Lack of carcinogenicity of agaritine by subcutaneous administration in mice.

Agaritine (A), an ingredient of the cultivated mushroom of commerce Agaricus bisporus, was administered by subcutaneous injection to two groups of randomly bred Swiss mice. In the first group the animals of both sexes were treated at a 100 micrograms/g body weight basis five times at weekly intervals, while in the second group the mice received a single A treatment of 100 micrograms/g body weight for females and 50 micrograms/g body weight for males. The administration of the compound resulted in no detectable carcinogenic effect in the animals. Since some of the breakdown products of A were shown to be carcinogenic in mice and the mushroom itself was found to be mutagenic, the field is discussed in the light of the obtained results.

Platelet adsorption-source of error in reference controls.

Preparation of platelet reference controls requires that the platelet membranes be strengthened by chemical cross-linking, usually with glutaraldehyde. The reaction produces a platelet which is strongly absorbed to plastic and glass surfaces. Tests of commercially available reference controls indicate adsorption to the counting containers. Surfactants inhibit adsorption but cause an apparent decrease in size. Polyethylene glyocol can eliminate adsorption without altering conductivity.

Sex dependent toxicity of four chemicals.

Toxicity investigations were conducted with 4 chemicals: hydrazine, 1,2-dimethylhydrazine di HCl, phenylhydrazine HCl and Beta-N-(gamma-L(+)-glutamyl)-4-hydroxymethylphenylhydrazine in Swiss mice. Hydrazine and phenylhydrazine HCl were administered daily in drinking water, the former at 0.01% and the latter at 0.001% concentrations. The 1,2-dimethylhydrazine di HCl and Beta-N-(gamma-L(+)-glutamyl)-4-hydroxymethylphenylhydrazine were given as a single subcutaneous injection at 45 mug and 100 mug/gr. body weight basis, respectively. The findings clearly showed that all four chemicals exerted a stronger toxic effect in male than in female mice. It is, therefore, recommended in similar situations to use different doses of chemicals for each sex in the long-term tumorigenesis studies.
