[Pathological changes of the blood vessels in rabbit femoral head with glucocorticoid-induced necrosis].

OBJECTIVE: To observe the pathological changes in the blood vessels in rabbit femoral head with glucocorticoid-induced necrosis and investigate the pathogenesis of glucocorticoid-induced osteonecrosis. METHODS: Twenty New Zealand white rabbits were randomly divided into two groups, namely group A. which was injected with horse serum and prednisone and group B as the control group. Chinese ink was injected into the femoral cavity of the rabbits to observe the blood vessels in the femoral head under optical microscope and the femoral head was examined histopathologically. RESULTS: Compared with the normal control group, the rabbits in group A had significantly decreased number of perfused vessels, which was featured by defective perfusion, osteocytie pyknosis or necrosis, increase of empty ostoocyte lacunae and fat cells, decrease of hematopoietic tissue, and blood vessel occlusion. CONCLUSION: Vascular occlusion and vasculitis due to glucocorticoid treatment may cause avascular necrosis of the femoral head.

The effects of sodium hyaluronate on mRNA expressions of matrix metalloproteinase-1, -3 and tissue inhibitor of metalloproteinase-1 in cartilage and synovium of traumatic osteoarthritis model.

OBJECTIVE: To observe the influence of intra-articular injection of sodium hyaluronate (HA) on the mRNA expressions of matrix metalloproteinase-1,-3 (MMP-1,-3) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in cartilage and synovium of traumatic osteoarthritis (OA). METHODS: Sixteen white rabbits underwent unilateral anterior cruciate ligament transection (ACLT) were divided into 2 groups randomly 5 weeks after transection. The experimental group rabbits received 0.3 ml of 1% HA by intra-articular injection once a week. Animals in the control group were treated under the same conditions using physiological saline. Ten weeks following surgery, cartilage and synovium were harvested. The mRNA expressions of MMP-1, MMP-3 and TIMP-1 were analyzed using reverse transcription-polymerase chain reaction (RT-PCR). RESULTS: In synovium, the mRNA expression of MMP-3 was suppressed in the HA injection group. HA treatment had no effect on the MMP-3 expression in cartilage. No significant difference of MMP-1 and TIMP-1 expressions in cartilage and synovium was found between the HA injection group and the control group. CONCLUSIONS: One of the mechanisms of the therapeutic effect of HA may be the inhibition of expression of MMP-3 in synovium during early stage of traumatic OA.

[Effects of carbachol on apoptosis of peripheral white blood cells and expression of cytokines in rats suffering from gut ischemia/reperfusion injury].

OBJECTIVE: To evaluate the effect of cholinergic drug carbachol on apoptosis and expression of certain cytokines of peripheral blood lymphocytes and neutrophils in rats subjected to ischemia/reperfusion injury of the intestine. METHODS: Thirty-six Wistar male rats were randomly divided into groups as follows: sham operation, 1 and 2 hour-gut ischemia, 1 hour-gut ischemia followed by reperfusion for 1 hour and 2 hours, carbachol + 1 hour-gut ischemia, and carbachol + 1 hour-gut ischemia followed by reperfusion for 2 hours. Then the gut was subjected to ischemia/reperfusion. At different time points after the said injury, the total number and differential count of leukocytes, apoptosis rate of lymphocytes and neutrophils, and the mRNA expression levels of certain cytokines in peripheral blood leukocytes, were determined. RESULTS: At 1 hour after gut ischemia, the total number of leukocytes decreased (the ratio of lymphocytes increased and that of neutrophils decreased), but it increased after gut reperfusion. Carbachol could reduce the apoptosis rate of lymphocytes, but enhance that of neutrophils after gut ischemia. In leukocytes, mRNAs expression of inflammatory (TNF-alpha) and antiinflammatory (IL-10) cytokines were upregulated at 1 hour after gut ischemia, while that of IL-10, IL-4 and interferon-gamma were down-regulated distinctly at 2 hours following reperfusion. All of these phenomena were ameliorated by giving carbachol. CONCLUSION: Carbachol could reduce the peripheral lymphocytes apoptosis subsequent to gut ischemia, and regulate the balance between inflammatory and antiinflammatory cytokines expression in leukocytes during gut ischemia and reperfusion. The results suggest that carbachol might be a potential therapeutic agent in preventing uncontrolled inflammatory response as a result of ischemia/reperfusion injury of the intestine.

[Construction of animal models of keloid by tissue engineering].

OBJECTIVE: To construct animal model of keloid by using tissue engineering and to explore its potential clinical significance. METHOD: Fibroblasts from keloid were isolated and cultured. After being cultured to generation six or eight, fibroblasts were inoculated into scaffolds constructed from copolymers of polylactic acid (PLA) and polyglycolic acid (PGA), and then cultured in rotatory cell culture system for 1 week prior to subcutaneous transplantation into 20 athymic mice. After 4, 8, 16, 24 weeks, the specimens were cut out and examined histologically. RESULTS: All the mice survived after surgery. The collagen patterns of all the keloids were remained after 24 weeks. A lot of fibroblasts and their secreted collagen were observed under light microscope. Enlarged rough endoplasmic reticula inside the fibroblasts were revealed by transmission electron microscopy. The fibroblasts in specimens remained the capability of synthesizing and secreting collagen. CONCLUSIONS There is a good affinity between PLA/PGA and fibroblasts. Complex of PLA/PGA and fibroblasts could be cultured into keloids in athymic mice.

[Clinical significance of serum transferrin receptor in differential diagnosis of anemia].

OBJECTIVE: To study the clinical significance of the serum transferrin receptor (sTfR) and ferritin in distinguishing between iron deficiency anemia (IDA), anemia of chronic disease (ACD) and ACD with complicating IDA (ACD-IDA). METHODS: One hundred and thirteen subjects enrolled into the study were divided into four groups including 28 health volunteers as control, 29 patients with IDA, 27 patients with ACD (sTfR level 20.0 nmol/L, ACD2 group). sTfR, serum ferritin (SF), serum iron (SI), total iron-binding capacity (TIBC), transferring saturation (TS) and complete blood count were measured in all the enrolled subjects or patients. Bone marrow iron stain was examined in 10 cases of ACD1 and 16 cases of ADC2. RESULTS: In IDA group, MCV (68.0 +/- 11.3) fl was the smallest; SI, TS and SF were significantly decreased but sTfR level was increased as compared with the control group (SI) (19.6 +/- 10.1) mg/L vs (81.7 +/- 30.6) mg/L, TS (5.5 +/- 2.3)% vs (27.0 +/- 12.0)%, SF (4.3 +/- 2.8) micro g/L vs (43.3 +/- 26.8) micro g/L and sTfR (67.2 +/- 40.3) nmol/L vs (15.6 +/- 4.1) nmol/L (P 0.05) but SF (627.3 +/- 40.3) micro g/L level was increased in ACD1 group as compared with the control group (P < 0.01). Bone marrow iron stain showed no iron deficiency in 10 cases of ACD1. In ACD2 group, SF (320.5 +/- 156.0) micro g/L level was higher than that of the control group, but lower than that of the ACD1 group (P <or= 0.01); bone marrow iron stain showed iron depletion in 14 of the 16 patients. CONCLUSION: High sTfR level may reflect iron deficiency status of the body and is a more sensitive parameter to diagnose iron deficiency anemia. sTfR level is less influenced by chronic inflammation, and able to distinguish ACD from ACD-IDA.

[Clinical research on the effect of Oxymatrine on serum cholinesterase].

BACKGROUND: To investigate the effect of Oxymatrine (OM) on serum cholinesterase (ChE) during the treatment of viral hepatitis and the relationship between the change of ChE and the change of albumin (ALB), prothrombin activity (PTA) and other liver function tests. METHODS: A total of 98 patients with viral hepatitis were divided into four groups. Group A consisted of 31 patients and were treated with OM intravenous infusion; Group B consisted of 30 patients, treated with OM orally; Group C consisted of 7 patients and were treated with OM intramuscular injection while Group D consisted of 30 patients, and were not treated with OM. ChE, ALB, PTA, liver function, renal function, soluble complement receptor-1 (sCR1) and erythrocyte innate immune adhesion function (EIIAF) were regularly determined. RESULTS: ChE in Group A,B,C was dropped obviously during the treatment (P less than 0.001, less than 0.001, 0.023=. But there were no change in ALB, PTA, sCR1, EIIAF (P greater than 0.05), and remarkable improvement of ALT, AST, TBiL was seen during the treatment in Groups A, B, C. After the treatment with OM, the level of ChE recovered soon. CONCLUSION: Serum level of ChE significantly declined during the treatment of viral hepatitis with OM, but no change was found in ALB, PTA, sCR1, EIIAF while liver function tests showed better results. So the drop of ChE does not mean deprivation of patient's liver disease.

Study on a non-contact life parameter detection system using millimeter wave.

OBJECTIVE: To study the basic principle and design method of a non-contact life detection system using 36 GHz millimeter wave. METHOD: Continuous wave radar techniques and signal processing techniques including zero intermediate frequency, frequency domain accumulation and adaptive noise cancellation were used in the system. RESULT: When the radiated power was about a milliwatt (mW) and detection range was up to 3 meters, the experimental results showed that this system can well detect the heartbeat and respiration signals from human subjects through common clothes and quilts. CONCLUSION: The system is feasible and effective due to its high detection sensitivity, small antenna, simple structure, easy miniaturization and safety to the human subject. It can be used for long-term continuous monitoring, especially suitable for the astronauts.

[The changes and significance of red cell nature-immune-adhesion function in liver diseases at different stages].

OBJECTIVE: To study the changes of red cells nature immune adhesion function (RNIAF) in liver diseases at different stages, and the feasibility of RNIAF in evaluating the severity of liver diseases. METHODS: Venous blood was extracted from 682 patients with liver disease, including cirrhosis, chronic hepatitis, acute hepatitis, hepatitis gravis, and 50 healthy blood donors as controls. Suspension of red cells in self-plasma was mixed with solution of mouse ascites carcinoma cells. A tumor cell attached with 5 red cells or 2 lymphocytes/granulocytes was counted as one rosette. The adhesion rate was calculated. Serum soluble complement receptor 1 (sCR1) was measured with a newly established sandwich enzyme-linked immunosorbent assay (ELISA). The CR1 expressed on erythrocytes has assayed by cell-ELISA. RESULTS: The expression of CR1 on erythrocytes was lower among the patients with liver diseases. Compared with that in healthy controls, RNIAF decreased among the patients with various liver diseases (all P < 0.01) in the order of acute hepatitis, chronic hepatitis, cirrhosis Child's A, cirrhosis Child's B, cirrhosis Child's C, and hepatitis gravis. CHE and PTA decreased and PT increased among the patients with cirrhosis and hepatitis gravis (all P < 0.05 or P < 0.01). RNIAF was positively correlated with CHE, PT, and PTA in patents with cirrhosis. CHE, PT and PTA remained at low levels while RNIAF almost returned to its normal level in the convalescents of hepatitis gravis. CR1 expressed on erythrocytes and sCR1 in serum in various liver diseases were decreased at different degrees, however, they changed significantly later than RNIAF. Serum sCR1 significantly increased in the patients with cirrhosis, particularly in those in grades Child's A to C. CONCLUSION: Low-cost, easy to operate and with early change and stable results, RNIAF may be used as an important indicator to evaluate the severity of liver diseases. of CHE, PT/PTA. It is an important to evaluate the severity of liver function in clinic.

[Changes of ECR1 genomic density polymorphism, quantitative expression and the activity of ECR1 natural adhesion in patients with chronic hepatitis].

OBJECTIVE: To study the changes of genomic density polymorphism, quantitative expression and the adhesion activity of complement receptor type 1 (ECR1) on erythrocytes in patients with chronic hepatitis. METHODS: Polymerase chain reaction (PCR) and Hind restriction enzyme digestion, the quantitative assay of ECR1 and the activity of erythrocytes immune adhesion test were applied. RESULTS: The spot mutation rate (25.0%-30.3%) of ECR1 density gene in patients with chronic hepatitis was not significantly different from that of healthy individuals (28.0%). The amount of ECR1 in patients with chronic hepatitis, except for the diseases with normal liver function, was significantly lower than that of healthy individuals (t=9.87,P<0.000 1). The quantitative expression of ECR1 in decompensated cirrhosis was obviously lower than that of compensated cirrhosis (t=2.21,P<0.05). CONCLUSIONS: Defective expression of ECR1 in chronic hepatitis B may be acquired through central and/or peripheral mechanisms. It is very important to study the quantitative expression in the patients with chronic hepatitis.

[Development of an analysis system for X-ray cephalometry by Windows platform].

Here in this paper introduced is an analysis system for X-ray cephalometry developed through Windows platform,which includes three parts: data input, cephalometry analysis & calculation, and display & printing of diagnostic results. It has been applied in orthodontic department of Stomatological Hospital of the Fourth Military Medical University, and it is quite convenient for doctors' cephalometry diagnosis.
