[Resveratrol induces HepG2 cell apoptosis by depolarizing mitochondrial membrane].

OBJECTIVE: To investigate the effects of resveratrol on the proliferation, apoptosis, mitochondrial membrane potential and cell morphology of human liver cancer cell line HepG2. METHODS: The changes in HepG2 cell growth and proliferation in response to resveratrol treatment were evaluated by MTT assay, and resveratrol-induced apoptosis of HepG2 cells was investigated by flow cytometry. Inverted microscope and electron microscope were employed for observing morphological changes of the treated cells. The whole-cell mitochondrial membrane potential was measured in separate experiments using two fluorimetric probes, rhodamine123 and TMRE, respectively. HepG2 cells treated with rhodamine123 were analyzed by flow cytometry and cells treated with TMRE by confocal microscope. RESULTS: MTT assay showed that low concentrations of resveratrol produced no significant effect on the growth of HepG2 cells, whereas at high concentrations, resveratrol could obviously inhibit the cell growth in a time- and dose-dependent manner. Resveratrol also induced apoptosis of HepG2 cells, and after a 24-hour treatment, resveratrol caused sharp increment of the mitochondria membrane potential. CONCLUSION: Resveratrol is capable of inhibiting the proliferation of HepG2 cells and inducing cell apoptosis by depolarizing mitochondrial membrane potential.

[As2O3-induced permeability transition pore opening in mitochondria depends on Ca2+].

OBJECTIVE: To explore the mechanism of arsenite-induced permeability transition pore (PTP) opening and the role of Ca(2+). in As(2)O(3)-induced PTP opening. METHOD: The mitochondria were prepared from Wistar rat liver and mitochondrial swelling was assessed spectrophotometrically at 540 nm to evaluate PTP opening. The membrane potential of the mitochondria was measured with fluorescence spectrophotometry. RESULTS: PTP opening was induced with 10 micromol/L As(2)O(3) in the presence of 10 micromol/L Ca(2+), and the absorbance at 540 nm of the mitochondria did not decrease in response to exclusive treatment with 10 micromol/L As(2)O(3), or to 10 micromol/L As(2)O(3) plus 10 micromol/L Ca(2+) treatment with 0.5 mmol/L EGTA pretreatment. Treatment with As(2)O(3) at 0, 5, 10 and 20 micromol/L in the presence of 50, 20, 10 and 5 micromol/L Ca(2+), respectively, resulted in decreased absorbance at 540 nm of the mitochondria. CONCLUSION: Ca(2+) mediates As(2)O(3)-induced PTP opening. As(2)O(3) lowers Ca(2+) threshold necessary for eliciting PTP opening and thereby regulates cell apoptosis.

Comparative studies of duodenal and macrophage ferroportin proteins.

Intestinal epithelial cells and reticuloendothelial macrophages are, respectively, involved in diet iron absorption and heme iron recycling from senescent erythrocytes, two critical processes of iron homeostasis. These cells appear to use the same transporter, ferroportin (Slc40a1), to export iron. The aim of this study was to compare the localization, expression, and regulation of ferroportin in both duodenal and macrophage cells. Using a high-affinity purified polyclonal antibody, we analyzed the localization and expression of ferroportin protein in the spleen, liver, and duodenum isolated from normal mice as well as from well-characterized mouse models of altered iron homeostasis. Ferroportin was found to be predominantly expressed in enterocytes of the duodenum, in splenic macrophages, and in liver Kupffer cells. Interestingly, the protein species detected in these cells migrated differently on SDS-PAGE. These differences in apparent molecular masses were partly explained by posttranslational complex N-linked glycosylations. In addition, in enterocytes, the transporter was mostly expressed at the basolateral membrane, whereas in bone marrow-derived macrophages, ferroportin was found predominantly localized in the intracellular vesicular compartment. However, some microdomains positive for ferroportin were also detected at the plasma membrane of macrophages. Despite these differences, we observed a parallel upregulation of ferroportin expression in tissue macrophages and enterocytes in response to iron-restricted erythropoiesis, suggesting that iron homeostasis is likely maintained through coordinate expression of the iron exporter in both intestinal and phagocytic cells. Our data also confirm a predominant regulation of ferroportin through systemic regulator(s) likely including hepcidin.

[Adrenal incidentalomas: report of 126 cases].

OBJECTIVE: To evaluate the diagnosis and treatment of adrenal incidentalomas. METHODS: One hundred and twenty-six patients were analyzed, in which 98 patients underwent operation. RESULTS: Of all types of the adrenal incidentalomas, 52 (41.3%) of them were adenomas and they were the most often; 43 (34.1%) of them were hypersecretory adrenal tumors, most of them were pheochromocytomas and Conn adenomas; all nonhypersecretory adrenal adenomas were under 6 cm, and all adrenal carcinomas were above 6 cm. CONCLUSIONS: It is important to search for hypersecretory adrenal tumors and to detect malignant adrenal tumors. To nonhypersecretory adrenal adenomas, the sizes of tumors are the most important indexes in determining weather the tumors are benign or malignant and weather the tumors need to be treated with operation or not.

[Clinical significance of RANTES and MIP-1 alpha in acute rejection episode in kidney transplantation].

OBJECTIVE: To discuss the correlation between acute rejection episodes and expression of regulated upon activation normal T cell expressed and secreted (RANTES) and macrophage inflammatory protein-1 alpha (MIP-1 alpha) gene following kidney transplantation. METHODS: A total of 76 kidney biopsies (episode biopsy) were performed on both 57 patients with allograft dysfunction following transplantation and 19 patients without rejection, and the latter were served as controls. All acute rejections were confirmed by histological examination. The expressions of RANTES and MIP-1 alpha mRNA in all samples were assayed by reverse transcription polymerase chain reaction. RESULTS: Thirty-eight (66.7%) of 57 cases with acute rejection had strong expression of RANTES, and 41 (72%) had expression of MIP-1 alpha. Only four (21%) of 19 controls had mild expression of RANTES, and 5 (26%) had expression of MIP-1 alpha, which evoked a statistical significance (P < 0.001) when compared with those with acute rejections. CONCLUSIONS: The expressions of chemokines RANTES and MIP-1 alpha are important in the process of immune reaction in the rejection of transplanted kidney, which may imply a potential way for early diagnosis and treatment of acute rejection after further research.

[Validation of lipids on body mass index reference recommended by Obesity Working Group, International Life Science Association of China].

OBJECTIVE: To assess the relationship between overweight, obesity and blood lipid profiles of children and adolescents and to validate body mass index (BMI) cutoff points for overweight and obesity screening to Chinese children and adolescents, recommended by Working Group of Obesity, China (WGOC), International Life Science Association. METHODS: 2293 children and adolescents (1124 males and 1169 females), aged between 10 and 18 years, were randomly selected as samples from 6 schools in Beijing area. Fasting serum lipids including total cholesterol (TC), total triglycerides (TG), high density lipoprotein cholesterol (HDLC), thropometrical index as weight and height were measured. BMI equals to weight in kilograms were then divided by the square of height in meters. RESULTS: According to BMI cutoff points recommended by WGOC, samples fell into 3 groups including normal group (BMI < 85 percentiles), overweight group (BMI >or= 85 and < 95 percentiles) and obesity group (BMI >or= 95 percentiles). Results clearly showed an increase of both serum TC and TG and a decrease of HDLC when BMI was increasing, among most age groups regardless of sex difference and the difference among BMI groups was statistically significant (P < 0.05 or P < 0.01). CONCLUSIONS: Results of this study indicated that there was an obvious dose-effect relationship between BMI and lipid profiles which accounted for some rationality of the BMI cutoff points recommended by WGOC. The authors reckoned the findings important to managing relevant adult diseases during childhood, in China.

[Expression of HGF and its receptor c-Met in renal cell carcinoma].

OBJECTIVE: To study the clinical significance of expression of hepatic growth factor (HGF) and its receptor c-Met in renal cell cancer (RCC). METHODS: Twenty-six patients with RCC and 10 benign renal tumor patients were examined. The expression of HGF and c-Met mRNA was detected using Northern blot. GAPDH was used as the internal control. RESULTS: Twenty-one out of 26 patients with RCC had positive gene expression of HGF and c-Met with the positive rate of 80.8%. According to the tumor TNM staging, the more advanced the cancer had a stronger expression of HGF and c-Met, the expression of c-Met was higher than that of HGF in the RCC. Whereas in the control group with benign tumor, the expression of c-Met was rather mild. CONCLUSION: The result suggests that HGF and its receptor c-Met may play an important role in the development and progression of renal cancer, which may also imply a potential clinical significance in diagnosing and assessing prognosis of RCC.

[Analysis of the factors associated with prognosis in patients with Ph chromosome positive adult acute lymphoblastic leukemia].

OBJECTIVE: To investigate factors associated with survival of patients with Ph chromosome positive adult acute lymphoblastic leukemia (aALL) in a period of 11 years. METHODS: All the clinical parameters of 31 Ph positive patients were statistically analyzed by SPSS software. RESULT: Ph(+) patients account for 15.3% (31/203) of all the aALL patients. Clinically, these patients manifested older in age, higher white blood cell counts with high blast fractions and lower platelet counts (PC). Phenotypically 82.6% of them were common ALL, 39.1% coexpressed myeloid antigens, and 56.5% expressed CD34 antigen. 65.4% of them (17/26) achieved complete remission (CR) and the median remission and survival durations were 4 months and 8 months, respectively. Patients with Ph(+) and additional chromosomal aberrations accounted for 42% of all the Ph(+) patients, including monosomy 7, +Ph, del(9)(p11-12) and add/t(16)(p13), and they had lower PC as compared with those with sole Ph(+) (P = 0.012) and variant Ph translocation (P = 0.01). CD34 positive patients had a shorter remission and survival duration than CD34 negative ones (0 vs 9 months for median remission time, P = 0.024; and 6 vs 12 months for median survival time, P = 0.034). There was no evidence to support the correlation between myeloid antigen expression and survival time in these patients. CONCLUSION: Ph(+) aALL is associated with adverse prognosis and CD34 expression is a poorer prognostic factor in Ph(+) aALL patients. There is no significant clinical difference between Ph(+) aALL with or without additional chromosomal aberrations.

[Differential expression of a homing-related molecule repertoire among umbilical cord blood, mobilized peripheral blood and bone marrow-derived hematopoietic stem/progenitor cells].

OBJECTIVE: To compare the expression profiles of a set of homing-related molecules (HRM) repertoire expressed on hematopoietic stem/progenitor cells (HS/PC) from different sources. METHOD: The expression levels of HRM on HS/PC from umbilical cord blood (UCB), mobilized peripheral blood (mPB) and bone marrow (BM) were assessed using a highly sensitive 4-color flow cytometric analysis. RESULTS: UCB-derived CD34(bright) cells, as well as mPB- and BM-derived CD34(bright) cells strongly expressed CD44, CD11a, CD18, CD62L, CD31 and CD49d. On the other hand, significantly lower expressions of CD49e, CD49f, CXCR-4 and CD54 on UCB-derived CD34(bright) and CD34(bright)CD38(-) cells, compared with those on mPB- and BM-derived CD34(bright) and CD34(bright)CD38(-) cells, were observed. None of UCB-, mPB- and BM-derived CD34(bright) cells expressed other chemokine receptors, including CCR-1, CCR-2, CCR-3, CCR-5, CXCR-1, CXCR-2, CXCR-3 and CXCR-5. Another striking finding was that only mPB-derived CD34(bright) cells expressed significant levels of both the matrix metalloproteinases MMP-2 \[(11.4 +/- 4.9)%\] and MMP-9 \[(27.6 +/- 7.8)%\]. CONCLUSION: HS/PC from UCB have some defects of expression of HRM repertoire, which might partly explain the cause(s) of delayed hematopoietic reconstitution after UCB transplant.

[Study on etiology and pathology of severe acute respiratory syndrome].

OBJECTIVE: To investigate the clinicopathologic characteristics of severe acute respiratory syndrome (SARS). METHODS: Three autopsy cases were studied retrospectively. Routine HE stain was used to study all the cases. Part of the lung tissue specimens were studied further with Macchiavello's stain, viral inclusion body stain, reticulin and PAS stains, immunohistochemistry, thin sections with staining, light microscopy and transmission electronic microscope investigation. RESULTS: The earliest symptom of all 3 cases was hyperpyrexia and followed by progressive dyspnea and appearance of lung field shadows in X rays findings. Pulmonary lesions included: bilateral and extensive consolidation, localized hemorrhage and necrosis, desquamative alveolitis and bronchitis, alveolar proliferation and desquamation, accumulation of protein exudates, mononuclear cells, lymphocytes, and plasma cells as well as hyaline membrane formation in alveoli and viral inclusion bodies were seen in the alveolus epithelial cells. The exudated organization tended to become glomeruloid organizing pneumonitis in a few avaoli. Lesions of the immune organs included: large patchy necrosis in the spleens and localized necrosis in the lymph nodes were seen. Bone marrow became restrained. There were lesions of systemic small vasculitis including edema of the perivascular tissue and vascular wall of the small veins with localized fibrinoid necrosis distributing in the heart, lungs, kidneys, adrenal glands and the striated muscles accompanying with mononuclear cells and lymphocytes infiltration. Thrombosis was seen in part of the small veins. In addition, there were also the systemic poisonous changes including: degeneration and necrosis of the parenchyma cells in lungs, liver, kidneys, heart and adrenals. Electronic microscopy demonstrated clusters of virus particles seen in the lung tissue. CONCLUSION: SARS is a systemic disease. Lungs, immune system and systemic small vessels are the main target organs attacked by the virus. Extensive consolidation of lungs, formation of hyaline membrane to a large extent, respiratory distress and decrease of immune function are the main causes of death.
