[Screening of botulinum neurotoxin type A-binding peptides by phage display peptide library].

To seek effective inhibitor against BoNT/A, in this study, BoNT/A-binding peptides were screened from phage display peptide library using synthesized and identified mimicry peptides that contained antigenic epitopes as targets. According to the homology of the amino acid sequences of displayed peptides, most had same motifs respectively. ELISA assay confirmed that identified positive clones respectively against P4 and P5 could specifically bind BoNT/A. The mice assay showed the specific BoNT/A-binding peptides could partially protect against challenge of BoNT/A. The results from the study may aid in the future identification of more potent small molecule inhibitors against BoNT/A.

[With genechip technique to investigate HLA-DQB1 gene polymorphisms in south and north regions of China].

OBJECTIVE: To research and compare HLA-DQB1 gene frequency(GF) and polymorphism distribution between south and north population of Chinese in China. METHODS: Combining PCR-sequence specific primers(SSP) and sequence specific oligonucleotide probe(SSOP) techniques and DNA Microarray Kit for HLA-DQB1 Low Res Genotyping from Shenzhen Yi-Shengtang Biological LTD. Co. was used to type HLA-DQB1 gene polymorphisms of 700 individuals living in south China and 320 individuals in north China. RESULTS: We inspected 10 alleles of HLA-DQB1 and got a series of comprehensive and accurate statistic data. CONCLUSION: It is tested that HLA-DQB1*02, 05, 0601, 0602, 0603 gene frequencies are different obviously(P<0.05) between south and north Chinese. And those data will be useful to kinds of research associated with disease relevant and anthropology research.

[Comparison of the long term results between two conditioning regimens MCC and BuCy in chronic myelocytic leukemia after allogeneic stem cell transplantation].

OBJECTIVE: To observe and evaluate the long term survival of patients with chronic myelocytic leukemia transplanted with MCC and BuCy conditioning regimens. METHODS: Fourteen cases were treated with MCC regimen (Melphanlan 170 mg/m2 x d x 1, MeCCNU 400 mg/m2 x d x 1, CTX 60 mg/kg x d x 2) and the median follow up time was 6 years; 16 cases were treated with BuCy regimen (Busulfan 4 mg/kg x d x 4, CTX 60 mg/kg x d x 2) and the median follow up time was 4 year. RESULTS: All the patients were engrafted successfully. 4 of 10 patients examined in MCC group showed mixed chimerism at day 100 after transplantation, whereas only 1 of 12 patients examined in BuCy group showed mixed chimerism. All the patients became complete donor source later without any DLI. The 5-year disease-free survival rate was 71.4% for MCC group and 62.5% for BuCy group. The transplant related mortality and relapse rate were 21% and 7% for MCC group, whereas those were 25% and 12% for BuCy group, respectively. The regimen related toxicity was relatively lower in MCC group and the median duration of hospitalization was 39 days (25-55 days) for patients with MCC regimen, and 55 days (39-90 days) for BuCy regimen. CONCLUSION: MCC regimen has a partial ablative effect on CML and the long term disease-free survival is the same as that of BuCy regimen. In regard to the cost-effect efficacy, MCC regimen has a substantial advantage over BuCy regimen.

[A multicentre study on the pathogenic agents in 665 adult patients with community-acquired pneumonia in cities of China].

OBJECTIVE: To investigate the pathogenic causes of community-acquired pneumonia (CAP) in adult patients in China, the relation of previous antibiotic use and the Pneumonia Patient Outcome Research Team (PORT) classification to microbial etiology, and the prevalence of drug resistance of common CAP bacteria. METHODS: A prospective study was performed on 665 consecutive adult patients with CAP at 12 centers in 7 Chinese cities during one year. The etiology of pneumonia was considered if one of the following criteria was met: (1) valid sputum sample yielding one or more predominant strains; (2) blood cultures yielding a bacterial pathogen; (3) seroconversion, a > or = 4-fold increase or decrease titers of antibodies to Mycoplasma pneumoniae, Chlamydia pneumoniae and Legionella pneumophila. Minimum inhibitory concentration (MIC) of respiratory tract isolates was determined using the agar dilution method. RESULTS: Pathogens were identified in 324/610 patients (53.1%) with valid serum samples and sputum cultures as follows: Mycoplasma pneumoniae (126, 20.7%), Streptococcus pneumoniae (63, 10.3%), Haemophilus influenzae (56, 9.2%), Chlamydia pneumoniae (40, 6.6%), Klebsiella pneumoniae (37, 6.1%), Legionella pneumophila (31, 5.1%), Staphylococcus aureus (23, 3.8%), Escherichia coli (10, 1.6%), Moraxella catarrhalis (8, 1.3%), Pseudomonas aeruginosa (6, 1.0%). Of 195 patients with a bacterial pathogen, an atypical pathogen was identified in 62 (10.2%) cases. The non-susceptibility rate of Streptococcus pneumoniae to penicillin, azithromycin, and moxifloxacin was 20.3%, 75.4% and 4.3% respectively. CONCLUSIONS: Atypical pathogens have important role in CAP, with Mycoplasma pneumoniae being the most common pathogen, and mixed infection of atypical pathogens with bacteria was found in 10.2% of the cases. Streptococcus pneumoniae and Haemophilus influenzae remain the most important bacteria for CAP. More than 75.0% of Streptococcus pneumoniae was resistant to macrolides and 20.3% was resistant to penicillin.

[Secretory expression of sCD14 in Pichia pastoris and its LPS binding activity].

AIM: To achieve secretory expression of mature sCD14 gene in Pichia pastoris and to analyze its LPS binding activity. METHODS: sCD14 gene was cloned into yeast expression vector pPIC9K. Recombinant plasmid was lineared by Sac I and transformed into Pichia pastoris GS115. Positive integrated clones were screened and expressed by methanol inducing. RESULTS: The mature sCD14 was expressed in BMMY with pH 6.5 after optimization of inducing expression condition. Western blot and ELISA data revealed recombinant products combined with specific antibody. The results from FACS showed recombinant product had biological activity of binding with LPS. CONCLUSION: The recombinant mature sCD14 molecules with favorable LPS-binding activity could be used for further study on its structure and functions.

[Treatment efficacy of surgical management for liver metastasis from colorectal cancer--a report of 198 cases].

BACKGROUND & OBJECTIVE: Liver is the most common site of metastasis in colorectal cancer, and 35% patients with colorectal cancer developed liver metastasis at diagnosis. The prognosis of the patients with liver metastases from colorectal cancer is poor. Surgical resection, radiofrequency ablation, and chemotherapy had been used in clinical treatment for liver metastasis from colorectal cancer with various outcomes. This study was to explore the treatment efficacy of surgical management for liver metastasis from colorectal cancer. METHODS: Clinical data of 198 patients with liver metastasis from colorectal cancer, treated from Jan. 1995 to Jan. 2000, were studied retrospectively. Of the 198 patients, 46 (23.2%) received radical resection, 43 (21.7%) received palliative resection, 29 (14.6%) received exploratory operation or supportive treatment, 41 (20.7%) received adjuvant hepatic arterial infusion, and 39 (19.7%) received adjuvant systemic chemotherapy. Survival statuses of the patients in different groups were compared. RESULTS: The median survival time of radical resection group was significantly longer than those of palliative resection group, exploratory operation or supportive treatment group, adjuvant hepatic arterial infusion group, and adjuvant systemic chemotherapy group (37.1 months vs. 14.3, 6.3, 21.3, and 18.7 months, P<0.01). The 5-year survival rates of the 5 groups were 31.2%, 0, 0, 7.5%, and 0, respectively. CONCLUSIONS: Radical resection could improve survival of the patients with liver metastasis from colorectal cancer. Palliative resection has no advantage over adjuvant therapy. Adjuvant hepatic arterial infusion should be applied in the unresectable cases.

[Correlation of multislice spiral CT findings with vascular endothelial growth factor expressions and microvessel density in renal cell carcinoma].

OBJECTIVE: To study the correlation of multislice spiral CT features and vascular endothelial growth factor (VEGF) expressions and microvessel density (MVD) in renal cell carcinoma (RCC). METHODS: Fourth-seven patients with pathologically confirmed RCC were examined by multislice spiral CT, and VEGF expressions and MVD of the RCC and the adjacent normal tissues were determined by immunohistochemistry with specific monoclonal antibodies. RESULTS: VEGF expression and MVD in the RCC and adjacent normal tissues increased with the pathological grades of RCC (P<0.05), VEGF expression was found to significantly correlate with MVD (r=0.67, P<0.01). The positive expression of VEGF and MVD were associated with the findings by multislice spiral CT scan of tumor size, intratumor necrosis, cystic degeneration, intensity signal, lymph node metastases, invasion of the renal vein or inferior vena cava, and invasion of the adjacent organs or distant metastases (P<0.01). CONCLUSION: Multislice spiral CT findings can be indicative of the histopathology of RCC, and some CT findings are closely correlated with MVD and VEGF expressions in RCC, which may help evaluate the biological behavior and malignancy of the tumor and predict tumor invasion and metastasis.

[Residues and distribution character of organochlorine pesticides in stream sediments in southwestern suburb of Nanjing].

Sediments and water samples were collected from different cross sections of 4 tributaries of Yangtze River in southwestern suburb of Nanjing city in spring 2004, and agricultural soil sample were collected also in those valleys. The residues and distribution of organochlorine pesticides in sediments from different streams were determined and the influence of soil character on organochlorine pesticides residues in sediments were studied. Result showed that accumulation of HCHs and DDTs occured significantly in the sediments and related to soil erosion. Residues of organochlorine pesticides in sediments may be influenced obviously by pH of agricultural soil in the valleys. In the alkaline soil, organic matters could be leached easily and entered the stream water and accumulated into sediments, resulting in accumulation of organochlorine pesticides adsorbed on soil organic matters in sediments. In the transfer process of materials from soil to water and sediment, constitutes of organochlorine pesticides in multiphase environmental medium were changed; usually those components easily leached from soils had higher residue levels in sediments. Meanwhile, sedimentation of soil in streams could reduce OCPs input to the Yangtze River.

[Bovine jugular venous conduit treated with the polyepoxy compound].

OBJECTIVE: To determine the feasibility whether the bovine jugular venous conduit (BJVC) can be fixed with polyepoxy compound (PC). METHODS: Twenty-four BJVCs were divided into 3 groups and fixed with polyepoxy compound (PC group, n = 8), glutaraldehyde (GA group, n = 8), and unfixed group (Control group, n = 8), respectively. The morphologic and mechanical properties of BJVCs in the 3 groups, including thickness, diameter, moisture content, denaturation temperature, tensile strength, elongation at break, and fixation index were measured. The rat subcutaneous model for the assessment of tissue calcification was used. The calcium content in bovine jugular vein patches and valves was determined by flame atomic absorption spectrophotometer. RESULTS: There was no difference in the wall thickness, diameter, and tissue water content between PC and the control group, but significant difference was found between GA and PC groups. The mechanical properties of PC group and GA group were not significantly different, but they were better than those of the control group. GA-fixed BJVC samples showed clear calcification, while PC fixed BJVC were calcified significantly less. CONCLUSION: PC is an effective and suitable choice for the treatment of BJVC since it can effectively preserve the structure and the anti-reflow function of valves in bovine jugular vein and it has better anti-calcification properties.

Regulation of DNA methylation on the expression of the FHIT gene contributes to cervical carcinoma cell tumorigenesis.

The purpose of this study was to determine the effect of the FHIT gene on tumorigenesis of cervical cancer. RT-PCR and MTT were used to detect the expression of FHIT and cell proliferation respectively. Flow cytometry was used to test cell cycle and cell apoptosis. The expression of FHIT was not induced at all four cervical cancer cells treated with demethylating agent 5-aza-2'-deoxycytidine (5-Aza-CdR). However, a constant level of FHIT expression was detected in the human umbilical vein endothelial cell before and after 5-aza-dC treatment. The proliferation of all four cervical cancer cells was inhibited evidently when treated with 5-aza-dC and the inhibiting rate of cell growth along with the increasing concentration of 5-aza-dC. There was no obvious inhibiting effect on the growth of the human umbilical vein endothelial cell treated with 5-aza-dC. An increasing G1 phase and high apoptosis rate were detected in all four cervical cancer cells. However, a negligible change was seen in the human umbilical vein endothelial cell. These results suggest that aberrant methylation of the FHIT gene might be a key mechanism for silenced FHIT gene, which could be reactivated and whose tumor suppressing function could be restored by demethylating agent treatment.

[Clinical significance for minimal residual disease detection by 4 color flow cytometry in adult and childhood B lineage acute lymphoblastic leukemia].

OBJECTIVE: To evaluate the clinical significance for minimal residual disease (MRD) detection by 4 color flow cytometry in B lineage acute lymphoblastic leukemia (B-ALL). METHODS: MRD was analyzed and followed up by using two panels of 4 color antibodies, mainly CD34/CD10/CD45/CD19, in 671 consecutive bone marrow specimens and 1 cerebrospinal fluid from 98 B-ALL patients. In 26 cases of them the immunophenotyping informations at diagnosis were not available. RESULTS: Of 671 bone marrow samples, 579 were MRD negative with leukemic cells below 0.0001 and 93 were MRD positive with leukemic cells over 0.0001. Of 93 MRD positive samples, leukemic cells below 0.05 were found in 64 bone marrow samples, meanwhile in the other 29 samples leukemic cells were over 0.05. Twenty patients relapsed, 19 were bone marrow relapse and one center nerves system. Fifteen of them were found MRD positive 7 - 17 weeks before relapse including 6 patients having no immunophenotyping data at diagnosis. The percentages of leukemia cells in these 15 patients were all over 0.0001. Two relapsed patients were MRD negative in 3 and 9 months before relapse, respectively. Two relapsed after MRD monitoring stopped. If MRD level was > 0.0001 at the end of induction chemotherapy and 12 weeks of treatment, the rate of relapse was 50% (6/12), while, it was 7.5% (3/40) in MRD negative patients (P = 0.000). CONCLUSION: Relapses can be predicted by MRD monitoring, if MRD was positive in the early phase of treatment, the risk of relapse was higher. Based on the characteristics of B cells ontogeny, MRD detection can be done independently of immunophenotypic information at diagnosis.

[Comparison of the immunophenotype of patients with B lineage acute lymphoblastic leukemia at diagnosis and relapse].

OBJECTIVE: To compare the leukemia-associated immunophenotypes (LAIP) in patients with B lineage acute lymphoblastic leukemia (B-ALL) at diagnosis and relapse, and investigate its implications for minimal residual disease (MRD) detection. METHODS: The immunophenotype of leukemia cells from 410 newly diagnosed and 6 relapsed patients with B-ALL were detected by four to six antibody combination, mainly CD34/CD10/CD45/CD19 of 4-color CD45/SSC gating flow cytometry (FCM). RESULTS: The proportion of CD45 under-expressed or negative in relapsed patients was much higher than that in newly diagnosed patients, being 69.2% and 37.8% respectively. Immunophenotypic changes occurred in 9 relapsed patients (including 8 hematological relapse and 1 central nerves system relapse) when analyzed by paired samples analysis at diagnosis vs at relapse: 4 cases showed CD45 down-modulation and 2 up-modulation; 4 CD34 down-modulation and 2 CD10 up-modulation, while the expression of CD19 remained no change. MRD was observed in all 7 cases of hematological relapse 2 - 4 months before relapse, and the immunophenotype of MRD cells was the same as that in relapse. CONCLUSION: A high frequency of immunophenotypic changes occurred at relapse and even in MRD before relapse, however the accuracy of MRD monitoring seemed not affected by the FCM strategy used in this investigation.

[Characteristics of atrazine-degrading genetically engineered microorganism (GEM) labeled with green fluorescent protein (GFP)].

The atrazine-degrading genetically engineered microorganism (GEM) was labeled by transforming a plasmid containing green fluorescent protein (GFP) gene. GFP gene could be expressed well in the cell of GEM after transforming. In the LB medium containing antibiotic, the expressing level of GFP was higher than in pure LB medium and in the basic medium. In the stable phase containing antibiotic, the expressing level of GFP was higher than in pure LB medium and in the basic medium. In the stable phase of cell growth, the expressing level of GFP was higher than in the lag phase and in the logarithmic phase. The transformation and expression of GFP gene wouldn't affect the degrading ability of GEM. The level of GFP expression was almost linear with the degrading activity. There exist two states of cells after inoculating GEM labeled with GFP into activated sludge: suspended state and adhered state to sludge floc, and the amount of suspended cells is higher than that adhered to sludge floc.

[Analysis of DC-SIGN and DC-SIGNR genetic polymorphism in Chinese Han population].

OBJECTIVE: To understand the genetic polymorphism of DC-SIGN's and DC-SIGNR's neck regions in normal Chinese Han population, and to obtain the genetic data of the two loci in Chinese Han population. METHODS: The genotypes and alleles of repeat sequences of DC-SIGN and DC-SIGNR neck region were typed by PCR, agarose gel electrophoresis and sequencing. Polymorphism information content (PIC) of DC-SIGNR was calculated. RESULTS: DC-SIGN genetic polymorphism was rare. Allele 7 was most and its frequency was 0.9808. 4-, 5-, 6- and 8- alleles were also found, although their frequencies were very low. Caucasians had only 6- and 8- allele mutants; DC-SIGNR genetic polymorphism was high, its PIC was 0.5312, 4-,5-,6-,7-,8-,9- alleles and 16 genotypes were found in normal Chinese Han population. The differences of 6/5,7/4,7/5,7/6,7/7,9/5,9/7,9/9 genotypes distribution and 5-,6-,7-,9- alleles frequency between normal Chinese Han population and Caucasian population were all extremely distinct (P<0.01). The inserted mutation seemed more in Chinese Hans than Caucasian population. CONCLUSION: DC-SIGN and DC-SIGNR genotypes and alleles distribution in Chinese Han population are significantly different from Caucasian population and with Chinese own population genetic characteristics, compared with Caucasians.

[Determination of trace elements in sika bone powder by inductively coupled plasma mass spectrometry with microwave digestion].

Contents of trace elements in sika bone powder were determined with microwave digestion and inductively coupled plasma mass spectrometry. Under the optimum conditions, the detection limits (3sigma, n = 11) are in the range of 0. 000 6-1. 498 ng x mL(-1) with relative standard deviations of 1.7%-6.8%. The recoveries are between 91% and 109%. The analytical results of national certified reference demonstrated the applicability of the proposed method.

[Experimental study on the survival of venous flap with different pedicle styles].

OBJECTIVE: To study a new method of venous flap that is improved on its persistence and quality. METHODS: New Zealand white rabbits were subdivided randomly into 4 groups. All rabbits were operated by harvesting a flap from the latero-abdominal wall and then sutured it in the original position. Group A: the superficial epigastric vein in the pedicle was left open (only one inflow vein remained). Group B: the pedicle vein of the proximal and distant end were left open (keeping an inflow vein and a principal out). Group C: the pedicle vein and a tributary vein were left open (keeping an inflow vein and a tributary outflow vein). Group D: the pedicle vein and two tributary veins were left open (keeping an inflow vein and two tributary outflow veins). Survival rate, MDA of the tissue, histology and ultra-microstructure were examined. RESULTS: Survival rate of A, B, C, D were improved in order. Statistic difference is significant (P < 0.05) between group and group other than C and D. The content of MDA was heightened with statistically significant differences (A > B > C > D) among the four groups 8 hours postoperatively, but fell back to the normal level in group D and C and kept a high level in group A and B at 72 hour postoperatively. Histology and ultra-microstructure exam showed that degeneration of collagen fiber and karyopyknosis of cell is more obvious in Group A and Group B than Group C and Group D. CONCLUSIONS: The higher survival rate of venous flap is possible by designing the more reasonable venous flap outputs pedicles which can alleviate the high tension dropsy and maintain the valid equilibrium of pour with flow in the venous flap.

[Clinical observation on treatment of active systemic lupus erythematosus with haoqin qingdan decoction].

OBJECTIVE: To observe the therapeutic efficacy and safety of Haoqin Qingdan Decoction (HQD) in treating active stage of systemic lupus erythematosus (SLE). METHODS: One hundred and twenty patients with SLE were randomly assigned into two groups, the treated group receiving HQD and prednisone, and the control group receiving prednisone and cyclophosphamide (CTX), both were treated for 3 months. SLE disease activity index (SLEDAI), various immunological indexes, blood and urine routine, hepatic and renal function, as well as the other adverse reaction of the treatment were observed. RESULTS: The total effective rate in the treated group was 81.67% (49/60), while 76.67% (46/60) in the control group, showing insignificant difference between the two groups. SLEDAI, erythrocyte sedimentation rate (ESR), complement 3 (C3), 24-hr urinary protein excretion and anti-double strain deoxyribonucleic acid (dsDNA) antibody were improved in the treated group, and no obvious adverse reaction was shown. CONCLUSION: HQD is obviously more effective in treating active stage of SLE with high safety.
