Protirelin (thyrotropin-releasing hormone) in thyroid gland: possible involvement in regulation of thyroid status.

AIM: To establish the presence of the hypothalamic hormone protirelin (thyrotropin-releasing hormone, TRH) in human thyroid and to investigate whether the concentration of this peptide in the thyroid gland is sensitive to thyroid status. METHODS: A procedure has been developed for the determination of TRH in the thyroid gland, distinct from TRH-like peptides which also react with TRH-antibody. RESULTS: Human thyroid was shown to contain both authentic TRH and TRH-like peptides, a similar pattern was seen in a range of animal thyroids. The concentrations of TRH in non-active goiter thyroids were substantial (41.6-248 pmol.g-1); in contrast the thyroids from hyperthyroid patients contained very little TRH (0.01-2.52 pmol.g-1). CONCLUSION: The physiologic role of TRH in the thyroid is not known but the large difference between the concentrations of this hormone in non-active and hyperactive thyroids suggests that thyroidal TRH may be involved in the regulation of thyroid status.

[Studies on the bioequivalence of canrenone using pharmacokinetic data and clinical effects].

Canrenon, an active metabolite of spironolactone, and the combination of Canrenon with hydrochlorothiazide (HCT) were examined on patients with essential hypertension, cardial oedemas and oedemas of different origin, and than compared to a product available on the market. A correlation of the clinical effect (e.g. systolic blood pressure) with the morning steady state-data (blood values of Canrenon and HCT) has been achieved. The new preparations showed a good resorption in the gastro-intestinal tract. For an equivalent clinical effect, less Canrenon substance has been needed. A therapeutically satisfactory result has been reached with the administered dosage of both Canrenon and the comparison group. In the combination the higher dosage level should be chosen.

[Influence of (+)-cyanidanol-3 on the course of acute viral hepatitis (author's transl)].

A double-blind study was conducted in four hospitals to test the effect of (+)-cyanidanol-3 (Catergen) on the course of acute viral hepatitis. Activities of a number of enzymes, bilirubin levels and gamma-globulin concentrations, as well as elimination of HBs antigen in blood were measured serially in 73 patients with viral hepatitis, 37 being treated with the drug, 36 patients receiving a placebo. In addition, liver biopsies before and after treatment were compared (19 patients treated, 23 patients on placebo). There was no significant difference between the treatment and placebo groups with regards to morphological and biochemical changes.

[Thrombophlebitis of internal cerebral veins in a case of systemic lupus erythematosus (author's transl)].

A 28-year-old man suffered from recurrent facial exanthema, arthritis and stomatitis for ten years and died six months after a catatonic episode with terminal cerebral convulsions. Three years before his death high KBR-Antititers to Herpes simplex- and cytomegalic virus were observed, while Lupus-Erythematosus-Tests (LE-Tests) only became positive in the last months. At autopsy, changes compatible with Systemic Lupus Erythematosus (SLE) were found in the mitral valves, the spleen, and the kidneys. The brain displayed hemorrhagic infarction of the striate bodies and thrombophlebitis of the internal cerebral veins, the wall of which exhibited circumscribed infiltrations with numerous hematoxilin bodies and LE cells. This seems to be the first observation of LE-specific changes in the brain. The importance of cerebral vein affection in SLE involving the nervous system is stressed and a hypothesis submitted proposing the viral etiology of SLE.

[Haemangiosarcomatosis after professional exposure to polyvinylchloride (author's transl)].

After taking a cursory look at neoplastic developments particularly at professional exposure to vinylchloride and polyvinylchloride, one case is reported in which a multilocular haemangiosarcomatosis occurred after a relatively short and only intermittent exposure to polyvinylchloride. The latent stage lasted 15 years. The vessel areas affected were the aorta abdominalis, the intrarenal kidney vessels, and the intracerebral vessel-systems. Symptomatically, the disease presented the picture of a progrediently advancing peripheral circulatory disturbance. The possibility of previous sclerotic damage caused by carbon sulphide is discussed.
