Wheat nuclear protein HBP-1 binds to the hexameric sequence in the promoter of various plant genes.

HBP-1 is a sequence-specific DNA-binding protein that interacts with the hexameric sequence ACGTCA, the putative cis-acting element of the wheat histone H3 gene. Gel mobility shift and DNase I footprint analyses showed that this protein interacts with homologous sequences in the regulatory regions for the transcription of the cauliflower mosaic virus (CaMV) 35S RNA and nopaline synthase (NOS) genes, evidence that HBP-1 may bind to hexameric sequences in the regulatory regions of various genes. An HBP-1-like protein, indistinguishable from wheat HBP-1 in its the DNA-binding specificity, is present in sunflower nuclear extract, an indication that HBP-1-like DNA-binding proteins also exist in dicots.

Multiplicity of the DNA-binding protein HBP-1 specific to the conserved hexameric sequence ACGTCA in various plant gene promoters.

A novel DNA-binding protein that specifically interacts with the hexameric sequence ACGTCA in the regulatory region of the wheat histone H3 gene has been identified in wheat nuclear extract and designated HBP-1a. The nuclear protein HBP-1 previously identified as a DNA-binding protein that interacts with hexameric sequences in the H3, cauliflower mosaic virus (CaMV) 35 S RNA, and nopaline synthase (NOS) promoter regions therefore has been renamed HBP-1b. The flanking sequences that surround the hexameric sequence may account for the difference in the binding properties of HBP-1a and HBP-1b.

Binding, Competitive↗

A protein that binds to a cis-acting element of wheat histone genes has a leucine zipper motif.

The structure and function of transcription factors of higher plants was studied by isolating cDNA clones encoding a wheat sequence-specific DNA binding protein. A hexameric nucleotide motif, ACGTCA, is located upstream from the TATA box of several plant histone genes. It has been suggested that this motif is essential for efficient transcription of the wheat histone H3 gene. A wheat nuclear protein, HBP-1 (histone DNA binding protein-1), which specifically binds to the hexameric motif, has previously been identified as a putative transcription factor. A cDNA clone encoding HBP-1 has been isolated on the basis of specific binding of HBP-1 to the hexameric motif. The deduced amino acid sequence indicates that HBP-1 contains the leucine zipper motif, which represents a characteristic property of several eukaryotic transcription factors.

Features of the anti-ulcer effects of Oren-gedoku-to (a traditional Chinese medicine) and its component herb drugs.

This report describes the features of the anti-ulcer effect of Oren-gedoku-to (OGT, a traditional Chinese medicine) and its component herb drugs. Coptidis rhizoma and Phellodendri cortex given orally dose-dependently inhibited the appearance of ethanol-induced gastric hemorrhagic lesions in a dose range of 25-100 mg/kg, but the formation of the lesions was not prevented by Scutellariae radix or Gardeniae fructus at the same doses. Coptidis rhizoma, Phellodendri cortex and Gardeniae fructus inhibited the gastric potential difference (PD) reduction induced by ethanol, whereas Scutellariae radix did not prevent the decrease in the PD reduction caused by ethanol. Phellodendri cortex, Scutellariae radix and Gardeniae fructus had no significant influence on the basal PD, while Coptidis rhizoma increased the basal PD. The four herb drugs prevented gastric acid secretion induced by 2-deoxy-D-glucose, but the three drugs except for Phellodendri cortex showed little effect on pentagastrin-stimulation. These results suggest that the gastric mucosal protection by OGT is ascribed to Coptidis rhizoma and Phellodendri cortex, and its antisecretory effect is due to the four drugs.

A possible mechanism for the gastric mucosal protection by oren-gedoku-to (OGT), a traditional herbal medicine.

We investigated the involvement of sulfhydryl-compounds in the cytoprotective effect of Oren-gedoku-to (OGT) against ethanol-induced gastric lesions and potential difference (PD) reduction in comparison with those of sucralfate and glutathione in rats. Pretreatment with indomethacin (IND) had little influence on the cytoprotective effects of OGT and glutathione, but attenuated the effect of sucralfate. Pretreatment with N-ethylmaleimide (NEM) blocked the cytoprotective effects of these three drugs. Pretreatment with iodoacetamide diminished the cytoprotection of OGT and glutathione, but had little effect on that of sucralfate. The cytoprotective activities of OGT, glutathione and sucralfate were little affected by pretreatment with 6,6'-dithiodinicotinic acid. The inhibitory effects of OGT and glutathione against the PD reduction were completely blocked by pretreatment with NEM, while they were not influenced by pretreatment with IND. On the other hand, the inhibitory effect of sucralfate disappeared both by IND and NEM. These results suggest that the cytoprotective effect of OGT may be mediated by endogenous sulfhydryl compounds, but not by endogenous prostaglandins in the gastric mucosa.

[Study on renal osteodystrophy using 2-compartment model analysis of bone scintigraphy. 1. Clinical significance of K Index].

The dynamic analyses of bone scintigraphy were performed in 30 cases of hemodialysed patients. The regression analyses between biochemical data or duration and K indexes obtained from the 2-compartment model analysis in the maxillofacial region were carried out. The obtained results were as follows. 1) The duration and K indexes were significantly correlated with 1 or 2% of significance level in the parietal bone and the zygomatic bone. 2) Serum Ca and K indexes were highly correlated in every settled ROI. The correlation coefficients were distributed from 0.58 to 0.79. In the cranial bone the correlation were more higher than in maxilla and mandibular bone. 3) Serum ALP and K indexes have more higher significant level of correlation than the results of Ca. The correlation coefficients were distributed from 0.48 to 0.83. The level of significance were changed by location. The highest correlation coefficient was 0.83 in the parietal bone. 4) Serum c-PTH and K indexes have significant correlation in every settled ROI with the correlation coefficient from 0.59 to 0.69. The significance level were nearly equal in every location. 5) In the case of subtotal parathyroidoectomy the K indexes which were abnormally high in preoperation showed the tendancy to decrease toward normal range in postoperation. These results indicated that K indexes reflected the bone change caused by renal osteodystrophy. And the K indexes considered to be usefull to estimate the bone improvement.

[A case of complete rupture of the righ TMJ disc].

A case of complete rupture of the right TMJ disc was described. A 43 year old woman visited our hospital with the chief complaint of a mild pain in the right TMJ. Initially, a conservative treatment using the occlusal splint (condylar repositioning appliance) was carried out. This was because she had reciprocal clicking of the right TMJ which had occurred not long before and it seemed that the symptoms were due to the trouble with the disc. In spite of this treatment, for 6 months no improvement was obtained and the reciprocal clicking changed into crepitation. Moreover the double contrast arthrotomographic image indicated a perforation or a wide rupture of the disc, and deformities of the articular bone. Then a right TMJ osteoarthrosis with wide (complete) disc rupture was diagnosed, and a discectomy was performed. The symptoms were greatly reduced by this treatment. It was conceivable that the steroid which she had used for a long time to treat bronchial asthma caused a remarkable inhibition of the tissue repairing, and an internal derangement of the TMJ rapidly progressed to osteoarthrosis.

[Study on gypsum hardener].

The purpose of this study was to search for an appropriate condition for shortening the treatment time of gypsum hardener, Plaster Aid. In order to keep the increased strength of plaster with this new hardener, compressive strength and dimensional change in various conditions of heating temperature, heating time and drying time were examined. The following results were obtained. 1. High compressive strength was maintained in the condition of 70 degrees C of heating temperature, with more than 15 minutes of heating time and more than 15 minutes of drying time. In this condition up to 90 minutes could be saved in comparison with the conventional method. 2. Plaster models had a tendency to shrink in the condition of 70 degrees C of heating temperature.

DNA-binding protein(s) interacts with a conserved nonameric sequence in the upstream regions of wheat histone genes.

A nuclear protein(s), HBP-2, that binds to the upstream region of the wheat histone H4 gene was identified from a fractionated nuclear extract of wheat germ by DNase I footprinting. The DNase I-protected region contained the conserved nonameric motif, CATCCAACG. Cross-competition experiments that used the mobility shift assay showed that this nuclear protein(s) binds specifically to the upstream sequence that has been postulated to be a cis element of the wheat H3 gene. Our findings suggest that this DNA-binding protein(s) may be a trans-acting factor in the regulation of the transcription of wheat histone genes.

Opsonic effect of monoclonal antibodies against Leptospira interrogans serovar copenhageni.

The opsonic effect of monoclonal antibodies (McAbs) against Leptospira interrogans serovar copenhageni strain Shibaura was examined in vitro using radiolabeled organisms and mouse peritoneal exudate macrophages. Four IgG McAbs (all IgG3) and 2 IgM McAbs were used, all showed different reactivities and were bound to homologous lipopolysaccharide (LPS). IgG3 McAbs at the sub-agglutinating concentration were opsonic, but the degree of the opsonic effect varied among IgG3 McAbs. Opsonization with IgG3 McAbs at the concentration showing the same level of ELISA binding showed that the highest opsonic effect was exerted by Sw-6 antibody. The other IgG3 McAbs showed a similar but lower opsonic effect. IgM McAbs, which were not opsonic at the sub-agglutinating concentration even in the presence of complement, showed opsonic effect at the agglutinating concentration.

[Pharmacological studies of some traditional Chinese medicines on gastric functions. (2) The effects of oren-gedoku-to (OGT), san'o-syasin-to (SST), antyu-san (AS) and dai-saiko-to (DST) on gastric acid secretion in rats].

The effects of OGT, SST, AS and DST on gastric acid secretion stimulated by histamine, pentagastrin, carbachol and 2-deoxy-D-glucose (2-DG) were studied in the perfused stomach of anesthetized rats, and these effects were compared with those of cimetidine, 16,16-dimethyl-prostaglandin E2(DMPGE2) and atropine. OGT and SST showed little or no effect on the acid secretion induced by histamine or carbachol at doses of 100 mg/kg, whereas the former showed a moderate inhibition and the latter showed a marked one against pentagastrin-stimulation. AS and DST had no effect on the acid secretion induced by carbachol at doses of 100 mg/kg, whereas they showed a moderate inhibition against histamine-stimulation, and the latter showed a significant inhibition against pentagastrin-stimulation. Further, each of the above four drugs showed a significant effect on 2-DG-stimulation. Cimetidine (1-10 mg/kg) inhibited gastric acid secretion stimulated by histamine, pentagastrin, carbachol or 2-DG in a dose-dependent manner. Similarly, DMPGE2 (10 micrograms/kg) strongly inhibited acid secretion induced by the four secretagogues. Atropine (50 micrograms/kg) inhibited acid secretion induced by carbachol or 2-DG, but not that by histamine or pentagastrin. These results suggest that OGT, SST, AS and DST clearly affect the mechanism of gastric acid secretion, and the site of action of OGT may differ from those of SST, AS and DST.

The effects of OGT, SST, AS and DST on ethanol- and aspirin-induced gastric hemorrhagic lesions in rats were studied in comparison with those of sucralfate, 16,16-dimethyl-prostaglandin E2 (DMPGE2) and cimetidine. 1) OGT given orally at doses of 25-250 mg/kg protected gastric mucosa from injury induced by ethanol or aspirin. 2) SST prevented the appearance of aspirin-induced lesions at doses more than 25 mg/kg, and ethanol-induced lesions at 250 and 500 mg/kg. 3) AS and DST inhibited aspirin-induced lesions at more than 250 mg/kg and inhibited ethanol-induced lesions at 500 mg/kg. 4) Sucralfate (500 mg/kg) significantly prevented ethanol- and aspirin-induced lesions. DMPGE2 significantly inhibited ethanol-induced lesions at doses more than 0.1 micrograms/kg, and it inhibited aspirin-induced lesions dose-dependently at doses ranging from 0.1 to 5 micrograms/kg. Cimetidine significantly inhibited aspirin-induced lesions at doses of 10-250 mg/kg and inhibited ethanol-induced lesions at doses of 100 and 250 mg/kg. These results suggest that OGT, SST, AS and DST have a prophylactic effect on ulcerogenics, and the potency of OGT may be superior to those of SST, AS and DST.

Effects of Triton WR 1339 and orotic acid on lipid metabolism in rats.

In order to investigate the effect of hepatic cholesterol flux on biliary bile acids, Triton WR 1339 and orotic acid were administered to rats, and the biliary cholesterol, phospholipids and bile acids were analyzed together with serum lipoproteins and hepatic lipids. Triton, which raised serum very low density lipoprotein and lipid levels and decreased serum high density lipoprotein liver lipid levels, increase the biliary cholic acid group/chenodeoxycholic acid group ratio (CA/CDCA) in the bile without affecting the total amount of bile acids and the other biliary lipids. Orotic acid, which decreased serum lipid and lipoprotein concentrations and increased liver lipid levels, increased the biliary excretion of cholesterol and phospholipids, but produced no significant change in the total amount of bile acids and in the CA/CDCA ratio in bile.

Purification, characterization and serological properties of a glycolipid antigen reactive with a serovar-specific monoclonal antibody against Leptospira interrogans serovar canicola.

A glycolipid antigen possessing a serovar-specific antigenic determinant of Leptospira interrogans serovar canicola was purified from a chloroform/methanol extract of the organism. The purification procedures included silicic acid column chromatography and preparative thin-layer chromatography (TLC). Antigenic activity was detected by a TLC-enzyme immunostaining technique using monoclonal antibody CT3, which specifically agglutinates serovar canicola and only weakly serovar sumneri but no other serovars of Leptospira. The purified glycolipid reacted with CT3 antibody, indicating that the glycolipid possessed a serovar-specific antigenic determinant. Infrared spectrum and proton nuclear magnetic resonance analyses showed that the glycolipid contained sugar and lipid moieties, which possessed amide linkages and an acetyl group. Gas-liquid chromatography-mass spectrometry analysis showed that the glycolipid contained two unknown sugars, one of which (unknown sugar II) appeared to be associated with the antigenic determinant specific for canicola. The serovar-specific antigenic determinant was destroyed by mild alkali treatment of the glycolipid. These findings suggested that the antigenic determinant was an alkali-labile moiety which may be related to the unknown sugar II.

[Pharmacological studies of some blended traditional Chinese medicines on gastric functions. (1). The effects of oren-gedoku-to (OGT), san'o-syasin-to (SST), antyu-san (AS) and dai-saiko-to (DST) on ethanol- and aspirin-induced injury of gastric mucosal barrier].

We studied the effects of OGT, SST, AS and DST on ethanol- and aspirin-induced reduction of gastric transmucosal potential difference (PD) in rats as an index of gastric mucosal damage, and these effects were compared with those of sucralfate, 16,16-dimethyl-prostaglandin E2 (DMPGE2), cimetidine and proglumide. 1) OGT and SST as well as sucralfate and DMPGE2 significantly inhibited the PD reduction induced by ethanol and aspirin. 2) AS and DST as well as cimetidine significantly inhibited the PD reduction induced by aspirin, but not by ethanol. 3) Proglumide did not influence the PD reduction induced by ethanol and aspirin. 4) OGT alone, like sucralfate and proglumide, showed no influence on the basal PD, while SST, AS and DST, like DMPGE2 and cimetidine, increased the basal PD. These results suggest that OGT, SST, AS and DST protect the gastric mucosal barrier, and this effect of OGT differs from that of SST, AS and DST.

Effects of castration and testosterone administration on serum lipoproteins and their apoproteins in male spontaneously hypertensive rat.

Serum triglyceride and very low density lipoprotein (VLDL) concentrations were higher in male spontaneously hypertensive rat than in male control Wistar Kyoto rat, whereas serum cholesterol, phospholipids, and high density lipoprotein (HDL) concentrations were lower. Castration of hypertensive rats induced an increase in serum cholesterol, phospholipids, and HDL, and a decrease in serum triglyceride and VLDL. The cholesterol content of HDL increased, whereas the triglycerides decreased after gonadectomy of hypertensive rats. These changes in serum lipids and lipoproteins could be reversed by the administration of testosterone. Apolipoprotein E contents in VLDL and HDL of hypertensive rats were low when compared with control rats but rose after castration and could be reduced by testosterone administration. Hypertensive rats accumulated triglycerides and cholesterol in the liver, which resulted in an increase of liver weight. Castration reduced the hepatic lipids as well as liver weight. The effects of castration and testosterone treatment on lipids and lipoproteins were more prominent in spontaneously hypertensive rats than in control rats. These results suggest that testosterone reduces VLDL catabolism which is related to changes of apolipoprotein composition, and that hypertensive rats are more sensitive to testosterone than control rats.
