[Specific cell immune response mediated by dendritic cells in multiple myeloma patients in vitro].

OBJECTIVE: To explore KM3 multiple myeloma (MM) cell line specific cytotoxic T lymphocyte (CTL) response in vitro mediated by autologous dendritic cells (DCs) aroused by KM3 cells' lysates and acid-eluted peptides. METHODS: Monocytes were isolated from the peripheral blood of healthy individuals and MM patients, and were cultured in serum medium with IL-4, GM-CSF, and TNF-alpha to generate DCs. These DCs were pulsed by KM3 cells' lysates or the acid-eluted peptides, then incubated with autologous T lymphocytes for 3 days to induce KM3 cell antigen specific CTL. MTT assay was performed to examine the specific KM3 cells' lysing ability of CTL. RESULTS: DCs were generated in peripheral blood monocytes cultured in the serum medium containing GM-CSF, IL-4, and TNF-alpha. Autologous T lymphocytes induced by IL-2 and DCs pulsed with KM3 cells' lysates or the acid-eluted peptides showed strong killing effect on KM3 cells. CONCLUSION: The DCs pulsed by KM3 cells' lysates or the acid-eluted peptides incubated with autologous T lymphocytes can induce KM3 cell antigen specific CTL.

[Species of Ligularia in the northwestern China and their medicinal uses].

OBJECTIVE: To investigate the species of Ligularia distributed in the northwestern China and their medicinal uses in the local area. METHOD: Field investigation, specimen collection, taxonomic study and datum check were adopted. RESULT: There are 29 species and 1 varieties of Ligularia distributed in the northwestern China, and 18 species of them had been used as folk medicines with the function of resolving phlegm, relieving cough, clearing heat and toxins. CONCLUSION: The northwestern China is abundant in medicinal resource of Ligularia.

[Preliminary studies on in vitro and in vivo thrombolytic activities of thrombolytic enzyme from an induced Bacillus subtilis strain].

OBJECTIVE: To separate and purify a novel thrombolytic enzyme (FSC2-13) from culture media of an induced Bacillus subtilis strain and to study its in vitro and in vivo thrombolytic activities. METHODS: Employed fibrin plate method, pyrogenation (heating up) fibrin plate method and SDS-PAGE were used respectively to study FSC2-13 hydrolyzing fibrin and fibrinogen in vitro. Investigations were made on its in vivo pharmacodynamics using rat thrombogenesis inhibition model after it was administrated by intestinal route. RESULTS: FSC2-13 could hydrolyze fibrin and fibrinogen in vitro and had hydrolysis sites on three peptide chains of fibrinogen. FSC2-13 could significantly inhibit the formation of rat venous thrombus after being administrated by intestinal route. Thrombogenesis inhibition rate was 68%. Activated partial thromboplastin time increased by 51%, fibrinogen content decreased by 49%, fibrin degradation product (FDP) content also increased obviously, but the plasminogen content did not change obviously. CONCLUSION: FSC2-13 possessed in vitro and in vivo thrombolytic activities in a special way. There seem to be prospects for researches to develop it into a new generation of oral thrombolytic drug.

[Clinical significance of serum vascular endothelial growth factor and interleukin-6 in multiple myeloma].

OBJECTIVE: To investigate the clinical significance of the changes of vascular endothelial growth factor ( VEGF ) and interleukin-6 ( IL-6 ) level in multiple myeloma (MM), solid tumor following bone metastasis. METHODS: Thirty- seven MM patients, including 7 in Stage I , 8 in Stage II , 22 in Stage III, 8 solid tumor with bone metastasis patientsly, and 17 healthy controls were enrolled in this study. The serum VEGF and IL-6 levels were determined by ELISA. RESULTS: Serum VEGF and IL-6 concentrations in patients with MM and solid tumor were significantly higher than those of the healthy controls (P <0.01 ), and the VEGF level was higher in MM than in solid tumor with bone metastasis. There was significant difference in VEGF and IL-6 levels in various clinical stages of MM. VEGF levels in Stage II were significantly higher than in Stage I (P < 0.05 ) and IL-6 levels in Stage II were significantly higher than in Stage I (P < 0.05). The levels of IL-6 showed great difference according to bone lesion scores (P < 0.05). There was a positive correlation between IL-6 and serum calcium or C-reactive protein( P <0.01) , and there was a positive correlation between VEGF and serum Cr or urinary Bene-Jones protein lambda (P < 0.01 ). The IL-6 levels had significant differences between patients with the normal serum CRP, serum calcium, and beta2-MG and patients with abnormal ones (P < 0.05). VEGF levels showed significant differences between the patients with normal serum Cr, serum calcium Bene-Jones protein lambda, and urinary Bene-Jones protein lambda and patients with abnormal ones (P < 0.05). CONCLUSION: Serum VEGF and IL-6 levels are helpful to diagnose the clinical stages, and understand bone lesion and serevity of MM.

[Study on the surface markers on peripheral blood dendritic cells and their secretion of IL-12 and IFN-alpha in SLE patients].

AIM: To analyze the surface markers on peripheral blood dendritic cells (DCs) in SLE patients and explore the relationship between the DCs and pathogenesis of SLE. METHODS: The peripheral blood monouclear cells (PBMCs) were separated by density gradient centrifugation. After culture of 3 hours in tissue culture flask, the suspended cells were removed and GM-CSF, IL-4 and TNF-alpha were used to stimulate the proliferation and maturation of the peripheral blood DCs from normal persons and SLE patients. The surface markers on the DCs were analyzed by flow cytometry and the levels of IL-12 and IFN-alpha in supernatants were measured by ELISA after culture of 9 days. RESULTS: The positive percentages of CD1a, CD11c, CD40, CD83 and CD123 expressed on DCs of SLE patients were (58.88+/-7.64)%, (54.4+/-10.88)%, (37.29+/-8.08)%, (57.76+/-11.54)% and (13.14+/-4.44)%, respectively, whereas those of normal subjects were (47.71+/-4.01)%, (43.12+/-8.82)%, (28.59+/-7.07)%, (48.31+/-8.79)% and (9.85+/-3.97)%, respectively, (P<0.05). But the positive proportion of CD80 expression was (55.16+/-10.12)% in SLE group and (47.95+/-12.21)% in the control group, without significant difference (P>0.05). The level of IL-12 in SLE group was (9.78+/-0.76) ng/L, higher than that in normal group. The level of IFN-alpha in SLE group (2.95+/-0.61) ng/L was not significant difference from that in control group (2.70+/-0.29) ng/L (P>0.05). And there was no significant difference in IL-12 and IFN-alpha levels between non-active and active stages of SLE patients. CONCLUSION: The DCs may be involved in the pathogenetic process of SLE possibly by means of enhancement of antigen presenting function of DCs and secretion of IL-12.

[Analysis on the geologic background and physicochemical properties of cultivated soil of Flos Lonicerae in the geo-authentic and non-authentic producing areas].

OBJECTIVE: To study the relationship between the Geo-authentic and Geologic Background System (GBS) and physicochemical properties of cultivated soil in the Flos Lonicerae. METHOD: The analysis on the physicochemical properties of cultivated soil, the geologic background system in 5 producing areas of Flos Lonicerae was compared. RESULT: The distribution of genuine Flos Lonicerae was restricted by GBS, which was mainly restricted in the temperate continental monsoon semi-arid climate zone between north latitude 34-36 degrees and east longitude 113-118 degrees. CONCLUSION: The most suitable soil for the genuine Flos Lonicerae is the neutral or weak alkali sandy loamy soil with higher ion exchanging capacity, and the soils from those producing areas are determined by soil-forming parent materials.

[Technology optimization and properties study of factor IV complex concentrates].

OBJECTIVE: To produce high quality factor IX complex concentrates with domestic ion exchanger. A human solvent-detergent (S/D) treated factor IX complex concentrates has been produced from fresh-frozen plasma using domestic DEAE-Sepharose fast flow ion exchange chromatagraphy and Ca3(PO4)2 adsorption. The properties of this new concentrates were compared with those of the concentrates obtained by foreign DEAE-Sapharose CL-6B. RESULTS: The recovery of activity was 93.67 +/- 2.56% (n = 6) and 88.71 +/- 2 (n = 6), respectively. The specific activity of the concentrates was 7.36 +/- 0.96 U Factor IX: C/mg protein (n = 6). This new concentrates retained all the useful components with higher purity and lower thrombogenicity. Conclusion Domestic ion exchanger could replace foreign ion exchanger in producing high quality factor IX complex concentrates.
