[Effects of finasteride on capillary in the ventral prostate of rat].

OBJECTIVE: To investigate the effects of finasteride on capillary in the ventral prostate of rat and study the mechanisms therein involved, METHODS: Male SD rats were given via gavage finasteride 40 mg/kg per day for 7 days (group B) and for 14 days (group C). The changes of capillary infused with Chinese ink were observed with the use of HE staining. Immunohistochemical (IHC) SP-method was adopted in measuring the protein expression of VEGF and eNOS in rat prostatic tissue. RESULTS: In comparison with the relevant values measured in the control group (group A), the microvessel density (MVD) values in group B and group C decreased by 35.2% and 56% respectively, the protein expression of VEGF in group B and group C decreased by 31.7% and 62.6% respectively, the protein expression of eNOS in group B and group C decreased by 15.7% and 52.5% respectively. CONCLUSION: Finasteride can inhibit the protein expression of VEGF and eNOS and thereby can suppress angiogenesis of capillary in the ventral prostate of rat.

[The influence of IL-3 on the differentiation and development of dendritic cells from mouse bone marrow].

In order to compare the effects of GM-CSF + IL-4 with the effects of IL-3 + IL-4 on the differentiation and development of dendritic cells (DCs) from mouse bone marrow, we cultured bone marrow hematopoietic progenitor cell in vitro and examined whether DCs induced by IL-3 + IL-4 (IL-3 DCs) differed from the DCs induced by GM-CSF + IL-4 (GM-CSF DCs) in morphology, yields, phenotype and endocytic activity. Scanning electron microscope, laser scanning confocal microscope (LSCM) and flow cytometer (FCM) were used. The results showed that the DCs induced by IL-3 + IL-4 and the DCs induced by GM-CSF + IL-4 had similar morphology, and the DCs induced by IL-3 + IL-4 expressed high level of major histocompability complex (MHC) class II. Besides, IL-3 DCs had the characteristics of higher yields and better consistency. But CD80 and CD86 were expressed at low levels or absent on IL-3-treated DCs. The capability of uptaking antigen of IL-3 DCs was more powerful than that of GM-CSF DCs. IL-3 could substitute for GM-CSF in culturing DCs from mouse bone marrow cells and could obtain the tolerance DCs that lack costimulatory molecules.

[Biological characteristics of human embryonic myoblasts in vitro].

This study was intended to observe the biological properties of primary human embryonic skeletal myoblasts cultured in vitro and to inquire about the possible differentiation mechanism. Isolated human embryonic myoblasts were identified by morphology and myosin immunohischemical staining. The proliferating myoblasts were shifted to DM (DMEM supplemented with 3% fetal bovine serum) to induce differentiation. The control group was cultured in GM (DMEM supplemented with 10% fetal bovine serum). The differentiation was tested by the rate of myotube formation (RMF); The change of cell cycle was tested by flow cytometry; the morphological features were observed by use of inverted microscope and TEM. The myogenic regulatory factors (MRFs) such as MyoD, Myogenin and Myf5 were assayed by RT-PCR. Results showed that the primary myoblasts cultured in DM, in comparison with those cultured in GM, had higher rate of myotube formation (RMF) and myofilament formation, and more of the myoblasts cultured in DM exited the S phase. The expression level of Myogenin mRNA was obviously higher than that of the control group, the increase of MyoD mRNA expression level was not so high, however, the expression of Myf5 mRNA was decreased. These data indicate that there is a possible mechanism between differentiation and cell cycle. Besides, the mRNA expression of myogenic regulatory factors (MRFs) occurs in different phase of differentiating myoblastes and implicates the diverse biological function.

[Role of cytokeratin expression in differential diagnosis of intraductal proliferative lesions of breast].

OBJECTIVE: To evaluate the expression of cytokeratins in intraductal proliferative lesions of breast, including usual ductal hyperplasia (UDH), atypical ductal hyperplasia (ADH), ductal carcinoma-in-situ (DCIS) and its role in differential diagnosis. METHODS: Ninety two cases of paraffin-embedded lesional breast tissue, 30 cases of frozen samples, cell cultures of hyperplastic ductal cells and 2 invasive ductal carcinoma cell lines (T47D and MCF-7) were used for this study. Immunohistochemistry was performed using EnVision method for 34betaE12, CK8 and CK14. RESULTS: The percentage of 34betaE12-positivity in paraffin-embedded samples of UDH, ADH, DCIS and invasive ductal carcinoma (IDC) was found to be 95.2%, 33.3%, 19.2% and 12.5% respectively. In frozen tissues, all UDH cases and 55% of IDC cases expressed 34betaE12. The primary UDH cell cultures and T47D cell line were also 34betaE12-positive, whereas MCF7 cell line showed negative staining. The expression rate of CK8 and CK14 in UDH was also different from that in ADH and DCIS. CONCLUSIONS: 34betaE12 can be useful in differential diagnosis of intraductal proliferative lesions of the breast. However application of this cytokeratin stain in intraoperative frozen sections is relatively limited. The expression patterns of CK8 and CK14 are also helpful in the differential diagnosis of similar lesions.

[Solitary fibrous tumor: the clinicopathologic and immunohistochemical characteristics of 26 cases].

OBJECTIVE: To study the clinicopathologic and immunohistochemical features of solitary fibrous tumor (SFT) and its biologic behaviour. METHODS: Clinicopathologic data of 26 cases were collected and analyzed. Among them, immunohistochemical staining (Envision method) for CD34, vimentin, CD99, bcl-2, S-100, SMA, HBME-1, EMA and Ki67 were performed in 23 cases. RESULTS: The 17 male and 9 female patients were between 15 and 66 years of age (mean, 44). Their SFTs were located in nose and nasopharynx, cranial cavity, soft tissue, mediastinum, and parietal and visceral pleura. The main clinical manifestations were local mass and pressure symptom. The characteristic microscopic features included patternless growth pattern, alternating hyper- and hypo-cellular areas, blunt spindle cells within keloid-like hyalinization, and hemangiopericytoma-like regions. Positive immunohistochemical staining: vimentin 100%(23/23), CD34 82%(19/23), bcl-2 87%(20/23), CD99 100%(23/23), SMA 30%(7/23). All cases were negative for S-100, HBME-1 and EMA. Follow-up information on 13 cases revealed that 3 patients had had relapse and died, the other 10 patients were alive without evidence of recurrence. CONCLUSION: SFT is a rare mesenchymal spindle cell tumor which may be found in various parts of human body and needs to be distinguished from other spindle cell tumors by differential diagnosis. The immunophenotype can be of help in this connexion. About 10% to 23% SFTs have malignant behaviour, manifesting as local recurrence or metastasis. The behavior of SFT is unpredictable, which requires careful, longterm follow-up.

[Interobserver reproducibility in the pathologic diagnosis of borderline ductal proliferative breast diseases].

OBJECTIVE: The aim of this study involving general pathologists was to assess interobserver reproducibility in the pathologic diagnosis of borderline ductal proliferative breast diseases. METHODS: Ten general pathologists independently reviewed 43 specimens chosen to represent the spectrum of borderline ductal proliferative breast lesions. All slides were blindly reviewed without given standardized criteria, and were classified as either mild usual hyperplasia, moderate-severe usual hyperplasia, mild atypical hyperplasia, moderate-severe atypical hyperplasia, ductal carcinoma in situ, or ductal carcinoma in situ with invasion. According to the years of training, these 10 general pathologists were divided into the experienced group and the less trained group. Interobserver agreement was statistically analyzed using Kappa statistic. Then, by comparing all the diagnoses of individual pathologist with the consensus opinion confirmed by two breast pathologists in terms of Page standard, we acquired the diagnostic accuracy and ascertained the undue diagnosis. RESULTS: The ten general pathologists' interobserver reproducibility in the diagnosis of borderline ductal proliferative breast diseases was rather low, especially that in their diagnosis of atypical hyperplasia. The reproducibility and accuracy were slightly higher in the experienced pathologists than in the less trained pathologists. Some pathologists made over-diagnosis or under-diagnosis of the lesions to different degrees. However, when the categories of diagnostic terms were simplified, the interobserver reproducibility increased. CONCLUSION: The use of standardized criteria is an important approach to increasing the diagnostic reproducibility and accuracy.

[Regulation of growth inhibition by transforming growth factor beta1 in rhabdomyosarcoma RD cell line].

OBJECTIVE: To study the regulatory effect of TGF-beta1 on growth of rhabdomyosarcoma RD cell line. METHODS: After various durations of TGF-beta1 treatment, the viability of RD cell line was examined by growth rate measurement, MTT assay and (3)H-thymidine incorporation. The cell cycle was analyzed by flow cytometry. Immunofluorescent staining was used to localize p15, p21 and p27 in RD cell line under laser scanning confocal microscope. The protein and mRNA of p15, p21 and p27 in RD cell line were detected by western blot and reverse transcriptase-polymerase chain reaction respectively. RESULTS: The viability of RD cell line treated with TGF-beta1 was obviously decreased. RD cell line was arrested in G(1) phase by TGF-beta1. There was increased expression of p21 and p27 in RD cell line with TGF-beta1 treatment at protein and mRNA levels. The expression of p21 in RD cell line was seen in both nucleus and cytoplasm after 24 hours of TGF-beta1 treatment. The expression of p15 showed no obvious changes upon TGF-beta1 treatment. CONCLUSIONS: TGF-beta1 inhibits growth of RD cell line and induces G(1)-arrest. It up-regulates protein and mRNA of p21 and p27 and shows no obvious influence on p15 expression. The growth arrest of RD cell line may result from the up-regulation of p21 and p27 by TGF-beta1.

[Progress in application of laser scanning confocal microscope to studies on dendritic cells].

The defensive function of immune response in vivo is very important. Antigen presenting cells (APC) is the hinge in starting specific immune response. Dendritic cells (DC) are the most potent APC, which participate in double-regulating the immune response. Laser scanning confocal microscope (LSCM) becomes a powerful tool in the fields of basic medicine and clinical medicine with its high resolving power, high sensitivity, optical section, three-dimensional reconstruction and dynamic analysis. And it makes the study on DC more thoroughly and deeper. In this paper, we review the recent progress in the applications of LSCM to studies on the morphology, immunofluorescence localization and the functional analysis of dendritic cells.

Systematic analysis of the TGF-beta/Smad signalling pathway in the rhabdomyosarcoma cell line RD.

Transforming growth factor-beta (TGF-beta) is a multifunctional regulator of cell growth and differentiation, whose actions are highly cell type specific. To study the role of the TGF-beta1 autocrine loop in regulating growth and myogenic differentiation in the human rhabdomyosarcoma cell line, RD, an attempt was made to establish a framework for the expression of several components of TGF-beta1/Smad signalling pathway at the mRNA and protein levels by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot analysis in RD cells compared with the normal myoblasts. Higher exogenous concentration of TGF-beta1 was necessary to reach a growth-inhibition effect, whereas TGF-beta1 downregulated the expression of myosin heavy-chain mRNA at lower concentrations than that was required for growth inhibition. Treatment with TGF-beta1 significantly decreased the number of sarcomeric actin and myosin-expressing cells. In this study, we have shown that RD cells displayed higher expression of TbetaRI, TbetaRII, Smad2 and Smad4 at both the mRNA and protein levels than myoblasts. Smad3 and Smad7 mRNA were expressed at higher level in RD cells than in myoblasts. The staining patterns of TbetaR and Smads suggest that they may transduce different TGF-beta1 signalling in RD cells than in myoblasts. TGF-beta1 signalling induced a rapid relocation of Smad2 to the nucleus; in contrast, Smad4 remained localized to the cytoplasm unless it was coexpressed with Smad2. These studies suggest that signalling from the cell surface to the nucleus through Smad proteins is a required component of TGF-beta1-induced cell response in RD cells. The RD cell line is a suitable model to study the TGF-beta autocrine loop involved in growth and differentiation of RMS.

[Specific loss of heterozygosity of chromosome 3p loci in soft tissue leiomyosarcoma].

OBJECTIVE: To analyze the loss of heterozygosity (LOH) at 5 loci on chromosome 3p in soft tissue leiomyosarcoma (LMS). METHODS: LOH was detected in 22 cases of LMS using PCR-silver staining targeting 5 microsatellite sites on 3p14.2-pter. Relation between LOH and LMS clinical pathological features was also analyzed. RESULTS: Ten of 22 LMS samples showed LOH at more than one locus (45.4%). Among the 5 loci, LOH occurred more frequently at D3s1295 (36.8%) and D3s1289 (10.5%), but absent at D3s1293. No significant difference was found on LOH incidence between different grade, size and location of LMS. CONCLUSIONS: LOH on chromosome 3p14.2-23 region is relatively frequent in LMS. Region around D3s1295 and D3s1289 may harbor tumor suppressor gene relating to LMS.

[Study on rabbit mesenchymal stem cells differentiation to the adipogenic or osteogenic lineage in vitro].

Rabbit bone marrow-derived mesenchymal stem cells(MSCs) are multipotent. We studied the adipogenic and osteogenic differentiation potent using adipogenic supplement (AS) or osteogenic supplement (OS) in vitro. Specific markers of this induced adipogenic and osteogenic lineage were identified. The findings showed that the rabbit MSCs are capable of differentiating into adipogenic and osteogenic lineages spontaneously. On the 21st day, approximately 75% rabbit MSCs were induced to adipogenic or osteogenic cells in medium containing AS or OS, respectively. These results demonstrated that the differentiation of MSCs could be regulated in vitro. The underlying molecular mechanisms of adipogenic or osteogenic differentiation await elucidation.

[Comparative study on image analysis and manual counting of immunohistochemistry].

With the use of computer image analysis (IA) and manual counting, the immunohistochemistry (IHC) results on PS2, ER and c-erB-2 of breast cancer which represent positive expression of cytoplasm, cell nucleus and cytomembrane are analyzed and compared. Then the advantages, disadvantages and repetition of the two methods are discussed and the correlation between image analysis and manual counting is estimated. The results of this study indicate that IA has better repetition in estimation of positive expression of cytoplasm, cell nucleus and cytomembrane, whereas manual counting has better repetition only in the positive expression of cell nucleus and cytomembrane, and the two methods have positive correlation in estimation of IHC results. In addition, some problems in image processing are discussed in this paper.

[Biological characteristics of human umbilical artery smooth muscle cells cultured in vitro and the preestablishment of immortalized cell line].

OBJECTIVE: To establish the best primary and propagated culture approach of the human umbilical artery smooth muscle cells (HUASMC) and immortalized the primary cells. METHODS: The primary culture of HUASMC was performed using the primary-explant method, and the effects on the cells' growth characteristic by different culture mediums were conducted. Primary cultures of HUASMC derived from normal human umbilical arteries were transfected with recombination plasmid containing human telomerase reverse transcriptase (hTERT) open reading frame. The transfected HUASMC was analyzed for cell biological characteristics, and growth characteristics, and specific alpha-actin. RESULTS: Highly purified primary HUASMC were gained and could be propagated about 10 population doublings (PD). After PD 4, the cells lost the proliferation ability gradually. However, the transfected HUASMC could be propagated at least 20 population doublings and could express specific alpha-actin. CONCLUSION: The hTERT transfected HUASMC could keep the basic biological characteristic of smooth muscle cells and prolonged life span at least 20 PD. It might be a good cell model for the study of cardiovascular disease, myoma of uterus and chronic graft disfunction. It might also bring the hope to engineered vascularization in the future.

[The quantity analysis of reverse transcriptase in porcine endogenous retrovirus expressed in Banna minipig inbred].

Quantitative RT(reverse transcriptase) assay was established to detect the reverse transcriptase in plasma of thirty-four Chinese Banna minipig inbred in this work. The protocol was given in the RT kit (Roche), using HIV-1 as the positive control of the kit and supernatant of PK-15 as the PERV positive control respectively. The results show that positive reverse transcriptase reaction can be detected in the plasma of the pigs, but the levels are much lower than that of HIV-1 and lower than that of PERV in supernatant of PK-15.

Differentiation between intestinal tuberculosis and Crohn's disease in endoscopic biopsy specimens by polymerase chain reaction.

OBJECTIVES: It is difficult to differentiate intestinal tuberculosis from Crohn's disease because of similar clinical, pathological, radiological, and endoscopic findings. The purpose of this study was to investigate the value of polymerase chain reaction (PCR) assay in the differentiation intestinal tuberculosis from Crohn's disease, and compare the histopathological features of endoscopic biopsy of the two disorders. METHODS: A total of 39 endoscopic biopsy specimens from patients with intestinal tuberculosis and 30 specimens from patients with Crohn's disease were subjected to pathological analysis retrospectively, Ziehl-Neelsen stain, and PCR assay. RESULTS: Except for granuloma with caseation and confluence, which was the characteristic of intestinal tuberculosis, other pathological features of intestinal tuberculosis and Crohn's disease were very similar or were difficult to find in endoscopic biopsy specimens. The positivity rate by PCR in 39 intestinal tuberculosis specimens was 64.1% (25/39), but was zero by PCR in 30 Crohn's disease specimens. Moreover, in the tissues of intestinal tuberculosis with granulomas similar to those of Crohn's disease, there were 71.4% (10/14) positive by PCR, and there were 61.1% (11/18) positive in intestinal tuberculosis tissues without granulomas. CONCLUSIONS: Biopsy is of limited diagnostic value in the differentiation intestinal tuberculosis from Crohn's disease, and PCR is valuable in the differentiation between intestinal tuberculosis and Crohn's disease.
