Interactions between secreted GRA proteins and host cell proteins across the paratitophorous vacuolar membrane in the parasitism of Toxoplasma gondii.

Interactions between GRA proteins of dense granules in Toxoplasma gondii and host cell proteins were analyzed by yeast two-hybrid technique. The cMyc-GRA fusion proteins expressed from pGBKT7 plasmid in Y187 yeast were bound to host cell proteins from pGADT7-Rec-HeLa cDNA library transformed to AH109 yeast by mating method. By the selection procedures, a total of 939 colonies of the SD/-AHLT culture, 348 colonies of the X-alpha-gal positive and PCR, 157 colonies of the X-beta-gal assay were chosen for sequencing the cDNA and finally 90 colonies containing ORF were selected to analyze the interactions. GRA proteins interacted with a variety of host cell proteins such as enzymes, structural and functional proteins of organellar proteins of broad spectrum. Several specific bindings of each GRA protein to host proteins were discussed presumptively the role of GRA proteins after secreting into the parasitophorous vacuoles (PV) and the PV membrane in the parasitism of this parasite.

How does light intensity influence evening dressing behavior in the cold?

Core temperature (tympanic and rectal temperatures) is lowered for several hours under diurnal bright light exposure and its evening fall is inhibited under evening bright light exposure. Melatonin may be involved in the behavior of these core temperatures. Diurnal bright light exposure for several hours may make dressing behavior and thermal sensibility in the evening cold slower and dull, compared with diurnal dim light exposure. On the contrary, evening bright light exposure for several hours may make the dressing behavior and thermal sensibility in the evening cold quicker and sharper, compared with evening dim light exposure. The underlying physiological mechanisms for these findings are that the thermoregulatory set-point would be reduced more markedly in the evening under the influence of higher elevation of melatonin under the diurnal bright light exposure, and its evening decline would be inhibited by suppression of the nocturnal rise of melatonin under evening bright light exposure.

The effect of safety hat on thermal responses and working efficiency under a high temperature environment.

The purpose of this study is to examine the effect of a safety hat on thermal responses and work efficiency under a high temperature environment. Five healthy male subjects participated in the repeated 'Rest' and 'Exercise' periods in order to compare a safety hat without holes (annoted as 'without hole') and a safety hat with holes (annoted as 'with hole') in a climatic chamber of 30 degrees C, 50%RH. The main findings are as follows: (a) the core temperature (tympanic temperature) and heart rate showed significantly lower levels in the subjects who are under the 'with hole' condition than those who are under the 'without hole' condition; (b) the forehead skin temperature was significantly higher in the subjects who are under the 'without hole' condition than those who ar uder the 'with hole' condition; (c) blood pressure was significantly lower in the 'with hole' condition; and (d) sweat rate which was measured by weight loss before and after the experiment was higher in the 'without hole' condition; and (e) work ability which was measured by a grip strength dynamometer was higher in the 'with hole' condition. Making a hole in the safety hat, designed for proper ventilation and hygiene, is practical in letting out heat and decreasing the physiological burden under a hot working environment. The safety hat with holes is useful in maintaining the homeostasis of the body temperature by releasing body heat efficiently and it is meaningful to keep the working efficiency.
