Purification and point-mutation analysis of human interleukin-2 (Ser-125).

A new type of IL-2(Ser-125) was purified by semi-preparative RP-HPLC, and its purity was analyzed with microbore HPLC, IEF, SDS-PAGE and capillary electrophoresis (CE). The N-terminal sequencing indicated the microheterogeneity of the N-terminus, i.e., N-Met(2/3) and N-Ala(1/3), which was identified with the results of CE and IEF. The amino acid sequence of the point mutated peptide confirmed the replacement of Cys-125 with Ser.

The upstream activating sequence for L-leucine gene regulation in Saccharomyces cerevisiae.

The upstream activating sequence (UAS) conferring leucine-specific regulation of transcription in Saccharomyces cerevisiae was identified by analysis of the LEU2 promoter and by comparison to other genes regulated by leucine. The UAS was localized with deletions and cloned synthetic DNA. Point mutations and sequence rearrangements were used to identify important basepairs and to construct an improved UAS with increased regulation and expression. The improved UAS contains a core ten basepair, GC-rich, palindromic sequence, which is sufficient to confer minimal levels of activation and regulation, within a 36 basepair palindromic sequence which confers maximal activation and regulation. Deletions downstream of the UAS indicated that the UAS must act in conjunction with at least one other site, perhaps a TATAA region, in order to confer high levels of activation. Tandem copies of the UAS in front of LEU2 increased expression and regulation. Tandem UAS elements in trans on a multi-copy 2 mu-based plasmid decreased expression and regulation. These results are consistent with a model that the UAS serves as the DNA-binding site for diffusible activation factor(s), possibly the LEU3 gene product.

Morbidity associated with temporomandibular joint arthrography in clinically normal joints.

To document the short- and long-term morbidity of arthrograms done on clinically normal temporomandibular joints, 62 arthrograms were performed on 31 asymptomatic volunteers. All subjects kept a record of any joint pain they experienced during the next five days and were examined by one of the authors at five days and again at two months. Maximum interincisal opening distance was recorded before the arthrogram and at both postarthrographic evaluations. No subjects complained of severe (grade 3) joint pain. Sixteen joints were recorded as moderately painful (grade 2), 32 as mildly painful (grade 1), and 14 as not painful (grade 0). Ten of 31 subjects described transient changes in occlusion one week after arthrography but none of the changes persisted until the two-month examination. Maximum interincisal distance was significantly decreased at one week but not at two months. These findings, although gained from asymptomatic subjects, suggest that complications of arthrography are probably minor and transient when performed by an experienced arthrographer.

Temporomandibular joint arthrography of normal subjects: prevalence of pain with ionic versus nonionic contrast agents.

Bilateral temporomandibular joint (TMI) arthrography was performed in 31 healthy subjects to determine the prevalence and degree of postprocedural pain and to establish if morbidity could be decreased by use of a nonionic contrast agent. Ionic contrast material (Hypaque 60%) was injected on one side and nonionic contrast material (Amipaque) on the other side. The pain experienced in the 5 days following arthrography was graded and recorded. Discomfort, when present, was described as mild or moderate and usually lasted for 2 days after the procedure. In no instance was the pain considered severe. There was significantly less pain on the side that was injected with nonionic contrast material. The invasiveness of the examination should not be a deterrent since discomfort from TMJ arthrography is minimal when performed by an experienced arthrographer; use of a nonionic contrast agent can further decrease the pain.

The role of cytosine arabinoside maintenance in acute nonlymphoblastic leukemia.

A series of 30 unselected patients with acute nonlymphoblastic leukemia (ANLL) was treated with combination chemotherapy, including three courses of cytosine arabinoside (Ara-C) by 5-day continuous i.v. infusion, vincristine i.v. weekly, and prednisone daily to complete remission. Ara-C was administered alone as a 5-day continuous i.v. infusion monthly for maintenance. Ten (33%) achieved a complete remission (CR). The remaining 30 (67%), including temporary partial remissions, hematologic improvements, inadequate trials, and early deaths, were all considered failures. The CR rate was 57% in those 17 cases receiving an adequate trial. After After 5 1/2 years' followup, the overall median survival, including cases failing to achieve CR, was 3.1 months. For those having adequate trials the median survival was 16.6 months, and for those achieving a CR, 36.6 months. Two patients are still alive, one at 55.2 months on maintenance therapy, and the other at 62.8 months, currently unmaintained.

Anti-HBe titre in patients infected with wild-type and e-minus variant of hepatitis B virus.

Serum samples from 77 HBsAg- and anti-HBe-positive chronic hepatitis B patients were studied for a G to A point mutation at the 1896 nucleotide of the precore region by polymerase chain reaction and oligonucleotide hybridization. Patients were grouped as: A, infected only by the wild-type virus; B, infected only by e-minus variant; C, coinfected by the wild-type and e-minus variant; and D, no precore/core gene of hepatitis B virus detected. All samples were assayed for the titre of anti-HBe by ELISA and compared as grouped. A significantly higher titre was found in the coinfected group C compared to groups A (p < 0.01) and D (p < 0.05). Results indicate that a high titre of anti-HBe is required as immune selective pressure for E-minus variant to overtake the wild-type virus.
