Genomic analyses implicate hormonal and metabolic dysregulation in polycystic ovary syndrome.

Polycystic ovary syndrome (PCOS) and its underlying features remain poorly understood. In this genetic study (n = 544,513), we expand the number of genetic loci from 16 to 29, and additionally identify 31 associated plasma proteins. Many risk-increasing loci were associated with later age at menopause, underscoring the reproductive longevity related to an increased oocyte number and/or availability across the lifespan. Hormonal regulation in the etiology of this condition, through metabolic and reproductive features, was emphasized. The proteomic analysis highlighted metabolic biology known to be related to PCOS. A polygenic risk score (PRS) was associated with adverse cardiometabolic outcomes, with differing relevance of testosterone and body mass index in women and men. Finally, while oligo-anovulation and anovulatory infertility are features of PCOS, we observed no impact of PCOS susceptibility on childlessness. We suggest that PCOS susceptibility confers balanced pleiotropic influences on fertility in women, and life-long adverse metabolic consequences in both sexes.

ADHD medication use in a population-based sample of twins.

OBJECTIVE: To determine treatment patterns for youth attention-deficit/hyperactivity disorder (ADHD) symptoms in a general population sample of 1,610 twins. METHOD: Twin pairs ages 7 to 17 years and parents ascertained from birth records in the state of Missouri were interviewed using the Missouri Assessment of Genetics Interview for Children between 1996 and 2001. Multivariate logistic regression analyses were used to determine which factors were significant in answering three questions about treatment for ADHD. RESULTS: Of 302 boys, 177 (58.6%) and 26 of 57 girls (45.6%) who met full DSM-IV criteria for ADHD received stimulant medication in this sample. Of 314 youths, 111 (35.4%) who received stimulant medication did not meet DSM-IV criteria for ADHD. When controlling for comorbidity and other factors, the number of impairing ADHD symptoms and having a cotwin who was also brought to treatment for ADHD correlated with referral or treatment in youths without ADHD. Youths without ADHD who were treated had a large number of ADHD symptoms. CONCLUSION: As is shown in earlier studies, children with ADHD are being undertreated. Complex factors, including comorbid disorders and family history of ADHD treatment, affect treatment patterns in the general population.

[Morphologic quantitative analysis of nucleus in laryngeal squamous cell carcinoma with Automatic Image Cytometry].

OBJECTIVE: Analyze morphological character of nucleus in laryngeal squamous cell carcinoma with Automatic Image Cytometry, discuss the relation between morphological parameters and tumor clinic. METHOD: We discussed morphological character of nucleus from 83 cytology specimens which included 21 laryngeal squamous cell carcinomas and 20 benign diseases with Automatic Image Cytometry. RESULT: Nucleus from laryngeal squamous cell carcinomas had bigger nuclear area nuclear radius, nuclear perimeter and more DNA content per nuclear area than from laryngeal benign diseases, furthermore, these parameters showed distinct difference in different anatomic positions and different pathologic types. We concluded that nuclear areas of nucleus in S phase from laryngeal squamous cell carcinomas are more than 77 microm, Second Order Regression Equation(Y =aX2+bX+c) described scattered diagrams between mean nuclear radius and nuclear area of laryngeal cytology specimens with different larynx pathology and different cell phase in cell cycle. The more the value of coefficient "c"(c>1.82) and the less the value of coefficient "b" (b< 0.045), the more the probability of malignant tumor diagnosis. CONCLUSION: Morphologic quantitative analysis parameters, such as nuclear area, mean nuclear radius, nuclear perimeter and DNA content per nuclear area, are suitable for diagnosing laryngeal squamous cell carcinomas. They could offer valuable morphologic quantitative criterion in assisting cytology diagnose and evaluating tumor clinical characters when these parameters associated with nuclear area of nucleus in S-phase and the coefficients of Second Order Regression Equation.

Overexpression of mouse GlcNAc-1-phosphotransferase-gamma subunit in cells induced an I-cell-like phenotype of mucolipidosis.

In a screen of signal peptide-containing proteins from a mouse hypothetical protein library, we identified the mouse UDP-GlcNAc:lysosomal enzyme N-acetylglucosamine-1-phosphotransferase-gamma chain (GlcNAc-1-phosphotransferase-gamma) (GenBank accession no. , HYP36 in this study). The mouse GlcNAc-1-phosphotransferase-gamma was localized in the Golgi complex in cells and was expressed ubiquitously in mouse tissues, as shown by fluorescence microscope analysis and a semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) assay, respectively. Domain analysis showed that the mouse GlcNAc-1-phosphotransferase-gamma had a conserved mannose-6-phosphate (M-6-P)-binding domain. Interestingly, we found that overexpression of the mouse GlcNAc-1-phosphotransferase-gamma in fibroblast cell line NIH-3T3 induced accumulation of macromolecules, formation of large cytoplasmic vacuoles and decrease of lysosomal enzymes in cells. This phenotype was reminiscent of inclusion cells (I-cells) that were reported in mucolipidosis diseases caused by abnormal sorting of lysosomal proteins. Transient ectopic expression of GlcNAc-1-phosphotransferase-gamma in endoplasmic reticulum (ER) induced lowered lysosomal enzyme activity in cells. These results suggested on one hand that GlcNAc-1-phosphotransferase-gamma is an essential subunit of the GlcNAc-1-phosphotransferase, and on the other hand, the molecule might not only recognize the substrates of GlcNAc-1-phosphotransferase, but also the lysosomal proteins with M-6-P residuals.

Collaborative analysis of DRD4 and DAT genotypes in population-defined ADHD subtypes.

BACKGROUND: It has been proposed that some of the variability in reporting of associations between attention deficit hyperactivity disorder (ADHD) and candidate genes may result from mixing of genetically heterogeneous forms of ADHD using DSM-IV criteria. The goal of the current study is to test whether population-based ADHD subtypes defined by latent class analysis help resolve issues of variable findings across individual gene association studies. METHODS: Three studies which had previously reported no associations between polymorphisms of the DRD4 and DAT genes and DSM-IV defined ADHD were reanalyzed using population-based and DSM-IV defined ADHD subtypes. RESULTS: Across studies no significant associations were found for either DRD4 or DAT polymorphisms using DSM-IV ADHD subtypes. In contrast, a significant association was found between the combined data set for the 440 base pair 3' DAT VNTR polymorphism and population-defined severe combined ADHD (OR=1.25, p=.01). A marginally significant association was also found between the 7 repeat DRD4 allele and population-defined severe combined ADHD. CONCLUSION: Use of alternative population-based defined ADHD subtypes may help resolve some of the variable results presented for candidate gene association studies in ADHD.

[DNA quantitative analysis of head and neck tumor].

OBJECTIVE: This research discussed the diagnostic value of cellular quantitative analysis in head and neck tumor. METHOD: We analysis quantitative parameters of 23 malignant tumors and 20 benign diseases with automated DNA image cytometry, they are the DNA Index (DI), S-phase Fraction (SPF), Proliferation Index (PI) and 5c Exceeding (5c Ex). RESULT: The masculine rate of cellular quantitative analysis and conventional method are 76.92% and 50.00%. The value of SPF, PI and 5c Ex are 14.05%, 19.59% and 1.59% in malignant tumor, respectively. The parameters of SPF and PI showed distinct difference in the area of malignant tumor than the 1 cm sideward area of tumor and the edge of tumor. CONCLUSION: Automatic DNA image cytometry has higher masculine rate of malignant tumor than conventional method in early detection of tumor. The cellular quantitative parameters, such as SPF, PI and 5c Ex, are able to guide clinical work, specially in diagnosing of head and neck tumor, estimating biological character of malignant tumor and evaluating the area of excision.
