Senile seminal vesicle amyloid is derived from semenogelin I.

Senile seminal vesicle amyloid (SSVA), one of the most common forms of localized amyloidosis, is associated with the male aging process. Although it had been posited that the amyloidogenic component originated from exocrine cells and that, on the basis of immunohistochemistry, that the amyloid was composed of lactoferrin, the nature of SSVA was never established definitively. To address this issue, we have used our microanalytic techniques to characterize the structure of the congophilic green birefringent protein extracted from 5 such amyloid-containing specimens. Mass spectrometric analysis revealed that in all cases, the fibrils were composed mainly of polypeptide fragments identical in sequence to the N-terminal portion of the major secretory product of seminal vesicles, namely semenogelin I (SgI). Although lactoferrin was detected in 3 instances, the trace amount and seemingly intact form of this molecule indicated that it was not the amyloidogenic molecule. The SgI nature of the amyloid was confirmed through demonstration that the deposits were immunostained specifically with Sgl-reactive antibodies. The results of our research provide unequivocal evidence that SSVA is derived from SgI, and we provisionally designate this form of amyloidosis as ASgI.

Mitogenomic perspectives on the origin and phylogeny of living amphibians.

Establishing the relationships among modern amphibians (lissamphibians) and their ancient relatives is necessary for our understanding of early tetrapod evolution. However, the phylogeny is still intractable because of the highly specialized anatomy and poor fossil record of lissamphibians. Paleobiologists are still not sure whether lissamphibians are monophyletic or polyphyletic, and which ancient group (temnospondyls or lepospondyls) is most closely related to them. In an attempt to address these problems, eight mitochondrial genomes of living amphibians were determined and compared with previously published amphibian sequences. A comprehensive molecular phylogenetic analysis of nucleotide sequences yields a highly resolved tree congruent with the traditional hypotheses (Batrachia). By using a molecular clock-independent approach for inferring dating information from molecular phylogenies, we present here the first molecular timescale for lissamphibian evolution, which suggests that lissamphibians first emerged about 330 million years ago. By observing the fit between molecular and fossil times, we suggest that the temnospondyl-origin hypothesis for lissamphibians is more credible than other hypotheses. Moreover, under this timescale, the potential geographic origins of the main living amphibian groups are discussed: (i) advanced frogs (neobatrachians) may possess an Africa-India origin; (ii) salamanders may have originated in east Asia; (iii) the tropic forest of the Triassic Pangaea may be the place of origin for the ancient caecilians. An accurate phylogeny with divergence times can be also helpful to direct the search for "missing" fossils, and can benefit comparative studies of amphibian evolution.

Maintaining a conserved methylation in plant and insect U2 snRNA through compensatory mutation by nucleotide insertion.

The extensive post-transcriptional modification of U2 snRNA is required for spliceosome assembly and pre-mRNA splicing in vertebrates. However, the rare modification of U2 snRNA in yeast implies a different mechanism for regulating spliceosome biogenesis in single-celled eukaryotes. To understand the evolutionary pattern of U2 snRNA methylation, we determined for the first time, the 2'-O-methylations of U2 snRNA in Oryza sativa, Arabidopsis thaliana and Drosophila melanogaster, and revealed two methylations which are conserved in a crucial region of U2 snRNA in plants. Interestingly, one of the methylations, U2-Cm29 is also methylated in D. melanogaster, but not in vertebrates. According to the methylation of U2-C29, computational analysis of databases identified three canonical box C/D snoRNAs, named OsmgU2-29, AtmgU2-29 and DmmgU2-28, as small methylation guides of U2 snRNA from O. sativa, A. thaliana and D. melanogaster, respectively. Although very divergent in their sequence, the three snoRNAs exhibit in common an 11 nucleotide-long sequence complementarity to corresponding U2 snRNA, implying a functional constraint on the modification during evolution. Interestingly, a nucleotide is found to be inserted both in U2 snRNA and DmmgU2-28 and maintains a perfect match of duplex specifying the methylation of C28 in Drosophila U2 snRNA. This is the first time a new model is being provided for compensatory mutations between a small guide RNA and its target by nucleotide insertion, instead of the known nucleotide substitution. In contrast to small Cajal body-specific RNAs (scaRNAs), the snoRNAs are similar to the reported singlet guide RNAs and are known to localize in nucleolus.

Differential immunoscreening identifies a glycosylation variant of the epidermal growth factor receptor in ME-180 cervical carcinoma cells.

In this report, we describe the development and characterization of an anti-ME-180 cervical cancer-specific epidermal growth factor (EGF) receptor monoclonal antibody (MAb). This MAb, 6C7, specifically binds to ME-180 cervical cancer cells and not to normal cervical epithelial cells. By immunoaffinity chromatography, we have shown that the 6C7 antibody binds to a 205-kDa protein. Subsequent mass spectrometry sequencing analysis identified this protein as an EGF receptor. In addition, treatment of the ME-180 EGF receptor with N- and O-linked glycosidases indicated that this antibody binds to the carbohydrate portion of the glycoprotein. Moreover, Western blotting analysis with an anti-EGF receptor antibody indicated that this protein is present in abundance in all cervical cancer cell lines, including ME-180, HeLa, Ca Ski, HT-3, SiHa, and Hs 588.T. However, the 6C7 antibody only binds to the EGF receptor from ME-180 cells, suggesting that this protein is differentially glycosylated in ME-180 cells, compared to other cervical cancer cell lines. Finally, we have shown that this antibody could selectively block EGF-mediated cell proliferation in ME-180 cells but not in HeLa cells. Overall, our study suggests that the differentially glycosylated EGF receptor could potentially serve as a unique target for the immunotherapeutic treatment of cervical cancer.

Lornoxicam pharmacokinetics in relation to cytochrome P450 2C9 genotype.

AIMS: To investigate the pharmacokinetics of lornoxicam and the relationship with CYP2C9 polymorphism in healthy Chinese subjects. METHODS: A single oral dose of 8 mg lornoxicam was administered to 18 healthy Chinese male subjects. Plasma was sampled for 24 h post dose, and plasma concentrations of lornoxicam were measured using a validated LC/MS/MS method. CYP2C9 genotype was determined by polymerase chain reaction-based restriction fragment length polymorphism or by direct sequencing of the coding region of the CYP2C9 gene. RESULTS: Of the 18 subjects, one subject was found to be a very poor metabolizer of lornoxicam with a long t(1/2) of 106 h, a low CL/F of 0.71 ml min(-1), and a high AUC(0-infinity) of 187.6 microg ml(-1) h. Genotyping studies revealed that this subject was heterozygous for CYP2C9*3 and a new variant CYP2C9 allele. Of the other 17 subjects, 13 were *1/*1 carriers, three were *1/*3 carriers, and one was a *1/*2 carrier. Mean AUC(0-infinity) values (95% confidence intervals) of lornoxicam were 9.25 (6.55, 11.95) vs. 4.75 (3.55, 5.95) microg ml(-1) h in *1 heterozygotes vs.*1 homozygotes, and mean CL/F values were 14.8 (10.2, 19.4) vs. 32.9 (24.5, 41.3) ml min(-1), respectively (P < 0.05 for both AUC and CL/F). CONCLUSIONS: The results show that the pharmacokinetics of lornoxicam are dependent on CYP2C9 polymorphism. In particular, the presence of the CYP2C9*3 allele impairs the oral clearance of lornoxicam.

Humoral immunity to vimentin is associated with cardiac allograft injury in nonhuman primates.

Immunity to autologous protein has not previously been described following nonhuman primate cardiac transplant. Native hearts and cardiac allografts from cynomolgus monkeys were assessed by immunohistology for vimentin, a highly conserved intermediate filament protein. IgM and IgG to vimentin were measured in serial sera from untreated (n = 4) or cyclosporine (CsA)-treated (n = 8, 2 with ATG) cardiac allograft recipients, and in groups treated with anti-CD154 antibody with (n = 6) or without ATG (n = 28). IgM or IgG reactive with vimentin was elaborated within 30 days with unmodified acute rejection (3/4) or in CsA-treated animals (5/6). CD154 blockade did not prevent anti-vimentin IgM (14/28) but tended to delay the IgG response during therapy (anti-CD154: 8/28, p = 0.10 vs. CsA; anti-CD154+ATG: 2/6). CAV and alloantibody were seen in 25 of 26 animals with grafts surviving over 30 days, including seven animals without increasing anti-vimentin antibody. Anti-vimentin antibodies and vascular complement deposition were found in rejected hearts. Acute and chronic alloimmunity disrupt modulation of autoreactivity to vimentin through pathways, which are resistant to CsA, but may be partially regulated by CD154.

Epidemiologic study of the predisposing factors in erythema toxicum neonatorum.

BACKGROUND: Erythema toxicum neonatorum (ETN) is a very common disease, but its predisposing factors are still unknown. OBJECTIVE: To determine the predisposing factors of ETN. METHODS: Seven hundred and eighty-three neonates born in the same hospital during the same period were investigated, and the factors predisposing to ETN were evaluated in a case-control study. RESULTS: (1) The incidence of ETN is about 43.68%, and it is significantly higher in males than in females (p < 0.001). (2) Term birth (p < 0.05), first-pregnancy birth (p < 0.001), the birth season (summer and autumn, p < 0.005), being fed with milk powder substitute or a mixed diet (p < 0.001) and vaginal delivery (p < 0.001) are the predisposing factors of ETN. (3) The severity of ETN in neonates born by vaginal delivery is significantly correlated with the total length of labor (p < 0.001). CONCLUSION: Our findings suggest that environmental factors play an important role in the onset of ETN.

[Effect of ornidazole on sperm in rats and its mechanism of action].

OBJECTIVE: To explore the reductive effect of ornidazole on sperm motility in rats and its mechanism of action. METHODS: Twenty rats were randomly divided into three groups, a low dosage group (LD group, n = 5), a high dosage group (HD group, n = 8) and a normal control group (n = 7). Ornidazole (200 mg/kg, 400 mg/kg) was given to the LD and HD groups, and 0.5% carboxymethylcellulose sodium (CMC) administered to the normal control, all for 20 consecutive days. Immediately after, sperm density, motility and the morphological changes of the testis and epidiclymis were measured, and the concentrations of lactate dehydrogenase (LDH), alpha-glycosidase, malondialdehyde (MDA) and fructose in the testis and epididymis tissues were monitored. RESULTS: Compared with the normal control, there were no obvious changes in sperm density (P > 0.05), but a significant decrease in sperm motility in the LD and HD groups (P < 0.01), and the concentration of LDH obviously declined (P < 0.01) while that of MDA distinctly increased in the HD group (P < 0.05). CONCLUSION: Spermatogenic cells could be damaged by the increase of inhibiting MDA, while sperm motility could be decreased by inhibiting energetic transferase or non-protein substance in the epididymis. This might be one of the mechanisms of ornidazole on weak sperm models in rats.

[The treatment of Semont maneuver for 40 patients with posterior semicircular canal benign paroxysmal positional vertigo].

OBJECTIVE: To evaluate the effectiveness of Semont maneuver for posterior semicircular canal benign paroxysmal positional vertigo (PC-BPPV). METHOD: Forty patients with PC-BPPV were treated with Semont maneuver from January to August in 2004. RESULT: After a single treatment session of Semont maneuver,the symptoms disappeared completely in 32 patients, the first success rate was 80%. Among 19 patients who had only 1 week duration of PC-BPPU, the symptoms disappeared in 16 patients after a single treatment. The symptoms disappeared in 3 patients after twice, 2 patients after 3 times and 1 patient after 4 times of Semont maneuver sessions, the total success rate was 95%. No effectiveness was found 2 patients; PC-BPPU recurred in 5 patients during follow-up and the recurrent rate was 12.5%. it was still effective for another Semont maneuver therapy session. CONCLUSION: The Semont maneuver therapy is safe and effective for PC-BPPV. Repeated Semont maneuver therapy is still effective for them.

[Construction of adeno-associated virus vector containing human papilloma virus 16 E7 gene and its expression in eukaryotic cells].

OBJECTIVE: To construct recombinant adeno-associated virus vector containing human papilloma virus (HPV) 16E7 gene and identify its effectiveness of expression in eukaryotic cell. METHODS: Recombinant adeno-associated virus particles containing HPV16E7 gene were generated by co-transfection of plasmids pAAV-MCS-E7, pAAV-RC and pAAV-helper into HEK293 cells. The viral particles were collected and purified. The vector titer was measured by southern blot. After the eukaryotic cells were transfected by virus particles, RT-PCR and western blot analysis were performed to identify rAAV expression. RESULTS: The recombinant adeno-associated virus vector containing HPV16E7 gene was correctly constructed. The vector titer measured by southern blot was approximately 1 x 10(11)/ml. After the eukaryotic cells were transfected by the recombinant adeno-associated virus vector, the expression of HPV16E7 gene was identified by RT-PCR and western blot. CONCLUSIONS: Recombinant adeno-associated virus HPV16E7 vector is successfully constructed and can stably express in eukaryotic cells.

[Impact of cytochrome P450 CYP2C9 variant allele CYP2C9 * 3 on the pharmacokinetics of glibenclamide and lornoxicam in Chinese subjects].

AIM: To investigate the impact of CYP2C9 * 3 on the pharmacokinetics of glibenclamide and lornoxicam. METHODS: CYP2C9 * 3 was measured in 83 non-related Chinese subjects by PCR-RFLP. The pharmacokinetics of lornoxicam and glibenclamide were investigated in 18 subjects (7 with CYP2C9 * 1/* 3 genotype and 11 with * 1/* 1 genotype). Glibenclamide and lornoxicam in plasma were determined by the sensitive liquid chromatography-tandem mass spectrometry, separately. RESULTS: After a single oral dose of 2.5 mg glibenclamide, C(max) was (70.0 +/- 11.5) microg x L(-1) in CYP2C9 * 1/ * 3 subjects and (51.9 +/- 12.3) microg x L(-1) in * 1/ *1 subjects. AUC(0-infinity) were (435 +/- 47) vs (287 +/- 95) microg x h x L(-1) (in * 1/ * 3 vs * 1/ *1 subjects), and CL/F were (96 +/- 9.3) vs (160 +/- 51) mL x min(-1), respectively. Statistic analysis results indicated that glibenclamide AUC(0-infinity) was significantly higher (1.5-fold) and subsequently CL/F was significantly lower (40%) in CYP2C9 * 1/ * 3 subjects than those in * 1/ * 1 subjects (P < 0.01). After a single oral dose of 8 mg lornoxicam, C(max) was (1.54 +/- 0.24) mg x L(-1) in CYP2C9 * 1/ * 3 subjects and (1.19 +/- 0.37) mg x L(-1) in * 1/ * 1 subjects. AUC(o-infinity were (14.9 +/- 2.2) vs (6.92 +/- 1.48) mg x h x L(-1) (in * 1/ *3 vs * 1/ * 1 subjects), and CL/F were (9.1 +/- 1.2) vs (20.1 +/- 4.6) mL x min(-1), respectively. Statistic analysis results indicated that lornoxicam AUC(0-infinity) was significantly higher (2. 2-fold) and subsequently CL/F was significantly lower (55% ) in CYP2C9 * 1/ * 3 subjects than those in * 1/ * 1 subjects (P < 0.001). CONCLUSION: CYP2C9 * 3 greatly affects both the pharmacokinetic profiles of glibenclamide and lornoxicam. The elimination of these drugs significantly decreased in subjects with CYP2C9 * 1/ * 3 genotype, especially lornoxicam.

[Study of water-soluble compounds from fungus garden of Odontotermes formosanus].

OBJECTIVE: To study water-soluble compounds from fungus garden of Odontotermes formosanus. METHODS: The chemical constituents of fungus garden were analyzed and identified by GC-MS. RESULTS: 28 compounds were separated and 11 chemical constituents were identified. CONCLUSION: The main constituents in water-solubles from fungus garden of Odontotermes formosanus are palmitic acid, linolei acid and oleic aid.

The SNPlex genotyping system: a flexible and scalable platform for SNP genotyping.

We developed the SNPlex Genotyping System to address the need for accurate genotyping data, high sample throughput, study design flexibility, and cost efficiency. The system uses oligonucleotide ligation/polymerase chain reaction and capillary electrophoresis to analyze bi-allelic single nucleotide polymorphism genotypes. It is well suited for single nucleotide polymorphism genotyping efforts in which throughput and cost efficiency are essential. The SNPlex Genotyping System offers a high degree of flexibility and scalability, allowing the selection of custom-defined sets of SNPs for medium- to high-throughput genotyping projects. It is therefore suitable for a broad range of study designs. In this article we describe the principle and applications of the SNPlex Genotyping System, as well as a set of single nucleotide polymorphism selection tools and validated assay resources that accelerate the assay design process. We developed the control pool, an oligonucleotide ligation probe set for training and quality-control purposes, which interrogates 48 SNPs simultaneously. We present performance data from this control pool obtained by testing genomic DNA samples from 44 individuals. in addition, we present data from a study that analyzed 521 SNPs in 92 individuals. Combined, both studies show the SNPlex Genotyping system to have a 99.32% overall call rate, 99.95% precision, and 99.84% concordance with genotypes analyzed by TaqMan probe-based assays. The SNPlex Genotyping System is an efficient and reliable tool for a broad range of genotyping applications, supported by applications for study design, data analysis, and data management.

[Study on the metabolic health risks of body mass index and waist circumference].

OBJECTIVE: To evaluate the effect of body mass index (BMI) and waist circumference (WC) on metabolic health risks (MHR). METHODS: Based on data from two community populations (Suzhou city and Changshu city) derived from a project on 'multiple metabolic disorders and metabolic syndrome' in Jiangsu province. 1604 subjects were divided into groups as normal weight (BMI: 18-23.9), overweight (BMI: 24-27.9) and obesity (BMI > or = 28) according to BMI measurement. According to the results from WC measurement, they were grouped as abdominal obesity (male: > or = 85 cm; female: > or = 80 cm) and normal. Relative risks (RR) for hypertension, hyperglycemia, dyslipidemia were calculated. RESULTS: In males and females, the mark values of hypertension, hyperglycemia, dyslipidemia as well as the prevalence of MS were related to BMI and WC. The relativity remained true when the confounding factors were under control. Among the overweight group, the aRR of all these metabolic health risks were basically lower than those when the overweight group and the abdominal obesity group were combined. Both in males and females, when groups combining normal weight with abdominal obesity group, the aRR of all these metabolic health risks were higher than overweight group combining with normal WC group in most of the cases. CONCLUSION: As to the effect of BMI and WC on metabolic health risks, WC--the mark of abdominal obesity seemed to be more effective than BMI.

[The relation between blood pressure level in prehypertensives and multiple metabolic disorders].

OBJECTIVE: To study the relation between blood pressure level in prehypertensives and multiple metabolic disorders. METHODS: Based on the populations in Jiangsu province from the project of Multiple Metabolic Disorders and Metabolic Syndrome, a case-control study was conducted including 585 cases of isolated systolic prehypertensive,746 cases of diastolic-systolic prehypertensive, 340 cases of isolated diastolic prehypertensive and 1755 individuals with normal blood pressure. RESULTS: The occurrence of triglyceride(TG), body mass index(BMI) and waist circumference(WC) disorders were significantly higher in the three case groups than in the normal blood pressare group. Among case groups, 50% of them had at least one more metabolic abnormalities as compared to 35% in the controls. CONCLUSION: Among patients in prehypertensive stage, TG disorder was most popular in multiple metabolic disorders. Prehypertensive was associated with WC, BMI and age, indicating that there had already been collections of cardiovascular risk factors in the prehypertensive stage.

Genetic diversity of small eukaryotes from the coastal waters of Nansha Islands in China.

Population structures and genetic diversity of the small eukaryotic plankton from the coastal waters of the Nansha Islands in China were investigated. Two genes libraries using 18S rDNA of the marine small eukaryotes were constituted, and 323 clones were identified within alveolates (more than 43%), acanthareas, viridiplantaes, and stramenopiles. Many novel clones were detected in the two libraries, including two groups of alveolates and two clades related to both acanthareas and polycystineas. Several sequences unrelated to any other known eukaryotes may represent early branches in the phylogenetic tree. Our results reveal that there is a high diversity and abundance of small eukaryotes in the marine regions of China.

Electrochemical studies of danthron and the DNA-danthron interaction.

Danthron is an important natural occurring component in laxative drugs. In this paper, electrochemical investigation of danthron and its interaction with DNA is reported. Via the electrochemical approach assisted by ultraviolet-visible (UV-Vis) spectroscopy, we have proved that danthron intercalates into DNA strands forming some nonelectroactive complexes, which results in the decrease of redox peak currents of danthron. In addition, the decrease of the peak currents is proportional to the concentration of DNA. The difference between the interaction of danthron with double-stranded DNA (dsDNA) and with single-stranded DNA (ssDNA) has also been studied. This character implies the potential of danthron to discriminate dsDNA and ssDNA.

An electrochemical study of hemoglobin in water-glycerol solutions.

The effect the composition of a water-glycerol mixture has on the electrochemical properties of hemoglobin (Hb) is studied. With the increased glycerol concentrations, the peak-to-peak separation of hemoglobin is found to increase from approximately 40 to 200 mV, with the apparent standard potential of hemoglobin negatively shifted, which demonstrate that the electron-transfer activity of hemoglobin will decrease at relatively high glycerol concentrations and the oxidized state of hemoglobin will be more stable with the increasing glycerol concentrations. Meanwhile, the electrocatalytic activity of hemoglobin to hydrogen peroxide, as well as the binding of ligands or effectors to hemoglobin in the presence of glycerol, are also been investigated. Our studies indicate that glycerol will decrease the electrocatalytic activity of hemoglobin, while have little effect on the microenvironment around the heme site.
