[Consumption of glucose under the influence of propranolol in human placentas perfused in vitro].

The consumption of glucose under the influence of various doses of propranolol in human placentas perfused in vitro was evaluated every 30 minutes during a two-hour perfusion. In the experiment the authors did not find a significant influence of propranolol on the level of the glucose consumption. The authors found a fourfold differentiation of glucose consumption between particular placentas. In the experiment these values differed from 2.63 mumol/g/h to 10.27 mumol/g/h.

[Placental perfusion. Observations based on examinations made at the Institute of Obstetrics and Gynecological Diseases of the Medical academy in Lublin in the years 1975-1984].

The paper presents the authors' experience resulting from the research into placental perfusion. The use of perfusion technique depended on a working hypothesis. The authors use the method of perfusion of foetal circulation. They analysed vascular resistance on the foetal side of the placenta, glucose consumption, production of lactic acid and pyruvic acid. They also analysed Dolantine, serotonin, papaverin, orciprenalin, phenoterol, theophylline ethylenediamine, isoxysuprin, propranolol, sodium fluoride. Basing on the results of the investigation of substances stimulating the betaadrenergic receptor, the authors came to the conclusion that there exists primary and secondary anoxia syndrome.

[Influence of betamethasone on glucose consumption in the human placenta perfused in vitro].

The study was carried out on 48 human placentas perfused in vitro. Betamethasone was administered into the arterial systems in the following single doses: 0.4 mg, 2 mg and 10 mg. The authors found a highly significant or statistically insignificant decreased glucose consumption after administering each dose of betamethasone and at all times of the experiment. The phenomenon of a decrease in glucose consumption was attributed to an increased gluconeogenesis and releasing glucose from placental glycogen. The action of betamethasone consisting in supplying an increased amount of glucose is of great importance in the process of maturing fetal lungs in the uterus and in states threatening the fetus.

[Metabolism of triglycerides of the placenta during extraorganic circulation].

An attempt has been made to evaluate the behaviour of triglycerides in the perfusional liquid of the human placenta under the influence of orciprenaline in the quantity of 40 micrograms/min and papaverine in quantity of 320 micrograms/min. In own experiments the authors discovered that these drugs which are given in the above mentioned doses increase the tension of the anaerobic metabolism of the placenta which is expressed by the quotient of lactate/pyruvate. The examinations were made on 30 placentas which were perfused extracorporally. In the control group of the placentas isotonic saline was administered. Concentration of triglycerides in the perfusion fluid was determined 30, 60, 90, 120 minute after beginning the experiment. Increase in the concentration of triglycerides after administration of orciprenaline was found and a constant decrease after the application of papaverine. Based on examinations of the metabolism of glucose in an identical experimental model and observations made by other authors a hypothesis was put forward that the increased synthesis of triglycerides is a reflection of the increasing tension of the anaerobic metabolism of the placenta. This process may constitute a compensatory mechanism of the metabolic activity of the placenta in the conditions of the limited accessibility of oxygen. The synthesis of triglycerides in ischemic tissue can perform two basic functions: 1. it assures constant regeneration of NADP, 2. it eliminates the endproducts of glycolyse transforming the pyruvate through the acetylo-CoA into the fatty acid and triglycerides. The increase of synthesis of triglycerides serves as replacement of oxygen and makes it possible to maintain the hexose-monophosphate shunt activity and Embden-Mayerhof pathway.(ABSTRACT TRUNCATED AT 250 WORDS)
