The initiation of fetal hemoglobin biosynthesis.

Biosynthesis of the alpha and beta chains of rabbit and human adult hemoglobin is initiated with a methionyl residue, which is removed during elongation of the peptide chain. To study the initiation of biosynthesis of the delta chain of human fetal hemoglobin, fresh placental blood was used for labeling experiments with radioactive amino acids. Labeled nascent peptide chains were purified from the polysomal fraction of placental blood reticulocytes. The number of amino acid residues in nascent gamma chain at the time of removal of its N-terminal methionine was estimated to be 40--60 from the relative yields of labeled tryptic peptides.

The occurrence of calicivirus in infants with acute gastroenteritis.

Calicivirus was detected in 8 (1.2%) of 647 hospitalized patients during a survey of acute gastroenteritis in infants and young children, conducted between December 1974 and September 1977. Morphologically calicivirus was approximately 30 nm in diameter with an easily recognizable staining "star of David" configuration. Its buoyant density in cesium chloride was 1.38-1.40 gm/ml. The serologic response to calicivirus by immune electron microscopy (IEM) was demonstrated only in paired sera from patients who shed the virus in their stools. The results suggest that calicivirus might be a cause of acute gastroenteritis in infants and young children.

Spontaneous endocrine tumors in Sprague-Dawley rats.

Spontaneous endocrine tumors were found in 81 of 100 Sprague-Dawley rats (42 males and 39 females) which survived for more than 2 years. Most of these tumors were medullary carcinomas of the thyroid, followed by tumors of the anterior pituitary gland, pheochromocytomas and cortical adenomas of the adrenal gland, and islet cell tumors of the pancreas. Multiple occurrence of these tumors was frequently observed. This study describes the morphology of these spontaneous endocrine tumors.

Role of Norwalk virus in outbreaks of nonbacterial gastroenteritis.

Twenty-five separate outbreaks of nonbacterial gastrointestinal illnesses were studied serologically for evidence of infection with the Norwalk virus and the rotaviruses that affect humans. Eight of 25 outbreaks appeared to be related to the Norwalk virus. In one of the 25 outbreaks, there was evidence of rotavirus infection. These observations suggest that the Norwalk virus or serologically related agents play an important role in epidemic nonbacterial gastroenteritis in adults and older children.

Effect of adrenalectomy on liver lipid content of fasted rats.

The interrelationship between thyroid and adrenal hormones in the regulation of lipid accumulation of the liver during fasting was examined with the use of intact, sham operated and adrenalectomized (ADX) rats previously fed a high carbohydrate diet. Adrenalectomy depressed the lipid accumulation of the liver induced by fasting. This was restored by cortisone administration, but not by epinephrine injection. Lipid content of the liver during fasting tended to be decreased in rats made hyperthyroid by intraperitoneal injection of thyroxine compared to euthyroid-fasted rats. In rats made hypothyroid by oral administration of propylthiouracil, liver lipid content during fasting was significantly higher than that of euthyroid-fasted rats. These responses of liver lipid to thyroid activity were minimized in ADX rats. Cortisone tended to restore completely the liver response to thyroid activity. The maximal response of liver lipid to cortisone was obtained in hypothyroid-ADX rats. The injection of epinephrine alone did not cause any significant change in liver lipid content of ADX rats regardless of the thyroid status. These findings suggest that glucocorticoid must be present for thyroid function to exert its effect on the lipid accumulation of the liver induced by fasting.

Formation of a TBA reaction product from crown ether in the presence of KO2.

Potassium superoxide (KO2), which can be dissolved in dimethyl sulfoxide containing crown ether, has been used as a source of O2-. for superoxide reaction systems. We have found that crown ether reacts with thiobarbituric acid (TBA) in the presence of KO2 to form a red pigment, which is a well-known reaction product of lipoperoxide.
