Effects of a bone lysine-rich 18 kDa protein on osteoblast-like MC3T3-E1 cells.

A lysine-rich 18 kDa protein was isolated from bovine bone and examined for its effects on osteoblast-like MC3T3-E1 cells. This protein is homologous to a heparin-binding protein in brain and uterus. This protein enhanced cell attachment independent of the Arg-Gly-Asp cell-binding sequence and stimulated proliferation during the growth phase. Addition of this protein to cell cultures on days 11, 12, and 13 after confluency resulted in a 1.6-2.0-fold increase in the alkaline phosphatase activity and little increase in the DNA content. These findings suggest that the 18 kDa protein may be functional in promoting the proliferation and differentiation of osteoblasts.

3T3 Cells↗

An osteonectin-like protein in the matrix of cultured osteogenic cell-line MC3T3-E1, which is associated with calcification.

Time-dependent changes of the [3H]-proline-labeled noncollagenous proteins synthesized by the osteogenic cell-line MC3T3-E1 were analyzed over a range starting from cell confluency to 13 days after confluency during which time cells formed a bone-like structure. It was found that the 40 kDa protein on SDS-polyacrylamide gel (SDS-PAGE) remarkably increased in the cell-matrix layer at about 9 days after cell confluence, just before calcification. This protein was highly purified and was found to contain high amounts of glutamic acid, glycine, and serine. An internal amino acid sequence of this protein was revealed to be K-X-M-A-P-E-E-X-P, which showed homology with the sequence of the EF-hand domain in osteonectin/SPARC (secreted protein, acidic, and rich in cysteine). This protein co-migrated with collagen in gel filtration, and ion-exchange chromatography. Furthermore, it showed high affinity to type I collagen.

t(13q;17p) and del(5q): possibly specific changes in Chinese patients with colorectal cancers.

Cytogenetic study of 18 colorectal carcinomas confirmed the extensive heterogeneity and the complexity of the karyotypic picture in this type of tumor. Karyotypic analysis showed that chromosomes 17p and 5q, in both numerical and structural aspects, were the most frequently involved chromosomes and prone to losses. The most common structurally rearranged forms were translocations of 17p with other chromosomes, especially t(13q;17p), which constituted over 50% of all 17p rearrangements, and an interstitial deletion of 5q that made up as much as 73% of all structural abnormalities of 5q. According to the results, we conclude that chromosomes 17 and 5 may play important roles in the evolution of colorectal cancer and t(13q;17p) and del(5q) may be possibly specific to Chinese patients with colorectal cancer.

Preliminary result of mixed bacterial vaccine as adjuvant treatment of hepatocellular carcinoma.

Mixed bacterial vaccine (MBV) was employed in the multi-modality treatment of hepatocellular carcinoma (HCC) during 1985-1988. Thirty eight patients undergoing palliative resection and cisplatin therapy (Series 1) and 48 patients with unresectable HCC who received hepatic artery ligation + intraarterial cisplatin infusion + radiotherapy (Series 2) were randomized to receive MBV or not. In series 1, the 1- and 2-year survival rates of MBV group and control were 75% vs 58% (P = 0.19) and 45% vs 39% (P = 0.23). In series 2, the 1-, 2- and 3-year survival rates were 59%, 41% and 41% for MBV group and 39%, 25% and 20% for the control, respectively (P1 = 0.07, P2 = 0.09, P3 = 0.07). In addition, MBV improved the "second look" resection rate to 40% as compared to 17% in the control (P greater than 0.05). MBV could also prevent such immunosuppression as decrease of macrophage activity caused by radiotherapy. We consider MBV a potential nonspecific immunostimulant in the multimodality treatment of HCC.

[Pharmacokinetics of [3H]ohmefentanyl in rats].

Ohmefentanyl (OMF) is a new potent analgesic derived from fentanyl. The pharmacokinetic characteristics of [3H]OMF in rats was studied in this paper. Time courses of concentration of [3H]OMF in plasma after bolus iv could be described as a 3-compartment open kinetic model. The major part of a single dose (1 microgram/kg) of [3H]OMF was rapidly eliminated from plasma by uptake of the active drug by tissues or organs and by biotransformation into hydrophilic metabolites. In respect to its lipophilic nature of OMF the concentrations of [3H]OMF in tissues or organs were much higher than those in plasma. At 1 min after iv, the concentrations of [3H]OMF in the lung, kidney, heart and brain stem were 40, 15, 7.5 and 6 times respectively higher than those in plasma. [3H]OMF and its hydrophilic metabolites were mainly excreted via the kidneys. The urine and feces in 72 h contained about 53% and 15% respectively of the total radioactivity administered. Approximately 65% of [3H]OMF was bound to plasma protein at pH 7.4, 4 degrees C. The bindings were the same for all concentrations of [3H]OMF between 0.02 and 1 ng/ml plasma in vitro.

Cloning of apoprotein AI gene and observation on DNA polymorphism of apoprotein AI/CIII gene-loci in genomic library from a liver of Chinese.

For the research on the structure and function of the HDL apoproteins, we have successfully constructed a complete genomic library, in self-prepared EMBL3 lambda vector and the packaging extracts of lysogenic strains BHB2688, BHB2690, from a Chinese fetal liver, which will be widely used in the research on the structure and function of apoproteins. In this study, a DNA sequence polymorphism, revealed by digestion of human DNA with the restriction endonuclease Sst1 and hybridization with an apoprotein AI cDNA probe, has been shown to be located at apoAI/CIII gene-loci. Three gene related fragments (5.7 Kb, 4.0 Kb and 3.0 Kb) were detected. The 5.7 Kb fragment is common and the polymorphism is demonstrated by the presence of either a 4.0 Kb fragment (S1 allele) or the 3.0 Kb fragment (S2 allele). Individuals were genotyped S1S1 or S1S2. Our result showed that the frequency of genotype S1S2 was higher in hypertriglyceridemic subjects than that in normolipidemic subjects.

A comparison of the care of hypertension in Hunan and Connecticut.

The demographic, clinical, and social characteristics of 300 hypertensive patients (150 seen in the Hypertension Clinic at Yale University and 150 seen in a newly opened clinic at the First Affiliated Hospital of the Hunan Medical College) were compared at their initial visit to the clinic. The mean blood pressure of both treated and untreated patients and the frequency of mild hypertension and rarity of secondary causes of hypertension were similar. Americans drank more alcohol and were more likely to be heavy smokers, and the Chinese were older, had more end organ damage, and had many more symptoms. The majority of patients in China (77%) were being treated most often with combination pills containing multiple agents. We feel that hypertensive patients are similar in the United States and China, though the Chinese patients appeared to have been seen at a later stage of their disease.

A comparison of the treatment of hypertension with Chinese herbal and Western medication.

Forty-five patients with diastolic blood pressure (DBP) greater than or equal to 105 mmHg were randomly assigned to receive Western (group 1, n = 21) or a classical Chinese herbal preparation (group 2, n = 24) to treat their hypertension (HBP). All remained hypertensive after 4 days in the hospital without treatment. Except for baseline Na+ excretion (higher in group 1) and somewhat more evidence of end organ damage in group 1, the patient groups were comparable. Those in group 1 were given a thiazide diuretic and propranolol if needed, and those in group 2, a mixture of 12 herbs. Patients on active therapy in group 1 had a drop in blood pressure (BP) from 172.6 +/- 27.8/107.4 +/- 13.6 to 141.2 +/- 26.2/89.6 +/- 12.0 mmHg, whereas those in group 2 had no change in BP, 168.8 +/- 22.0/107.7 +/- 9.8 mmHg to 165.7 +/- 23.7/106.0 +/- 11.8 mmHg. Although 66% of patients in group 1 had a DBP under 90 mmHg by discharge, only 8% of those in group 2 did. Except for a fall in serum K+ in group 1, there were no significant biochemical or clinical problems in either group. We conclude that standard Western medication is more effective than a classical Chinese herbal preparation used to treat HBP.

Oral immunization with a live recombinant attenuated Salmonella typhimurium protects mice against Toxoplasma gondii.

The natural site of infection for T. gondii is the mucosal surface of the intestine, so the protective immunity obtained after natural infection with T. gondii points to the importance of developing a vaccine that stimulates mucosal defences. In this study, an aroA- and aroD- attenuated strain of Salmonella typhimurium (BRD509) has been used to deliver the recombinant eukaryotic plasmid pSAG(1-2)/CTA2/B expressing a multi-antigenic gene encoding SAG1 and SAG2 of T. gondii linked to A2/B subunits of cholera toxin as a candidate oral T. gondii vaccine. Immunoblot analysis showed compound gene expression in HeLa cells in vitro and intragastric immunization of mice with the recombinant salmonella resulted in the induction of humoral and Th1 type cellular immune responses and afforded protection against RH strain T. gondii challenge. Anti-T. gondii IgG values increased markedly in the BRD509/pSAG(1-2)-CTA2/B immunized group; these values were significantly higher than those in the negative controls (P = 0.008). With CTA2/B genetic adjuvant, the T. gondii-specific response was predominantly Th1, indicating that the CTA(2)/B genetic adjuvant was able to overcome the strong Th2-bias of the antigen (IgG2a >> IgG1). Antigen-specific T cell proliferative responses and CTL activity were significantly enhanced when cholera toxin CTA2/B genetic adjuvant was used (P = 0.009; P = 0.006). Culture supernatants from antigen-stimulated splenocytes from mice in these groups were also examined by ELISA for Th1- and Th2-type cytokines; mean IFN-gamma levels produced after oral immunization with BRD509/pSAG(1-2)-CTA2/B were about nine-fold higher than after immunization with BRD509/pSAG(1-2) (P = 0.007). On the other hand, the levels of IL-4 were low for all groups and no increase was seen in the presence of CTA2/B genetic adjuvant. When the immunized mice were intraperitoneally challenged with 10(3) tachyzoites of the highly virulent RH strain, the survival time of the mice immunized with BRD509/pSAG(1-2)-CTA2/B was markedly longer than other groups (P = 0.003) and a 40% survival rate was achieved. This is the first report that demonstrates that an oral attenuated salmonella DNA vaccine can induce protective immunity against the acute phase of T. gondii infection.

A comparison of Chinese traditional and Western medical approaches for the treatment of mild hypertension.

We compared the efficacy of Chinese traditional treatment for mild hypertension with that of a standard Western medical regimen in a group of 50 well-matched patients (24 allocated to Western medicine and 26 to Chinese traditional medicine) with mild hypertension (diastolic blood pressure 90-104 mmHg). Those receiving Western therapy were treated in a stepped-care fashion with dihydrochlorothiazide and atenolol. Those in the Chinese traditional therapy group received one of two mixtures of nine herbs and other ingredients, depending on symptoms at initial evaluation. Blood pressure dropped significantly in both groups after only a few days on therapy. After 19 days on treatment, the group receiving Western therapy had a fall in blood pressure from 168.2/96.3 mmHg to 137.3/76.7 mmHg (p less than 0.01), while those on Chinese traditional therapy fell from 168.2/95.9 mmHg to 146.4/80.5 mmHg (p less than 0.01). The fall in blood pressure was significantly greater, however, in those given Western therapy. The relief of existing symptoms or development of possible drug side effects was similar in both groups, except for nocturia, occurring more often in the group treated with Western therapy. We conclude that Western therapy is more effective in reducing blood pressure as compared with Chinese traditional therapy, but effective control of blood pressure in mild hypertensives is possible with either form of treatment.
