Synthesis and evaluation of N-(phenylalkyl)acetohydroxamic acids as potential substrates for N-arylhydroxamic acid N,O-acyltransferase.

N-(4-Phenylcyclohexyl)acetohydroxamic acid and a series of N-(phenylalkyl)acetohydroxamic acids were synthesized and evaluated as substrates for partially purified rat and hamster hepatic arylhydroxamic acid N,O-acyltransferase systems (AHAT). The compounds were assayed for their abilities to function as acetyl donors in the AHAT-mediated transacetylation of 4-aminoazobenzene and for their abilities to participate in the AHAT-mediated conversion of N-arylhydroxylamines to electrophilic intermediates that form methylthio adducts upon reaction with N-acetylmethionine. None of the newly synthesized compounds displayed significant activity in either of the assays. The results of this study indicate that acetohydroxamic acids that have the nitrogen atom of the hydroxamic acid group attached directly to aliphatic or cycloalkyl groups are not likely to serve as substrates or inhibitors of AHAT.

Beta-adrenergic receptors in rat cerebellum after neonatal X-irradiation: effect of prolonged imipramine and lithium treatment.

The actions of prolonged treatment of rats within imipramine or lithium chloride on the beta-adrenergic cAMP generating system of the cerebellum were compared in normal animals and in rats degranulated by neonatal X-irradiation. Whereas in normal animals imipramine treatment is without significant effect on the cerebellar cAMP system but lowers the beta-adrenoceptor density significantly, in degranulated animals the cAMP system becomes subsensitive towards norepinephrine after prolonged imipramine treatment. These data support the hypothesis that in the cerebellum imipramine acts predominantly on beta-adrenoceptors located particularly on Purkinje cells. Prolonged lithium treatment exhibited no significant effects on beta-receptors nor the noradrenergic cAMP generating system of normal and degranulated cerebella.

Silicone intubation of the lacrimal system: loop retraction as a complication.

Inadvertent retraction of silicone tubing after nasolacrimal intubation occurs with some frequency. This problem may be managed in sequential fashion with probing, dacryocystostomy or dacryocystorhinostomy depending on the needs of the patient at the time of occurrence and the original rationale for utilization of the technique.

A pulmonary venous indentation on the esophagus-a normal variant.

An extrinsic indentation on the anterior left wall of the esophagus, about 4-5 cm below the carina, is a relatively common finding and should be considered a normal variant, although rarely it may be due to an aberrant insertion of a pulmonary vein into the left atrium. The indentation is best seen in a steep left posterior oblique horizontal position. In most cases it is caused by pressure of the left inferior pulmonary vein or a common confluence of the left pulmonary veins near the insertion into the left atrium. Generally, it does not cause any symptoms and should not be mistaken for a mediastinal mass.

Cellular immunity delimits adenoviral gene therapy strategies for the treatment of neoplastic diseases.

BACKGROUND: Adenoviral gene therapy is a promising new approach for the treatment of neoplastic diseases. To design rational clinical trials and distinguish the effects of therapeutic transgene expression from those caused by viral infection alone, the immune response to the vector must be understood. In these experiments, we further define cellular immunity to recombinant adenovirus. METHODS: The immune response to hepatic adenoviral gene transfer was studied in infected mice by depleting T cells with an anti-CD3 antibody, measuring splenocyte cytokine production, determining the impact of transgene expression on inflammation, and assessing liver MHC protein expression. RESULTS: The cellular immune response to recombinant adenovirus is (1) averted by T lymphocyte depletion, (2) marked by a TH1 response with increased IL-2 production, (3) directed against both the transgene product and viral proteins, and (4) associated with increased hepatocyte MHC Class I expression. CONCLUSIONS: It is necessary to take into consideration the constraints imposed by the immunogenicity of recombinant adenovirus and its transient transgene expression in the clinical application of adenoviral gene transfer for the treatment of cancer.

Cerebral gigantism (Sotos' syndrome) and cataracts.

A five-year-old girl with cerebral gigantism (Sotos' syndrome) and cataracts is described. Sotos' syndrome, characterized by generalized gigantism with normal endocrine studies has rarely been reported with ocular abnormalities and never with cataracts. It is important to study any child with cataracts for systemic disease.
