Distinct regulatory elements mediate the dynamic expression pattern of Nkx3.1.

Loss of Nkx3.1 function in mice results in defects in prostate development and epithelial hyperplasia, indicating that this gene plays important roles in both the initiation and maintenance of prostate differentiation. In humans, decreased NKX3.1 expression is associated with the progression of prostate cancer. Despite these roles in prostate development and disease, the transcriptional regulation of Nkx3.1 has not been systematically addressed. A reporter gene approach in transgenic mice was used to identify regulatory regions that dictate the expression pattern of Nkx3.1. A 32-kb DNA fragment from the Nkx3.1 locus that specifies the expected expression pattern during embryogenesis and postnatal life has been identified. Deletion analyses demonstrated that cis-regulatory elements that mediate expression in distinct sites are separable. A 5-kb fragment downstream of the Nkx3.1 coding region contains elements that support expression in the prostate and bulbourethral glands, whereas an upstream fragment contains elements that direct expression in somites and testes. Reporter gene expression analyses also revealed several previously unknown sites of Nkx3.1 expression in males, including urethral glands, glandular cells in the urethral diverticulum and basal epithelial cells in the prostate. In addition, these analyses revealed Nkx3.1 expression in female urethral glands. The identification of Nkx3.1 cis-regulatory elements provides a unique starting point to dissect signaling pathways involved in prostate organogenesis and pathogenesis and provides a system to perturb gene expression throughout prostate development.

Quantitation of synthetic polymers using an internal standard by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry.

MALDI-TOF MS is utilized to perform quantitative analysis on synthetic polymers. Despite the inherent limitations of MALDI, good quantitative results have been obtained in the three sets of experiments described here. An internal standard with similar molecular properties as the analytes is introduced. Plots of relative integrated intensity ratios as a function of theoretical ratios of stoichiometry are drawn based on the results. The satisfactory slopes and correlation coefficients illustrated the practicality of quantitative measurement by MALDI-TOF MS.

The cationization mechanism study of novel oligo (p-phenyleneethynylene)s.

Two series of new oligo(p-phenyleneethynylene)s (OPEs) O1-O4 and O5-O8, which have been proven to be one of the chief classes of molecules mainly used as the wires and other potential backbones of molecular electronic devices, have been synthesized by stepwise synthetic approach. The characterization of these oligomers was performed on MALDI TOF MS. Different cationization salts have been applied to investigate the ionization processes of these series of oligomers under MALDI conditions. The experimental results show that these oligomers display a strong tendency to undergo radical cationization and varied ionization efficiency with different cationization agents attributable to their difference in cationic diameters. Furthermore, we found that these two series of oligomers differed in ionization properties because of their different end-groups even when the same cationization agent was used.

Effect of short-term cigarette smoke exposure on body weight, appetite and brain neuropeptide Y in mice.

Although nicotinic receptors have been demonstrated in hypothalamic appetite-regulating areas and nicotine administration alters food intake and body weight in both animals and humans, the mechanisms underlying the effects of smoking on appetite circuits remain unclear. Conflicting effects of nicotine on the major orexigenic peptide, neuropeptide Y (NPY), have been observed in the brain, but the effects of smoking are unknown. Thus, we aimed to investigate how cigarette smoking affects body weight, food intake, plasma leptin concentration, hypothalamic NPY peptide, adipose mass and mRNA expression of uncoupling proteins (UCP), and tumor necrosis factor (TNF) alpha. Balb/C mice (8 weeks) were exposed to cigarette smoke (three cigarettes, three times a day for 4 consecutive days) or sham exposed. Body weight and food intake were recorded. Plasma leptin and brain NPY were measured by radioimmunoassay. UCPs and TNF alpha mRNA were measured by real-time PCR. Food intake dropped significantly from the first day of smoking, and weight loss became evident within 2 days. Brown fat and retroperitoneal white fat masses were significantly reduced, and plasma leptin concentration was decreased by 34%, in line with the decreased fat mass. NPY concentrations in hypothalamic subregions were similar between two groups. UCP1 mRNA was decreased in white fat and UCP3 mRNA increased in brown fat in smoking group. Short-term cigarette smoke exposure led to reduced body weight, food intake, and fat mass. The reduction in plasma leptin concentration may have been too modest to increase NPY production; alternatively, change in NPY or its function might have been offset by nicotine or other elements in cigarette smoke.

Detection of cytokeratin 20 mRNA in the peripheral blood of patients with colorectal cancer by immunomagnetic bead enrichment and real-time reverse transcriptase-polymeras chain reaction.

BACKGROUND: Detection of circulating cancer cells is a useful indicator for the risk of recurrence of advanced carcinoma. The aim of the present study was to evaluate the potential value of a novel approach to detect the circulating cancer cells in patients with colorectal cancer. This method is based on a combination of isolation of epithelial cell by a combination of negative and positive immunomagnetic beads with detection of cytokeratin 20 (CK20) mRNA by reverse transcriptase-polymeras chain reaction (RT-PCR). METHODS: Peripheral blood samples were collected from 40 patients with colorectal carcinoma on the day before operation or chemical therapy. Mononuclear cells (MNC) were isolated by centrifugation through a Ficoll gradient. Each MNC sample was equally divided into three parts and then CD45 immunomagnetic beads and/or Ber-EP4 immunomagnetic beads were used to enrich colon cancer cells. Finally, the CK20 mRNA was detected by real-time quantitative RT-PCR. As a control, LS174T colon cancer cells were serially diluted with blood from healthy individuals. RESULTS: When CD45 and Ber-EP4 immunomagnetic beads were used successively, a significant correlation between CK20 mRNA levels and the initial cell concentrations was found in the control recovery experiment. The sensitivity of the assay was one cancer cell in 1 mL healthy blood. In the patient group, CK20 mRNA was detected in 80.0%, 82.5% and 72.5% of patients when CD45, Ber-EP4, and CD45/Ber-EP4 immunomagnetic beads were used, respectively. The positive detection rates of patients with colorectal carcinoma at Dukes A, B, C, and D stage were 0.0% (0/2), 33.3% (3/9), 86.7% (13/15), and 92.9% (13/14), respectively. The CK20 mRNA positive detection rate in peripheral blood was significantly correlated with tumor diameter (P < 0.01, chi(2)), lymphatic metastasis (P < 0.05) and hepatic metastasis (P < 0.05), but not with the differentiation of tumor cells. CONCLUSION: The combined use of negative and positive immunomagnetic beads followed by amplification of CK20 mRNA by means of RT-PCR is a non-invasive, sensitive, and specific assay for the detection of circulating colonic cancer cells.

Assessing the predictive accuracy of QUICKI as a surrogate index for insulin sensitivity using a calibration model.

The quantitative insulin-sensitivity check index (QUICKI) has an excellent linear correlation with the glucose clamp index of insulin sensitivity (SI(Clamp)) that is better than that of many other surrogate indexes. However, correlation between a surrogate and reference standard may improve as variability between subjects in a cohort increases (i.e., with an increased range of values). Correlation may be excellent even when prediction of reference values by the surrogate is poor. Thus, it is important to evaluate the ability of QUICKI to accurately predict insulin sensitivity as determined by the reference glucose clamp method. In the present study, we used a calibration model to compare the ability of QUICKI and other simple surrogates to predict SI(Clamp). Predictive accuracy was evaluated by both root mean squared error of prediction as well as a more robust leave-one-out cross-validation-type root mean squared error of prediction (CVPE). Based on data from 116 glucose clamps obtained from nonobese, obese, type 2 diabetic, and hypertensive subjects, we found that QUICKI and log (homeostasis model assessment [HOMA]) were both excellent at predicting SI(Clamp) (CVPE = 1.45 and 1.51, respectively) and significantly better than HOMA, 1/HOMA, and fasting insulin (CVPE = 3.17, P < 0.001; 1.67, P < 0.02; and 2.85, P < 0.001, respectively). QUICKI and log(HOMA) also had the narrowest distribution of residuals (measured SI(Clamp) - predicted SI(Clamp)). In a subset of subjects (n = 78) who also underwent a frequently sampled intravenous glucose tolerance test with minimal model analysis, QUICKI was significantly better than the minimal model index of insulin sensitivity (SI(MM)) at predicting SI(Clamp) (CVPE = 1.54 vs. 1.98, P = 0.001). We conclude that QUICKI and log(HOMA) are among the most accurate surrogate indexes for determining insulin sensitivity in humans.

[Changes in neuropeptide substance P and calcitonin gene-related peptide in scarless wound healing in fetal rabbit skin].

OBJECTIVE: To explore the role of the neuropeptide in fetal scarless wound healing. METHODS: The animal models were replicated in which a full-thickness wound was inflicted on the back of rabbit fetuses at 22 days gestation (term=31 days). The progress of wound healing was observed on micro-and macro-levels at the 1 st, 2 nd, 3 rd, 5 th, 7 th, 14 th post-injury days respectively. The expression level of substance P(SP) and calcitonin gene-related peptide (CGRP) during wound healing was assessed by immunohistochemistry staining. RESULTS: The cutaneous wounds in fetal rabbits healed more rapidly than that in adult and without the formation of scar. The expression of SP and CGRP of wound decreased in fetal rabbit at the early stage post-injury, down to the lowest levels on the 2 nd post-injury day (75%, 80%, vs. fetal control, both P<0.01), then increased gradually, up to the normal levels. The expression of SP and CGRP of wound decreased in adult rabbit at the early stage post-injury, down to the lowest levels on the post-injury 1 st day (60%, 76%, vs. adult control, both P<0.01), then increased quickly, up to a higher level than normal group, reaching the peak on the post-injury 7th day(168%, 126%, vs. adult control, both P<0.01), then returned to the normal levels. CONCLUSION: The low level of neuropeptide may contribute to scarless wound healing in fetal rabbit.

[Expression and biology identification of the human epididymis-specific gene ESC42 in E. coli].

OBJECTIVE: To provide materials for the study of the function of ESC42 protein specifically expressed in the human epididymis. METHODS: The ESC42 gene was amplified from the human epididymis cDNA library by PCR and then cloned into prokaryotic expression vector pGEX-4T-1, expressed and purified by recombinant DNA techniques. The specificity of ESC42 protein was identified by Western blot and MALDI-TOF-MS. The database was searched by Ms-Fit. RESULTS: The recombinant plasmid expressed a Mr 38 x 10(3) fusion protein in E. coli at a level of 30% of the total protein, and the purity was as high as 99%. The ESC42 protein was identified by ESC42 monoclonal antibody and its molecular weight was 11 978.12, tested by MALDI-TOF-MS. The peptide mass fingerprint analysis showed that the coverage rate of the sequence reached 48% with 100% matching. The motif scan in Prosite database reveal that ESC42 belonged to the beta-defensin family and had antibacterial activity. CONCLUSION: Obtaining high purity of rhESC42 protein may lay a foundation for the study of its functions.

[A practical method for the calculation of wound water loss in burn patients].

OBJECTIVE: To evaluate a calculating method of wound water loss (WWL) in hospitalized burn patients. METHODS: Eighty-five adult burn patients with total burn area over 50% were enrolled in the study. The WWL formula after shock stage (after 3rd postburn day) was given as: WWL (ml/%/kg/24 h) = [input (ml/24 h)-urine output (ml/24 h)-500 ml] / actual burn wound area(%) / body weight (kg). The result was compared with that determined by other formulas such as Davies, S.M.S.P and Lamke and palm rule. RESULTS: The calculated daily WWL in burn patients after shock stage was found out to be 0.9 +/- 0.1 ml/%/kg/24 h by this method. This figure is comparable with that calculated by several different methods. Palm rule could be simply applied in the patients with body weight around 60 kg. CONCLUSION: This method and palm rule were proved to be simple and practical for the calculation of WWL in burn patients, which could be applied in clinical work.

[High temperature Raman spectra and structure character study of BSO crystal].

The structure character of BSO crystal at room temperature was generalized. The main Raman shifts of lattice vibration at room temperature were interpreted. The Raman spectra of BSO crystal were measured in a temperature range from 293 K to 1123 K with high temperature Raman spectroscopy and time-resolved detection techniques. Temperature-dependence character of the Raman spectra of the crystal was investigated. The vibration mode of the longest bond Bi-O(1) in crystal shifts from 542 cm(-1) to 512 cm(-1) with the temperature increasing from room temperature to 1123 K. It can be attributed to the fact that oxygen atoms are electrostatic bond to Bi atoms. The intensity of 88 cm(-1) modes, which belongs to combination mode of bending and stretching in Bi3O4 unit, changes not so noticeably as other modes, and even the 58 cm(-1) mode of Bi atoms motions in crystal lattice decreases rapidly when the temperature is higher than 873 K, which indicates that the framework structure of the crystal is broken down at high temperature, while the Bi3O4 unit still exists.

[Analysis of spontaneous reperfusion in acute myocardial infarction and its effect on short-term prognosis].

OBJECTIVE: To analyse the clinical and angiographic characteristics of spontaneous reperfusion (SR) in AMI, and to evaluate its effect on short-term prognosis. METHODS: 112 consecutive AMI patients without intravenous thrombolytic therapy received emergent coronary angiography and primary PCI. The patients were divided into SR group (antegrade TIMI grade 2-3 flow) and non-SR group (antegrade TIMI grade 0-1 flow). The clinical, angiographic and prognostic features of the patients were analyzed. RESULTS: 31 patients (27.7%) were in SR group, and there was no significant difference in base-line clinical characteristics between the two groups. Compared with non-SR group, peak values of CK and CK-MB, Ventricular wall motion abnormality and mortality were lower in SR group, ejection fraction was higher in SR group. Logistic regression analysis showed that there was good correlation between SR and peak value of CK, collaterals, ventricular wall motion abnormality and pre-dilation in PCI. CONCLUSION: SR decreased infarction size, improved heart function and reduced 30-day mortality.

[Structure analysis on growth solid-liquid boundary layer of BSO crystal at real time].

The micro Raman spectra of solid-liquid boundary layer, the melts and crystal side, were measured at real time, concerning BSO crystal grown with zone-melting method. The structure characters in boundary layer, melts and crystal were analyzed. The process, of which the growth unit structure changed while they transited from melts through boundary layer to crystal lattice, was analyzed. The results show that, there exists Bi3O4 and [SiO4] bonding structure in the melts of BSO crystal. While in the solid-liquid boundary layer, the Bi3O4 molecular units converge into [BiO7] octahedron monomer of polymer in form, the monomer or the polymer converge with the [SiO4] structure units, then all these converged structure enter into crystal lattice sites.

[Effect of etomidate on hemodynamics in elderly and shock patients during general anesthesia induction].

OBJECTIVE: To investigate the effect of etomidate on the hemodynamics in elderly and shock patients during general anesthesia induction. METHODS: Totally 100 elderly patients or patients with hemorrhagic shock (ASA II -III ) undergoing surgery were studied. Anesthesia was induced with 4 microg/kg of fentanyl, 0.1 mg/kg of vecuronium and 0.2-0.3 mg/kg of etomidate, and the mean arterial blood pressure (MAP), heart rate (HR), stroke volume (SV), stroke index (SI), cardiac output (CO), cardiac index (CI), cardiac acceleration index (ACI), left cardiac work (LCW), and left cardiac work index (LCWI) were recorded using thoracic electrical bioimpedance (TEB) hemodynamic monitoring system before induction and at 1, 2 and 3 min after etomidate injection as well as at 3 min after intubation. RESULTS: After etomidate injection, MAP, HR, SV, SI, CO, CI, ACI, LCW and LCWI were decreased significantly as compared with those before induction (P<0.05). MAP, HR, CO, CI and ACI at 3 min after intubation were higher than those before induction (P<0.05), and the other indices resumed the baseline level. The amplitudes of such changes in the recorded indices was below 20% of the baseline level in the course of anesthesia induction. CONCLUSION: Cardiovascular hemodynamics may vary slightly during the course of induction with etomidate, which is an ideal drug for anesthesia induction for elderly or shock patients with unstable hemodynamics.

[Effects of three madriatics on the human ocular aberrations].

OBJECTIVE: To evaluate the effects of three common madriatics-phenylephrine, tropicamide and Mydrin-P on aberrations in human eye. METHODS: General aberrations, lower- and higher-order aberrations (HOA) were measured at the pupil diameter 4.0 mm and 7.0 mm with Allegretto Wave Analyzer on 37 eyes of 19 young volunteers whose spherical equivalence (SE) ranged from -0.25 D to -6.00 D. Four measured conditions were used for each subject. (1) Topical installation of 3 drops 5.0% phenylephrine hydrochloride. (2) Topical installation of 3 drops 0.5% tropicamide. (3) Topical installation of 3 drops Mydrin-P. (4) Topical installation of 3 drops 0.3% Tarivid, prior to aberometry measurements. The differences were analyzed on the values of the root-mean-square (RMS) of general (RMSg) and higher-order aberrations (RMSh), coefficient of defocus, astigmatism, the third aberrations, primary spherical aberration and the dominating type of HOA among three madriatics or between the madriatics and Tarivid solution respectively. RESULTS: There was no significant difference among the three madriatics in the average value of RMSg, RMSh, C3-C9 and C12 (P > 0.05). However, small but statistically significant difference was found in the disparity between any madriatic and Tarivid at the condition of 4.0 mm diameter of pupil. The disparity of 5% phenylephrine over RMSg and C4 was the smallest, and showed significant difference (P < 0.05). The third order aberration was more frequent dominating type of HOA over a 4.0 mm and 7.0 mm pupil for 4 eye drops, and C7 was the most frequent one among all the volunteers; there were 28 eyes (75.7%), 20 eyes (54.1%) which dominating type of HOA changed. CONCLUSIONS: All the three madriatics can be used to measure the higher-order aberrations, spherical aberration and coma in the young myopic persons. However, phenylephrine measurement may be the choice for lower-order aberrations.

[Uptake HTO from seawater by seashell and formative dynamics of bound tritium in seashell].

Uptake HTO from seawater by five kinds seashell and formative dynamics of bound tritium in organ of seashell are studied by using the isotope-tracer techniques in order to get a better understanding of the marine environmental behavior of HTO. The results show that the tritium in seashell exists in free water tritium and bound tritium form. The free water tritium is main form; its content observed was more than 97.4% of total tritium. The bound tritium content was very lower, only being 0.4% - 2.6% in total tritium. The speed of HTO untaken by seashell is very rapid, specific activity of HTO in three kinds seashell had reached their maximum only 2 h after treatment. The bound tritium in seashell increases slowly with time. The results of analysis of concentration factor (CF) values indicate that HTO would not concentrate in the seashell.

[Effect of sinomenine on adjuvant arthritis and its mechanisms].

OBJECTIVE: To discuss the anti-inflammatory mechanism of sinomenine on inflammatory media in joint of adjuvant arthritis rats. METHOD: Rats were randomly divided into the normal group and the model group, the prednisone group, the small, medium, large of sinomenine group (30, 60, 120 mg x kg(-1)). Except for the rats in the normal group, animals were modeled to adjuvant arthritiswith freund's complete adjuvant. The arthritis index (AI) and the swelling degree of paws were recorded, and the activity of IL-1, TNF and the levels of NO, PGE2 in joint fluids of secondary arthritis were determined. RESULT: Compared with the normal group, the activity of IL-1, TNF and the levels of NO, PGE2 in joint fluids of secondary arthritis were increased significantly in the model group (P < 0.05). Compared with the model group, it was shown to exert a dramatic inhibitory effect on secondary reaction of freund's adjuvant arthritis of rats, and the activity of IL-1, TNF and the levels of NO, PGE2 in joint fluids of secondary arthritis were significantly decreased in the sinomenine group (P < 0.05). CONCLUSION: Sinomenine has a remarkable treatment effect on RA. It is via NO to inhibit the activity of cytokines and decrease the level of inflammation mediators, which may be one of its curing RA mechanism.

[Improving thermostability of Aspergillus niger phytase by elongation mutation].

The phytase gene phyA(m) from Aspergillus niger N25 was recombined into E. coli expression vector pET-30b(+). Recombined at expression vectors pET30b-FphyA(m) was served as a template to amplify phytase gene, and the PCR product named elongation mutation gene phyA(e) was expanded with a 13 amino acid sequence from pET-30b-FphyA(m) vector at C-terminal of phytase gene phyA(m). Furthermore, phyA(e) gene was recombined into expression vector pPIC9k and expressed in Pichia pastoris. The comparison experiment of mutant phytase PP-NP0 with wild-type phytase PP-NP(m)-8 showed that: the optimum temperature of PP-NPe was increased by 3 degrees C, and its thermostability was increased by 21% when it was exposed to 10 min at 75 degrees C. Its effective reaction pH range with catalysis efficiency above 70% was pH 4.6 - pH 6.6, and wider 0.4 pH value than that of wild-type phytase.
