The aim of this study is to characterize the pattern of release of soluble vascular cell adhesion molecule-1 (sVCAM-1) in patients with acute cerebral infarction. The level of serum sVCAM-1 was measured serially with two-layer antibody sandwich enzyme-linked immunosorbent assay (ELISA) in 89 patients with acute cerebral infarction, 43 patients with cerebral haemorrhage and 30 normal controls. It is shown that the level of serum sVCAM-1 in patients with cerebral infarction 24 hours after the onset was higher than that in patients with cerebral haemorrhage and normal controls (P < 0.01). The level of serum sVCAM-1 was significantly higher in the patients with large cerebral infarction than that in patients with medium and small cerebral infarctions (P < 0.01). The level of serum sVCAM-1 in patients with cerebral infarction increased from 24 hours to 7 days and decreased from 7 days to 14 days, but it was still higher than that in patients with cerebral haemorrhage and normal controls on the 14th day. The level of serum sVCAM-1 was higher in patients with infection as compared with those without. These findings suggested that sVCAM-1 is closely related to the development of acute cerebral infarction. It is essential to have further study on the change of serumsVCAM-1 in cerebral infarction.
OBJECTIVES: To detect and compare the levels of vascular endothelial growth factor (VEGF) in the vitreous of patients with proliferative diabetic retinopathy (PDR) and to investigate the role of VEGF in the pathogenesis of PDR. METHODS: Undiluted vitreous samples of 7 normal subjects and 19 patients with PDR were obtained in different ways, and VEGF levels in the vitreous were detected by using enzyme linked immunosorbent assay (ELISA) for quantitative analysis. RESULTS: In the normal group (n = 7), the mean value of VEGF in the vitreous was 0.35 ng/ml (range 0.18-0.60 ng/ ml) in comparison with that of 5.66 ng/ml in the PDR group (n = 19; range 0.84-15.64 ng/ml), there was a significant difference between the two groups statistically (P < 0.05, Primer t test). In the comparison between male (n = 12; range 0.84-15.64 ng/ml) and female PDR group (n = 7; range 1.34-12.34 ng/ml), as their mean values of VEGF in the vitreous were 4.83 ng/ml and 7.08 ng/ml respectively, there was no significant difference between the two groups statistically (P > 0.05). CONCLUSIONS: The VEGF levels in the vitreous of patients with PDR were elevated, and as a neovascular growth factor, VEGF may play a role in the pathogenesis of PDR.

A technique of vital microcirculation observation and electron microscope were used to study the microcirculation in hamster cheek pouch, in which an acute inflammatory lesion was induced by oxygen free radicals (OFR). At the same time, the effect of superoxide dismutase (SOD) on the microcirculation was assessed. The results showed that in the model group the microcirculation disturbance in the cheek pouch was very serious. However, in the SOD group the microcirculation was almost unchangable.
Using our ELISA kit for GST-alpha, we tested the serum GST-alpha levels of patients with hepatitis A, acute hepatitis B (HB), chronic active HB, chronic persistent HB and liver cirrhosis. Serum GST-alpha levels in all these groups of patients were significantly higher than those in the controls (P < 0.01). Serum GST-alpha levels was closely (P < 0.01) correlated with serum alanine transaminase (ALT) levels in various groups of patients except chronic persistant HB. The combined application of the two markers, GST-alpha and ALT, raised the sensitivity of detection for liver diseases. In detection of chronic persistent HB patients, the GST-alpha marker was more sensitive than ALT (P < 0.01). The follow-up data of GST-alpha and ALT markers in serum showed that the GST-alpha level could reflect the clinical progression of liver disease more exactly as observed in 79 liver cancer patients in then GST-alpha and ALT positive rate were 81% and 61% respectively (P < 0.01). In 30 persons with positive HBsAg, the positive rates of GST-alpha and ALT were 70% and 37%, respectively. These results indicated that, (1) the detection of GST-alpha combined with ALT was capable of increasing the sensitivity for recognizing hepatocellular damage; (2) the elevation of serum GST-alpha level mainly resulted from the increased expression of GST-alpha in liver cells during hepatocarcinogenesis, thus, the GST-alpha is thought to be a tumor marker for liver cancer; (3) GST-alpha measurement offers advantage over ALT for the detection of minor degree of hepatocellular damages. GST-alpha may act as an early, sensitive and specific enzyme marker.
In this work, the contents of amino acids in three kinds of cotton-seed-cake powder from domestic and abroad were determined by using Waters Automatic Amino Acid Analyzer with post-column derivatization and fluorescence detection. The operating conditions were cation exchange resin column, 250 mm x 4.6 mm i.d., gradient elution, detection wavelength lambda(ex) = 338 nm and lambda(em) = 425 nm. The results showed that contents of amino acids from both domestic cotton seed cakes were less than those from abroad.
A reversed-phase high performance liquid chromatographic method for the determination of metronidazole (MTZ) in rabbit serum and cerebro-spinal fluid has been developed. The method is simple and convenient. The analysis was carried out with Zorbax C8 column, NaH2PO4(0.0075mol/L, pH 4.0)-CH3OH(4:11) as mobile phase and UV detector at 320nm, by using hydrochlorothiazide as internal standard. The standard curve is linear over the range from 0.5ng/L to 80.0ng/L in sample with correlation coefficient of 0.9998. The recovery of metronidazole from rabbits serum and cerebrospinal fluid are 97.1% +/- 1.5% and 97.4% +/- 1.6% respectively. The minimum detectable quantity is 80ng/mL in sample (S/N = 3). Under the chromotographic condition, the consistency of MTZ of the serum and cerebrospinal fluids of rabbits were detected separately after taking MTZ (0.1mg/g) orally. The drug-time curve was obtained and a satisfactory result was achieved. The new findings provide conditions for further exploration of the process of pharmacokinetics.
The design and preparation of a novel chiral stationary phase (CSP) were described. The chiral resolutions of N-3,5-dinitrobenzoyl-DL-amino acid methyl esters were performed on the CSP. The selectivities were between 1.04 and 1.18. The applications of the CSP and the optimization of the chromatographic conditions will be further studied.
A quantitative analytical method for 3,5-di-t-butyltoluene in reaction mixture by gas chromatography with methyl benzoate as internal standard is reported. A stainless steel column of 2m x 3mm i.d. packed with 15% FFAP/Chromosorb W/AW (60-80 mesh) and flame ionization detector were used with column temperature of 180 degrees C and injection port and detector temperatures of 250 degrees C. There was a good linear calibration curve in the range from 1.1 to 9.2 microg (r = 0.9990, n = 8). It was taken 10 minutes to complete an analysis.
In this work we found that there were five decomposition products of di-t-butyl peroxide at 320 degrees C by GC-MS. The peak area of acetone as a chief product in the thermal decomposition was related to di-t-butyl peroxide concentration in a linear manner. The range was from 22.91 to 114.93 microg (r=0.9997, n=4). By use of the calibration, we can possibly determine the content of the peroxide rapidly. The present method is more suitable for the determination of peroxide that is liable to decompose and difficult to be vaporized.
The levels of R- and S-mephenytoin in human urine were determined by chiral capillary gas chromatography with nitrogen-phosphorus detector. The conditions of the chromatography and detection included a chiral capillary column (Chirasil-Val, 25 m x 0.25 mm i.d., Alltech), column temperature (T) 190 degrees C, injector T 220 degrees C, detector T 240 degrees C, and the flow-rates of 0.85, 3.5 and 120 mL/min for nitrogen, hydrogen and air respectively. Based on the above conditions, the satisfactory separation of R- and S-mephenytoin was gained, and other interference from urine sample was not found. The linear curves for both tested compounds ranged between 12.5 and 2500 microg/L, with a minimum detectable concentration of about 6 microg/L. Because of its simplicity, rapidity, sensitivity and accuracy, this method has been extensively used for testing metabolic ability of S-mephenytoin polymorphic oxidation in the Chinese populations and for the determination of the activity of hepatic drug-metabolizing enzyme CYP2C19 using mephenytoin S/R ratio of the urine samples of subjects as an indicator.
A 51-year-old drunken male was carried to a hospital with acute abdominal pain and was suspected of acute pancreatitis. The patient was treated with fasting, electrolyte transfusion, and anodyne, but took a sudden turn for the worse and died in 16 hours. In the judicial autopsy, rupture of a small intestine was detected. As the police investigated, he had been kicked in the abdomen by an assailant before coming to the hospital. The cause of death was diagnosed to be acute peritonitis due to the rupture of a small intestine. Several problems were pointed out on medical examinations and treatments of this case.
Verbascoside is a natural antioxidant extracted from Pedicularis striata Pall (Jueyehesen). After being treated with 20 mumol/l verbascoside, the growth curve and mitotic index of human gastric adenocarcinoma MGc80-3 cells decreased remarkably, cell doubling time was delayed, the cellular growth inhibitory rate amounted to 53.2%, cell surface charge assayed by cell electrophoresis obviously changed, the electrophoresis rate dropped from 3.51 microns/s/v/cm to 2.74, i.e., the percent of retardation reached 28.4%. There was a 75% decrease of the tumorigenicity for the treated cells compared with the untreated cells inoculated subcutaneously in BALB/C nude mice. Scanning electron microscopy revealed that the microvilli on the surface of treated cells had been reduced obviously. It confirmed that verbascoside, similar to DMSO, could reverse MGc80-3 cells' malignant phenotypic characteristics and induced redifferentiation of MGc80-3 cells.
OBJECTIVE: To investigate the effects of pulse methylprednisolone (MP) and monthly intravenous cyclophosphamide (CTX) therapy (MP + CTX) on renal infiltrating cells in patients with rapid progressive glomerular nephritis (RPGN). METHODS: Twelve patients with RPGN (> 50% crescents) were given MP + CTX therapy and received repeated renal biopsies within 4 to 12 weeks after MP + CTX treatment. Seven were diagnosed as type II RPGN, including one case of IgA nephropathy, 2 cases of idiopathic RPGN and 4 cases of lupus nephritis and five were diagnosed as type III RPGN, including 2 cases of idiopathic RPGN and 3 cases of vasculitis. The changes of infiltrating CD4+, CD8+, CD68+ and proliferating cell nuclear antigen-PCNA+ cell levels were determined by four PAP method in glomeruli and interstitium. RESULTS: In the patients before MP + CTX therapy, there were higher levels of infiltrating CD4+ and CD8+ cells (306 +/- 118 and 223 +/- 98.4Num/mm2) in renal interstitium, CD68+ cells (17.2 +/- 9.95 Num/G) in glomeruli and (1120 +/- 229 Num/mm2) in interstitium, and PCNA+ cells (7.56 +/- 3.57 Num/G) in glomeruli and (17.6 +/- 6.85 Num/mm2) in interstitium as compared with those in the patients after MP + CTX therapy (CD4+/CD8+ cells were 171 +/- 87.5/121 +/- 38.4 Num/mm2, CD68+ cells were 9.04 +/- 4.33 Num/G and 600 +/- 107 Num/mm2, and PCNA+ cells were 2.04 +/- 1.43 Num/G and 9.40 +/- 4.45 Num/mm2). These changes were associated with improving renal dysfunctions (the levels of serum creatinine and proteinuria decreased gradually from 766 +/- 356 to 284 +/- 192 mumol/L and 2.60 +/- 1.46 to 1.29 +/- 0.85 g/day). CONCLUSIONS: Our data indicate that the renal infiltrating cells may play an important role in renal injury in patients with RPGN. The effects of MP + CTX therapy on improving renal dysfunctions may partially contribute to its amelioration of infiltrating cells in renal tissues. The degrees of CD4+, CD68+, and PCNA+ cells in the kidney may be useful indicators of MP + CTX therapy for RPGN.
OBJECTIVE: To investigate the changes and the clinical significance of plasma D-dimer in normal pregnant women and patients with pregnancy induced hypertension (PIH). METHODS: Using ELISA method, we measured the level of plasma D-dimer in 160 cases, including 106 normal pregnant women and 25 patients with mild and moderate PIH, 18 patients with preeclampsia and 11 patients with eclampsia. RESULTS: The value (2.27 +/- 0.92 mg/L) of plasma D-dimer in mild and moderate PIH patients was markedly higher than that of normal third trimester pregnant women (1.45 +/- 0.38 mg/L, P < 0.01). The value (3.09 +/- 1.65 mg/L) of plasma D-dimer in preeclampsia patients was markedly higher than that of mild and moderate PIH patients (P < 0.01). The value (5.62 +/- 1.34 mg/L) of plasma D-dimer in eclampsia patients was markedly higher than that of preeclampsia patients. CONCLUSION: Determination of plasma D-dimer level in second and third trimester pregnant women was of great significance in diagnosing early PIH and predicting the prognosis of PIH.
OBJECTIVE: To study the practicability of using red blood cells (RBC) as the carrier of SOD or GSH.Px and to observe the effects of RBC-SOD and RBC-GSH.Px on cerebral ischemia-reperfusion (I-R). METHODS: The method of hypotonic dialysis was used in encapsulating SOD or GSH.Px into rabbit RBC. The circulation half-lives (T1/2) of RBC-SOD and RBC-GSH.Px were determined in rabbits by 51Cr radiolabel. Lipid peroxidation (LPO) and nitric oxide (NO) in plasma and TTC staining of cerebrum were examined. RESULTS: T1/2 was 13.4 +/- 1.5 days (n = 6) for RBC, 13.7 +/- 1.0 days (n = 5) for RBC-SOD, and 7.3 +/- 2.5 days (n = 5) for RBC-GSH.Px. The concentration of NO reduced gradually as reperfusion continuing in control group, but it was a state of significant increase in NO concentration during the 4.5 hours of reperfusion, in RBC-SOD group. The increase of NO was inhibited by the administration of RBC-GSH.Px or (RBC-SOD + RBC-GSH.Px). The increase of LPO during cerebral I-R was inhibited by RBC-SOD or (RBC-SOD + RBC-GSH.Px). Staining with TTC showed that the RBC-SOD, RBC-GSH.Px and (RBC-SOD + RBC-GSH.Px) could effectively protect cerebrum For I-R damage, especially the last one. CONCLUSION: Using RBC as the carrier of SOD or GSH.Px is practicable. Intact RBC-SOD and RBC-GSH.Px could scavenge active oxygen produced during cerebral I-R.
