Simulation study of ratio calculation formulae of two-colour cDNA microarray data.

In cDNA microarray image processing, there are different methods for calculating the channel ratios. Standard microarray image analysis software, such as the Axon GenePix Pro, calculate the channel ratio from pixels that define a given spot using different methods (i.e. ratio of means, ratio of medians, mean of ratios, median of ratios, and regression ratio). Ratio values calculated using the different methods will then be listed in an output file. Microarray users have to choose one of the available methods at their own discretion, as no guidelines are provided. Therefore, we aim to address one of the most frequently asked questions by the microarray users: which ratio quantity provided by the image analysis software should be used? In this study, we have evaluated the five different ratio calculation approaches using simulation studies. Our results suggest that in most circumstances the ratio of means appears to be the best approach, particularly when the coefficient of variance (CV) of two-channel pixel intensities are small (<0.5) and channel intensities are large. Conversely, the ratio of medians and the median of ratios are more favourable when the CV is large.

[Pharmacokinetics of honokiol in rats].

OBJECTIVE: To investigate the pharmacokinetics of honokiol in rats. METHODS: Honokiol injection was delivered by vein injection to SD-rats. The blood samples were gathered at a series of time lags. Honokiol in rat plasma was determined with an RP-HPLC method and the data were analyzed with program 3P87. RESULTS: After i.v. injection of honokiol, concentration-time curves were fitted to a 3-compartment model: with halftime of 2.8 min, 11.9 min, and 56.8 min. CONCLUSION: Honokiol was quickly distributed in rats after i.v. and the concentration decreased rapidly. Our studies provided important referrence to the research on the pharmacodynamics and the pharmaceutics of Honokiol.

[Application of BaSO4 diffuser plate in 250-400 nm spectral radiance calibration].

Sprayed BaSO4 diffuser plate is the most Lambertian surface actually used in spectral radiance calibration known by now. Its hemispheric reflectance and Bi-directional Reflectance Distribution Functions (BRDF) were measured in the experiment. Its diffuse characteristics were compared with Lambertian surface. In order to calibrate spectral radiance more accurately, the small variation of diffuser's BRDF with scattered angles and the nonuniformity of spectral irradiance on diffuser surface illuminated by the standard lamp should be considered. By integrating the radiation flux reflected by the element area and that entering the entrance slit within the viewing area of spectrometer, the measured spectral radiance can be calculated. Furthermore, the spectral radiance of Lambertian surface whose BRDF was derived from hemispheric reflectance was compared with that from the average of the measured BRDF.
