[Study on the expression and genomic imprinting status of insulin-like growth factor 2 gene in hepatocellular carcinoma].

OBJECTIVE: To study on the expression and genomic imprinting status of IGF2 gene in hepatocellular carcinoma. METHODS: RT-PCR semi-quantitation analysis was used to test IGF2 in paired hepatocellular carcinoma and adjacent normal tissue. Restriction fragment length polymorphism was used to detect the imprinting status of IGF2. The relationships between the expression level of IGF2, its imprinting status, and clinical information were analyzed. RESULTS: The expression pattern of IGF2 was quite variable among the various HCC cases; the expression of IGF2 mRNA on cancer tissue was significantly higher than that on paired normal tissues (t=3.695); LOI was found in 11 HCC cases. CONCLUSIONS: It is supposed that the enhanced expression of IGF2 gene was involved in the carcinogenesis of HCC; LOI of IGF2 may be a prophase manifestation of hepatocellular carcinoma

[Effect of niupo zhibao pellet on transforming growth factor-beta1 and its receptor's expression in endotoxic shock rats with lung injury].

OBJECTIVE: To observe the influence of Niupo Zhibao pellet (NZP) on transforming growth factor-beta1 (TGF-beta1) and its receptor's expression. METHODS: Endotoxic shock model was established by intravenous injection of lipopolysaccharide (LPS) 1.5 mg/kg and intraperitoneal injection of D-galactosamine 100 mg/kg, and intervened by NZP, TGF-beta1 and its receptor's expression in lung tissue were detected by immunohistochemical method. RESULTS: NZP could enhance the TGF-beta1 and its receptor's expression in endotoxic shock lung tissue, and reduce the injury of lung. CONCLUSION: The mechanism of NZP in reducing endotoxic shock lung injury is possibly related with its effect in enhancing the TGF-beta1 and its receptor's expression in lung tissue.

[Partial middle turbinatectomy and folded for nasonsociliary neuralgia by transnasal endoscopic surgery].

OBJECTIVE: To explore into the feasibility and outcome of partial middle turbinetectomy and folded for nasociliary neuralgia by transnasal endoscopic surgery. METHOD: Forty patients with nasociliary neuralgia were treated by partial middle turbinatectomy and folded. RESULT: All patients have got sufficient of factory sulcus without hyposmia. The curable rate and the improve rate were 97.5% and 2.5%, respectively, with the total effective rate 100%. CONCLUSION: This procedure present an ideal effect. It is a safe, minimally invasive and efficient procedure for nasociliary neuralgia.

[Value of measurements of blood flow velocity in central retinal artery in thyroid-associated ophthalmopathy].

OBJECTIVE: To study the clinical values of measurement of blood flow velocity in central retinal artery in thyroid-associated ophthalmopathy (TAO). METHODS: Thirty normal subjects without any thyroid disorders or any eye abnormalities were assigned into the control group, and 30 TAO patients were assigned into the TAO group. Color Doppler ultrasonography was done to measure the blood flow velocity in central retinal artery. Measurement results were compared between these two groups. RESULTS: The peak systolic velocity and end diastolic velocity of blood flow were significantly lower in TAO group (P < 0.001), while the resistance index significantly increased (P < 0.001). CONCLUSION: Measurement of blood flow velocity in central retinal artery with Color Doppler ultrasonography is an easy and useful way to appraise the hemodynamics of the central retinal artery in TAO patients.

[Molecular cloning for an alternatively splicing cDNA of human PKNOX1 gene and it's expression analysis].

Down's syndrome (DS) is the most common chromosomal abnormality in live born infants. The cloning and characterization of genes on HC21 are necessary steps for the understanding of the molecular basis of Down's syndrome. To search for new Down's syndrome related genes, we have used bioinformatics analysis and rapid amplification of cDNA ends to identify an alternatively splicing cDNA of human PKNOX1 in Down's syndrome critical region on the long arm of human chromosome 21. The alternatively splicing full length cDNA of human PKNOX1, named PKNOX1B, is 2 793 bp in length and encodes a 405 amino acid protein. Bioinformatical studies show PKNOX1B gene spans over 58 kb on chromosome 21q22.3 and contains 11 exons and 10 introns. The predicted molecule weight of PKNOX1B is 44.628 kDa, and the deduced iso-electric point is 6.28. Compared with PKNOX1 gene, PKNOX1B is 30 aa shorter at the C terminus than PKNOX1 owing to alternative splice between (10th exon and 11th exon) last two exons. PKNOX1B posses a same homeobox-domain as PKNOX1, so the isoform of PKNOX1 may be involved in the genetic control of development like other members of homeobox-containing gene family. RT-PCR results showed that PKNOX1B is expressed in all examined tissues except in marrow tissue. By Northern blot, PKNOX1 has three transcripts in adult testis, one about 5 kb, second about 2.9 kb, and third about 2 kb. The two larger transcript is expressed in all examined tissues, but the smallest transcript is only expressed actively in adult testis, so that it may play potential roles in spermatogenesis.

[Saline tolerance white clover transformed with the betaine aldehyde dehyrogenase gene by Agrobacterium tumefaciens].

The white clover has been transformed with the Betaine Aldehyde Dehydrogenase (BADH) gene cloned from Atriplex hortensis by Agrobacterium tumefaciens. The relative electronic conductivity of the transgenic plants under 1% NaCl stress for 48 hours was about 20%, less than the control plant's relative electronic conductivity (more than 40%), these showed the cell membrane of the transgenic plants has been less injured than control plants under salt stress. The other experience showed that the transgenic plant could grow well in water culture included 0.5% NaCl for more than two weeks, but the control plants could not.

[Expression and correlation of apoptosis-related gene c-IAP2 and caspase-4 in sinonasal squamous carcinoma].

OBJECTIVE: To examine the expression of apoptosis-related genes c-IAP2 and caspase-4 in sinonasal squamous carcinoma. To investigate the role and mechanism of the two apoptosis-related genes in nasal cavity and sinus squamons carcinoma. METHODS: Forty-one pathologically confirmed specimens of nasal cavity and sinus squamons carcinoma were subjected for immunohistochemical staining to analyze the expression of c-IAP2 and caspase-4, in addition 10 normal nasal mucosa were used as control. RESULTS: The positive rate of c-IAP2 in sinonasal squamons carcinoma was 87.8%, among which 14 cases were strongly stained(+ + +), 15 cases were stained(+ +), 7 cases were stained (+), only 5 cases were negative. In 10 normal control tissues, no case was stained(+ + +), 2 cases were stained (+ +), 3 cases were stained(+), the other 5 cases were negative. Higher level expression of c-IAP2 protein was detected more often in squamons carcinoma specimens than that in normal nasal mucosa. There was statistically significant difference between the two groups for the staining(P < 0.05). The expression of c-IAP2 was not correlated with clinical staging (P > 0.05), but there were positive correlation with clinical prognosis (P < 0.01) and pathological classification (P < 0.01); The positive rate of caspase-4 in nasal cavity and sinus squamons carcinoma was 58.5%, among which 4 cases were strongly stained (+ + +), 7 cases were stained (+ +), 13 case was stained(+), 17 cases were negative. The expression of caspase-4 protein was more deletion in squamons carcinoma specimens than that in control group, there was statistically significant difference between the two groups for the staining (P < 0.05). The expression of caspase-4 protein was negatively correlated with clinical prognosis (P < 0.01) and pathological classification ( P < 0.01), not correlated with clinical staging(P > 0.05). The expression of c-IAP2 was negativelly correlated with the experssion of caspase-4. CONCLUSIONS: The results indicates that in sinonasal squamous carcinoma, c-IAP2 is activated, while caspase-4 is restrained, the signal transduction pathway of apoptosis is arrested, leading to surviving of tumor cells, this maybe directly associated with the development of sinonasal squamons carcinoma, and with the failure of tumor trentment.

[Characterization of tissue expression of peroxisome proliferator activated receptors in the chicken].

The peroxisome proliferator activated receptor (PPAR) is the member of super family of nuclear receptors. A great number of researches in rodent and human have showed that PPARs are involved in the lipids metabolism, especially in regulating the microsome,expression of genes coding the various enzymes during the beta-oxidation of peroxisome and the differentiation of adipose cell. The PPARs are one of the important gene clusters that are viewed as the objective genes correlated with the lipids metabolism and corresponding diseases. Great differences in lipids metabolism are existed between the poultry and mammals. The mechanism of the lipids metabolism in poultry is not completely understood. The current study was designed to investigate the expression characterization of PPAR genes in chicken in order to understand the effects of PPAR gene on adipose and other tissues. The eight weeks age Arber Acres broilers were used and quantitative RT-PCR and Northern blot were developed to analyze the expression characterization of PPAR-alpha and PPAR-gamma gene in heart, liver, spleen, lung, kidney, stomach, intestine, brain, muscle and adipose tissues. Results of the quantitative RT-PCR showed that the expression level of PPAR-alpha gene was higher in brain, lung, kidney, heart and intestine, medium in stomach, liver and adipose, and lower in spleen. But did no expression was detected in breast muscle. The expression level of PPAR-gamma gene was higher in adipose, medium in brain and kidney, lower in spleen, heart, lung, stomach and intestine, but no expression was detected in liver and breast muscle. Northern blot results showed that PPAR-alpha gene expressed in heart, liver, kidney and stomach, and the intensity of hybridization signal was the greatest in liver tissue; PPAR-gamma gene only expressed in adipose and kidney tissues, and the intensity of hybridization signal was greater in adipose tissue. The present study results showed that the tissue expression characterization of PPARs gene in chicken was almost consistent with that in rodent and human, but, in chicken, PPAR-alpha did not expressed in breast muscle and PPAR-gamma gene expressed in kidney. The results of current research indicated simultaneously that the PPAR genes were related with the growth and development of various tissues and organs.

[Three-dimensional ultrasound evaluation of the efficacy and safety of single-dose systemic or local methotrexate injection for tubal pregnancy].

OBJECTIVE: To compare the efficacy and safety of single-dose systemic and local injection of methotrexate (MTX) in the treatment of tubal pregnancy (TP). METHODS: The systemic group included 127 women with TP who received a single-dose MTX of 50 mg/m(2) given by intramuscular injection, and the local group included 118 women receiving single-dose MTX of 100 mg via local injection guided by three-dimensional (3D) ultrasound. The volume of ectopic masses and blood beta-human chorionic gonadotropin (beta-HCG) were measured before and after MTX administration. The TP was detected by 3D images reconstructed 3 months after MTX administration. RESULTS: The success rate of treatment was 100% in the local group, significantly higher than that of 91.34% in the systemic group. After the first week of MTX injection, the speed of the ectopic mass resolution was 0.85+/-0.04 cm(3)/d and 0.47+/-0.03 cm(3)/d in systemic group and local group respectively (P<0.05), and the time of the ectopic mass resolution was 27.66+/-12.77 d and 32.62+/-12.75 d, respectively (P<0.01). The rate of tubal deformity in the local group was significantly lower than that in the systemic group. CONCLUSION: Local injection of MTX under ultrasound guidance is safe, efficient and reliable for conservative management of unruptured TP, which is superior to systemic injection.

Responses of chlorophyll fluorescence and nitrogen level of Leymus chinensis seedling to changes of soil moisture and temperature.

Controlled experiment of Leymus chinensis seedlings grown in the environmental growth chambers at 3 soil moisture levels and 3 temperature levels was conducted in order to improve the understanding how leaf photosynthetic parameters will respond to climatic change. The results indicated that soil drought and high temperature decreased the photochemical efficiency of photosystem (Fv/Fm), the overall photochemical quantum yield of PSII(yield), the coefficient of photochemical fluorescence quenching(qP), but increased the coefficient of non-photochemical fluorescence quenching(qN). Severe soil drought would decrease Fv/Fm and yield by 3.12% and 37.04% under 26 degrees C condition, respectively, and 6.60% and 73.33% under 32 degrees C condition, respectively, suggesting that higher temperature may enhance the negative effects of soil drought. All the soil drought treatments resulted in the decline in leaf nitrogen content. There was no significant effect of temperature on leaf nitrogen level, but higher temperature significantly reduced the root nitrogen content and the ratio of root nitrogen to leaf nitrogen, indicating the different strategies of adaptation to soil drought and temperature. It was also implied that higher temperature would enhance the effect of soil drought on leaf photosynthetic capacity, decrease the adaptability of Leymus chinensis to drought.

[Genetic structure of Tujia as revealed by Y chromosomes].

Tujia people call themselves "Bizika", which means aboriginal. Genetic study of Tujia is virtually absent. To characterize the genetic structure of Tujia,the distribution of 14 Y haplogroups was studied in Tujia populations sampled from Enshi, Hubei (31 males) and Jishou, Hunan (68 males). A total of eight haplogroups were observed in the Enshi and Jishou populations. The haplogroup frequencies of Tujia were compared with the frequencies of other related ethnic groups, including Northern Han, Southern Han, Tibetan-Burman speaking populations,Daic and Hmong-Mien. The principal component (PC) analysis was conducted and the PCs were plotted to explore the historical migrations. In addition, partial correlation analysis was performed to study the relationship between the first three PCs and the haplogroups. The PC2 revealed a cluster of Tujia groups including Longshan,Yongshun, and Enshi with Lahu, suggesting possible interaction between Tujia and the Di-Qiang groups. However,a similarity between Han and Tujia populations, though differentiated, were also observed. We postulated, by incorporating the results of archaeological and historical evidences, that the Ba people, the ancestors of the Tujia,might be related with Di-Qiang groups and inhabited the Tujia area initially before a substantial interaction with Han and other ethnic groups.

Establishment of liver specific glucokinase gene knockout mice: a new animal model for screening anti-diabetic drugs.

AIM: To characterize the liver-specific role of glucokinase in maintaining glucose homeostasis and to create an animal model for diabetes. METHODS: We performed hepatocyte-specific gene knockout of glucokinase in mice using Cre-loxP gene targeting strategy. First, two directly repeated loxP sequences were inserted to flank the exon 9 and exon 10 of glucokinase in genomic DNA. To achieve this, linearized targeting vector was electroporated into ES cells. Then G418- and Gancyclovir-double-resistant clones were picked and screened by PCR analysis and the positives identified by PCR were confirmed by Southern blot. A targeted clone was selected for microinjection into C57BL/6J blastocysts and implanted into pseudopregnant FVB recipient. Chimeric mice and their offspring were analyzed by Southern blot. Then by intercrossing the Alb-Cre transgenic mice with mice containing a conditional gk allele, we obtained mice with liver-specific glucokinase gene knockout. RESULTS: Among 161 double resistant clones 4 were positive to PCR and Southern blot and only one was used for further experiments. Eventually we generated the liver specific glucokinase knockout mice. These mice showed increased glucose level with age and at the age of 6 weeks fasting blood glucose level was significantly higher than control and they also displayed impaired glucose tolerance. CONCLUSION: Our studies indicate that hepatic glucokinase plays an important role in glucose homeostasis and its deficiencies contribute to the development of diabetes. The liver glucokinase knockout mouse is an ideal animal model for MODY2, and it also can be applied for screening anti-diabetic drugs.

[Experimental study on the antiviral mechanism of Ceratostigma willmattianum against herpes simplex virus type 1 in vitro].

OBJECTIVE: To study the antiviral effect and mechanisms of the liquid extract from Ceratostigma willmattianum against herpes simplex virus type 1 (HSV-1) in vitro. METHOD: C. willmattianum in various concentration was applied to different steps of HSV-1 replication cycle. 50% Tissue culture infective dose (TCID50), cytopathic effect (CPE), MTT staining method, dot blotting and Northern blotting analysis were used to estimate index of antiviral activity. RESULT: 50% Toxic concentration (TC50) was 1077 mg x L(-1), IC50 29.46 mg x L(-1) and therapeutic index (TI) 36.56 in C. willmattianum. TC50 330 mg x L(-1), 50% Inhibiting concentration (IC50) 9.12 mg x L(-1) and TI 36.18 in ACV by MTT staining method. The liquid extract from C. willmattianum had remarkable effect on inhibiting HSV-1 in vitro. Ceratostigma could interfere absorption of HSV-1 to Vero cells to prevent HSV-1 infectivity, inhibit HSV-1 gD DNA replication and HSV-1 gD mRNA expression. CONCLUSION: C. willmattianum possesses strong anti-HSV-1 activity in vitro. The antiviral mechanisms are related to inhibiting virus absorption, HSV-1 gD gene replication and HSV-1 gD gene transcription.

[Experimental study on hemostatic effect of flos sophorae and its extracts].

OBJECTIVE: To compare the hemostatic effect of Flos Sophorae in crude, parched and carbonized forms and its extracts, including rutin, quercetin and tannin. METHODS: All the testing samples were orally administered to the experimental animals for 5 days, then the bleeding time (BT), coagulation time (CT), platelet count and capillary permeability (CP) in the treated mice were tested, and the prothrombin time (PT), fibrinogen (FBG) content and platelet aggregation rate (PAR) in the treated rats were determined. RESULTS: All the samples could lower the CP, BT and CT in mice and also decrease the plasma PT in rats. All three forms of Flos Sophorae could increase FBG in rats, while the three extracts of it could inhibit the PAR in rats obviously. In addition, rutin had the effect of raising the platelet count. CONCLUSION: All the three forms and three extracts of Flos Sophorae have hemostatic effect, the effect of parched and carbonized form is higher than that of crude drug. The mechanism of the hemostatic effect of the six kinds of sample might be various.

[Cloning and sequencing of the introns of UCP gene and construction of molecular phylogenetic tree in chicken].

The UCP genes were the newly discovered genes that can increase the energy expenditure and involve in the metabolism of fat and regulation of energy. Four pairs of primers in chicken UCP exon region were designed to amplify the introns of chicken UCP gene according to the splice ways of the mouse UCP2 gene (Accession No.AF096288). The sequence results showed that the chicken UCP gene also had five GT-AG type introns. The molecular phylogenetic tree was constructed based on the sequence of cds, intron 2 and intron 3 region, respectively. The phylogenetic tree based on the UCP cds region was consistent with the species phylogenetic tree. This result implicated that UCP gene can be regarded as the useful gene for the study of animal phylogenesis. On the contrast, the phylogenetic tree based on the intron 2 and intron 3 region was different from the species phylogenetic tree, which showed that the evolution of intron and cds region is different.
