Mapping of genes controlling aluminum tolerance in rice: comparison of different genetic backgrounds.

Aluminum toxicity is the main factor limiting the productivity of crop plants in acid soils, particularly in the tropics and subtropics. In this study, a doubled-haploid population derived from the rice ( Oryza sativa L.) breeding lines CT9993 and IR62266 was used to map genes controlling Al tolerance. A genetic linkage map consisting of 280 DNA markers (RFLP, AFLP and SSR) was constructed to determine the position and nature of quantitative trait loci (QTLs) affecting Al tolerance. Three characters - control root length (CRL), Al-stressed root length (SRL) and root length ratio (RR) - were evaluated for the DH lines and the parents at the seedling stage in nutrient solution. A total of 20 QTLs controlling root growth under Al stress and control conditions were detected and distributed over 10 of the 12 rice chromosomes, reflecting multigenic control of these traits. The two QTLs of largest effect, qALRR-1-1 and qALRR-8 for root length ratio (a measurement of Al tolerance) were localized on chromosomes 1 and 8, respectively. Three other QTLs in addition to qALRR-8 were apparently unique in the CT9993 x IR62266 mapping population, which may explain the high level of Al tolerance in CT9993. Comparative mapping identified a conserved genomic region on chromosome 1 associated with Al tolerance across three rice genetic backgrounds. This region provides an important starting point for isolating genes responsible for different mechanisms of aluminum tolerance and understanding the genetic nature of this trait in rice and other cereals.

Dramatic effect of an additional CH2 group on the temperature variation of the Sm-C(*)(alpha) short helical pitch.

High-resolution differential optical reflectivity was used to study the temperature evolution of the short helical pitch in the Sm-C(*)(alpha) phase of successive members from two liquid-crystal homologous series. With the addition of one CH2 group, the magnitude and temperature evolution of the pitch change dramatically, and the molecular arrangements between consecutive surface layers found in free-standing films change from being anticlinic to synclinic.

Identification of the EcoKI and EcoR124I Type I restriction--modification enzyme subunits by non-equilibrium pH gradient two-dimensional gel electrophoresis.

Effectively optimized and reproducible procedure for monitoring the composition of type I restriction-modification endonucleases EcoKI and EcoR124I by non-equilibrium pH gradient two-dimensional (2-D) gel electrophoresis is described. Three subunits of the enzyme complex, which widely differ from one another in their isoelectric points and molar mass, were identified in crude cell extracts of E. coli. For the first time all three subunits of both EcoKI and EcoR124I were detected as distinct spots on a single 2-D gel. A sensitive immunoblotting procedure was suggested suitable for routine use in determining the identity of individual subunits. Potential application of this method for detailed studies of regulation of the function and stoichiometry of the enzyme complexes is discussed.

The effect of retinoids and butyrate on the expression of CRX and IRBP in retinoblastoma cells.

We sought to determine whether differentiation agents such as retinoids and butyrate regulate transcript levels of interphotoreceptor retinoid binding protein (IRBP) and cone rod homeobox (CRX), a homeodomain transcription factor that regulates IRBP promoter activity. WERI-Rb1 retinoblastoma cells were treated with all-trans retinol, all-trans retinoic acid, or butyrate. IRBP and CRX mRNA levels were determined by quantitative RT-PCR. Butyrate at low concentrations increased both mRNA levels but suppressed them at higher concentrations. Retinoic acid had minimal effects. Retinol increased CRX mRNA over four fold. IRBP and CRX transcript levels are sensitive to butyrate and CRX expression is sensitive to retinol.

Dielectric properties of twist grain boundary phases: influence of the anchoring and the distance between grain boundaries.

The dielectric properties of the twist grain boundaries TGB(A) and TGB(C) of liquid crystal phases differ from the smectic-A and smectic-C phase ones: a theoretical model confirmed by experimental results shows that the Goldstone mode of the TGB(C) phase and the soft mode of the TGB(A) phase are strongly reduced. This behavior is due to elastic strain, which is connected to two parameters: the anchoring at the grain boundaries and the distance between the grain boundaries. It is shown quantitatively that a relatively flexible anchoring in the TGB(A) phase becomes rigid in the TGB(C) one. The relaxation frequencies of these modes allow analysis of the rotational viscosity variations.

Detailed optical studies of several banana-shaped compounds in liquid crystal B2 phase.

Null-transmission ellipsometry has been performed on free-standing films of one compound in the liquid crystal B2 phase. We have studied films of thickness from 1 to 121 layers. One of the compounds used has an unusually wide 59 K window for the B2 phase. The tilt angle was investigated as a function of temperature and found to be constant over this temperature range to within our resolution of 1 degrees. The one-layer films studied exhibit the same structure as the thicker films, and have helped us to refine the optical model for the B2 phase. For thin films we find that modeling a single smectic layer as two uniaxial layers is a better description of the data than a single biaxial layer, but that for thick films the model used does not effect the simulated result appreciably. Preliminary results also find that the surface layers are less tilted than the interior layers, in contrast to rodlike liquid crystals, which show an enhanced surface tilt.

Evidence of a tissue-restricting DNA regulatory element in the mouse IRBP promoter.

The expression of interphotoreceptor retinoid binding protein (IRBP) is limited to photoreceptor cells of the retina and pinealocytes of the pineal gland. We sought to define cis-elements of the mouse IRBP 5' flanking region that are required for this restricted activity. In vitro transient transfections of retinoblastoma and neuroblastoma cells and in vivo experiments with transgenic Xenopus laevis indicate that -1783/+101 and -156/+101 IRBP gene fragments directed expression predominantly to the retina and pineal, with minor neuronal expression elsewhere. In contrast, a -70/+101 fragment was less restrictive in controlling expression, exhibiting activity not only in retina, but also in forebrain, hindbrain, spinal cord, and motor neurons innervating gills.

Structural properties of the ferrielectric phases of a chiral liquid crystal revealed by dynamic light scattering.

Dynamic light scattering is used to study the antiferro- and ferrielectric phases in a bulk sample of the chiral liquid crystal R-10OTBBB1M7. Comparison of the detected fluctuation modes in the two ferriphases to calculated selection rules for their coupling to light are consistent with the distorted clock structures recently reported by an optical ellipsometry study, but indicate that the distortion has a polar component in both ferriphases. We also find a surprisingly large amplitude and dispersion associated with scattering from fluctuations in the tilt angle magnitude. In terms of the discrete interlayer interaction models currently being used to account for the ferriphases, this finding suggests that the interlayer interaction coefficients are of comparable magnitude to the intralayer coefficients that stabilize the tilt angle.

Influence of the molecular tilt on the structure of smectic blue phases.

Smectic blue phases (BP(sm)) are original physical systems of thermotropic liquid crystals, displaying a double geometrical frustration: the extension of chirality in the three spatial dimensions, such as classical blue phases, and the competition between smectic order and helical twist, such as twist grain boundary (TGB) phases. We report experimental evidence of the influence of the underlying TGB((A) or (C)) phase on the BP(sm) structure.

Optical studies of the smectic-C(*)(alpha) phase layer structure in free-standing films.

The layer structure of the smectic-C(*)(alpha) phase of one liquid-crystal compound has been acquired from both differential optical reflectivity and ellipsometry measurements in the free-standing film geometry. The data from both techniques display characteristic oscillations as a function of temperature, which can be described by a model for the film consisting of surface anticlinic layers and an interior short-pitched azimuthal helix. These results are consistent with those found previously for another compound. Depolarized reflected light microscopy is used to study the films when the unique features of the aforementioned oscillations occur.

Optical rotatory power of different phases of an antiferroelectric liquid crystal and implications for models of structure.

The antiferroelectric liquid crystal (AFLC) under investigation possesses different helical smectic phases. The various phases have been identified through a texture under cross-polarizers with a homeotropic alignment of the AFLC. Measurements of the optical rotatory power (ORP) of these phases have elucidated the ability of this method for finding phase transitions between several phases and for investigating the helical structure of the antiferroelectric phases. The optical rotatory power as a function of temperature at a fixed wavelength of light was measured for different phases of the investigated AFLC material. The values of the pitch for some of the phases have been calculated from the ORP data. The results of the ORP rule out the simple "clock" model or a clock model with a long pitch superimposed on to it. The results can be explained only in terms of biaxial models, either Ising-type models or a highly distorted "clock" model. It is also found that in the SmC*A phase the sense of the helix in the investigated material is left handed, and is opposite to that observed in the SmC* phase. The reversal of the helix from left to right handed occurs during the phase transition from the SmC*FI1(SmC*gamma) to SmC*FI2(AF) phase. This fact also allows for SmC*A and AF phases to be distinguished from each other.

Structural investigations on smectic blue phases.

Smectic blue phases ( BP(Sm)) are thermotropic liquid crystalline phases which exhibit both three-dimensional orientational order, such as classical blue phases, and smectic positional order. BP(Sm) appear as the three-dimensional counterpart of twist grain boundary phases. X-ray scattering and optical polarizing microscopy provide information on the hexagonal and cubic symmetries of these new phases.

Optical studies of the liquid crystal B2 phase formed by banana-shaped molecules.

Null-transmission ellipsometry and depolarized light microscopy have been performed on free-standing films of three achiral banana-shaped compounds in the B2 phase. Our results support a two-layer unit cell previously proposed to explain the observed antiferroelectricity in thin films and bulk samples. We have studied thicker films than previously reported and have found no deviations in the film structure from the earlier findings. Moreover, we can determine the layer spacing, the molecular tilt from the layer normal, and the three principal indices of refraction in the molecular reference frame.

Interactions between colloidal inclusions in two-dimensional smectic-C* films.

We report an experimental study of colloidal inclusions in free-standing films of smectic-C* liquid crystal. The inclusions are cholesteric droplets that form above the bulk smectic-C*-7cholesteric transition temperature. Each droplet confined in a two-dimensional (2D) system, is accompanied by a topological defect. The distortions of the in-plane orientational order of the smectic-C* film induce elastic interactions between the droplets. As in 3D water nematic emulsions, a short-range repulsion and a long-range dipolar attraction govern the stability of the inclusions and lead to their organization in chainlike structures. Our results are in agreement with recent theoretical predictions.

Cloning of new members of heat shock protein HSP101 gene family in wheat (Triticum aestivum (L.) Moench) inducible by heat, dehydration, and ABA(1).

We have cloned two cDNAs, TaHSP101B and TaHSP101C, encoding two heat stress-inducible members of HSP101/ClpB family in bread wheat (Triticum aestivum (L.) Moench.). Proteins encoded by these cDNAs are highly similar at the primary sequence level and diverged from the previously reported TaHSP101 (designated TaHSP101A) both in the consensus ATP/GTP-binding region II and in the carboxy terminal region. The HSP101 gene was determined to be a single copy gene or a member of a small gene family in hexaploid wheat. Messages encoding HSP101 proteins were inducible by heat stress treatments in both wheat leaves and roots. Accumulation of the TaHSP101C mRNA was less abundant than that of TaHSP101B mRNA. We are showing for the first time that in addition to heat stress, expression of HSP101 mRNAs in wheat leaves was induced by a 2-h dehydration and a treatment with 5x10(-5)M ABA, but not affected by chilling or wounding, indicating that HSP101 proteins may be involved in both heat and drought responses in wheat.

Fetal intervention for myelomeningocele: effect on postnatal bladder function.

PURPOSE: Myelomeningocele is the most common congenital malformation of the central nervous system noted on prenatal ultrasound. Due to its significant postnatal sequelae, treatment in utero could potentially have a profound impact on the newborn. Others have reported fetal surgical techniques for in utero repair of myelomeningocele and its potential benefits on motor and neurological function. We report our urodynamic findings in the newborn after in utero repair of spina bifida in an effort to characterize postnatal bladder function. MATERIALS AND METHODS: A retrospective review of the fetal surgery database at University of California San Francisco was performed identifying patients with a diagnosis of myelomeningocele. Prenatal surgical repair of myelomeningocele was considered if a normal karyotype was present, no other significant congenital anomalies were evident and gestational age was less than 24 weeks. The spinal defects were in the lumbar or lumbosacral region. All surgery was performed before 24 weeks of gestations. RESULTS: Fetal surgery to correct myelomeningocele was performed in 6 patients. All patients were born premature at 32 weeks of gestation or less. Videourodynamics performed at age 1 month in 4 patients indicated decreased bladder capacity for weight, increased detrusor storage pressures and significant post-void residual. Hydronephrosis was demonstrated in 4 patients on renal/bladder ultrasound, and moderate vesicoureteral reflux was seen in 3. CONCLUSIONS: Patients with spinal bifida treated in utero appear to have the same changes in urodynamic parameters and anatomical abnormalities in the urinary tract as other children with spinal defects who have undergone standard postnatal care. In utero treatment of spinal bifida may expose the newborn to the effects of prematurity. The long-term effects on bladder function in the fetus after in utero repair of myelomeningocele remain unknown. A randomized controlled trial is necessary to evaluate long-term bladder function as well as other outcome variables in this experimental approach to patients with myelomeningocele.

Extra-hepatic expression of serum albumin mRNA in mouse retina.

PURPOSE: In some mammals, serum albumin protein exists in the interphotoreceptor space (IPS), the space between photoreceptor cells and the retinal pigment epithelium. Serum albumin is synthesized largely in the liver, though low levels of extra-hepatic expression have been documented in several other tissues, including fetal rat kidney, pancreas, lung, and heart. The purpose of this study was to investigate whether serum albumin protein and mRNA are found in mouse retina. METHODS: Using albumin rabbit antibodies and HRP goat anti-(rabbit IgG), we performed immunoassays on mouse IPS washes to detect the presence of serum albumin protein. Protein extracts from IPS washes were subjected to Affigel Blue chromatography. This resin has an affinity for serum albumin. Reverse transcription-polymerase chain reaction (RT-PCR) of retina total RNA was performed to search for albumin mRNA. Also, real-time reverse transcription polymerase chain reaction (RT-RT-PCR) was employed to look at the levels of expression in different age groups. RESULTS: A constituent of the IPS washes specifically bound and eluted from Affigel Blue column, suggesting that the washes contained serum albumin. SDS PAGE revealed that the size of the constituent was 67 kDa, the size of serum albumin. This 67 kDa band reacted with mouse serum antibody. An RT-PCR amplified fragment of serum albumin mRNA from retina displayed the expected size. The sequence of this fragment is identical to authentic serum albumin cDNA sequence. RPE and choroid were negative for serum albumin mRNA. However, rd1(-)/rd1(-) retina was positive, suggesting that at least some serum albumin is synthesized in the inner layers of the retina. RT-RT-PCR showed that serum albumin mRNA levels in whole retina reached a maximum at about postnatal day 15 and gradually decreased to about one-sixth of maximum at 12 months age. CONCLUSIONS: Serum albumin protein and mRNA were found in mouse IPS and retina, suggesting that the protein is synthesized in the retina. The previously demonstrated ability of serum albumin to bind fatty acids and retinoids and its presence in the mouse IPS suggest a role for serum albumin in transporting retinoids in the retina or IPS, especially at young ages when concentrations appear greatest.
