[Fundamental and clinical studies on imipenem/cilastatin sodium in the field of obstetrics and gynecology].

Fundamental and clinical studies on imipenem/cilastatin sodium (MK-0787/MK-0791) in the field of obstetrics and gynecology were carried out. The concentration of MK-0787 in uterine tissue was 5.9-12.2 micrograms/g at 30 minutes after an administration of 500 mg/500 mg of MK-0787/MK-0791 by a 30-minute intravenous drip infusion and 0.9-1.1 micrograms/g at 185 minutes. The clinical application of the drug to 9 patients with gynecological infections produced "good" results with all the patients clearly showing clinical or bacteriological improvement. Neither side effects nor abnormal laboratory findings were observed. Based on these findings, MK-0787/MK-0791 appeared to be a useful antibiotic for the treatment obstetrical and gynecological infections.

13C nuclear magnetic resonance studies on bacteriochlorophyll a biosynthesis in Rhodopseudomonas spheroides S.

The 13C NMR spectra were analyzed in bacteriochlorophyll a and magnesium protoporphyrin methyl ester formed in Rhodopseudomonas spheroides S. in the presence of L-[1-13C]glutamate and [2-13C]glycine. After reassignment of three alpha-pyrrolic carbons (C-9, -14 and -16) of bacteriochlorophyll a, the spectra showed that C-2 of glycine was preferentially incorporated into the eight-carbon atoms in these tetrapyrrole macrocycles derived from C-5 of 5-aminolevulinic acid (ALA). C-2 of glycine was also incorporated specifically into methyl ester carbon of magnesium protoporphyrin IX methyl ester and methoxyl carbon of methoxycarbonyl group attached to isocyclic ring of bacteriochlorophyll a. No enrichment of these nine-carbon atoms was observed in the spectrum of bacteriochlorophyll formed in the presence of L-[1-13C]glutamate, showing exclusive operation of ALA synthase on bacteriochlorophyll biosynthesis.

Diurnal rhythms in calcium and phosphate metabolism in rodents and their relations to lighting and feeding schedules.

Diurnal fluctuations in serum Ca, inorganic phosphorus, Mg, and alkaline phosphatase activity and their relations to lighting and feeding schedules were investigated in nocturnal rats and diurnal asiatic chipmunks. The following results were obtained: all parameters in the animals fed ad libitum and adapted to a regulated light-dark cycle (with light from 7.00 to 19.00 h) showed diurnal rhythms; under the same experimental condition, the patterns of the rhythms in the nocturnal rat and the diurnal asiatic chipmunk were nearly 180 degrees out of phase with each other; under the continuous lighting condition for more than 10 days, the diurnal rhythms of the parameters in rats decreased in their amplitudes, and phase shifts or marked modulations in the diurnal curves occurred when the time of availability of food was restricted from 9.00 to 17.00 h without altering the lighting schedule. The above findings support an idea that the rhythmicity is one of the essential features involved in the Ca metabolism. It is demonstrated that the environmental photofraction is one of the essential factors in generating or in maintaining the circadian rhythm of Ca metabolism as in other physiological processes and that the feeding time also could have a marked effect on the diurnal rhythm in the Ca metabolism.
