[Expression of VEGF165 and VEGF121 genes in MC3T3-E1 cell line and biological function of their products].

OBJECTIVE: To construct vectors stably expressing bioactive VEGF165 and VEGF121 in MC3T3-E1 cells. METHODS: VEGF165 and VEGF121 genes were obtained by RT-PCR from HL-60 cells and inserted into eukaryotic expression plasmid pcDNA3.1(+). The constructed plasmids containing right sequence of target genes were transfected into MC3T3-E1 cells by lipofectin. The cells were limitedly diluted twice and selected by G418, the expression of VEGF was tested by ELISA, Western-blot and immunofluorescence essay. The bioactivity of expressed products was proved by MTT method. RESULT: DNA sequencing indicated the sequences of the obtained VEGF165 and VEGF121 were identical to the those reported in Genebank, and the newly-constructed plasmids were VEGF165/pcDNA3.1(+) and VEGF121/pcDNA3.1(+). The results of ELISA, Western-blot and immunofluorescence showed that the transfected cells could express VEGF165 and VEGF121 proteins. And MTT proved the proteins were bioactive. CONCLUSION: The transfected MC3T3-E1 cells could stably express high levels of bioactive VEGF165 and VEGF121.

[Sternocleidomastoid myocutaneous flap for reconstruction after resection of carcinoma in floor of mouth].

OBJECTIVE: To evaluate the clinic outcome and indications of sternocleidomastoid (SCM) myocutaneous flap for reconstruction after resection of carcinoma in floor of mouth. METHODS: Thirty-two patients underwent SCM myocutaneous flap for reconstruction of the floor of mouth were reviewed. According to the clinical data and the follow-up, the appearance and function were analyzed. RESULTS: Only one myocutaneous flap was partial necrosis in termination of the flap, and growth with granular. In eight cases, partial epidermal loss over the skin paddle occurred with secondary healing with mucosal epithelium. All patients had oral diet with good separation of the oral floor and the tongue, and had no difficulty in speech intelligibility. The patients' appearance, function and the overall quality of life were improved. CONCLUSION: The SCM myocutaneous flap appears to be easy to use and suitable for reconstruction of the defect after resection of oral floor carcinoma, the indications of this flap should be selected carefully.

[Effects of arsenic trioxideon the cell apoptosis and hTERT mRNA of human tongue cancer cells].

OBJECTIVE: To study the effects and its mechanisms of arsenic trioxide (As2O3) on cell growth and human telomerase reverse transcriptase (hTERT) of human tongue cancer cells (Tca8113 cell line). METHODS: The growth inhibition rates of Tca8113 by various concentrations of As2O3 were detected by MTF method. Cell apoptosis was detected by FCM labeled with Annexin V-FITC. hTERT gene expression was detected by RT-PCR method. hTERT protein of Tca8113 cells was determined by Western blot assay. RESULTS: The results showed that As2O3 could inhibit the growth of Tca8113 effectively and apoptotic rate of Tca8113 cells was obviously increased in a dose and time-dependent manner. (83.40 +/- 7.31)% cells treated with 5 micromol/L As2O3 were inhibited on 72 hour point, the early apoptosis rate reached on 26.40% +/- 3.42% at that time. Moreover hTERT mRNA and protein were decreased depended on the dose and time of As2OC3, mRNA expressions of hTERT in test groups were greatly lower than that of control group on 72 hour point. CONCLUSION: It was suggested that As2O3 could significantly inhibit the growth of Tca8113 cells by inducing causing cell apoptosis and down-regulating the expression of hTERT mRNA gene and protein which might be one of its action mechanisms.

[Effects of tea polyphenols on cell proliferation and hTERT of human Tca8113 cell lines].

OBJECTIVE: To investigate the effects of tea polyphenols (TP) on cell proliferation and human telomerase reverse transcriptase (hTERT). METHODS: The experiment was divided into tea polyphenols 0.100 g/L, tea polyphenols 0.050 g/L and blank control groups. The inhibitory ratio of cell proliferation was assayed with MTT, and hTERT mRNA was detected by RT-PCR, and hTERT protein analyzed by Western-blotting. The data were analyzed by one-way ANOVA and Student-Newman-Keuls of SPSS 11.0 for windows. RESULTS: Cell proliferations were significantly inhibited after exposure to TP. The proliferation inhibiting rate of TP 0.025, 0.050, 0.100, 0.200 g/L on Tca8113 cell at 72 h was 69.75% +/- 3.24%, 63.17% +/- 3.19%, 50.35% +/- 4.21% and 34.75% +/- 3.71%, respectively (F = 270.19, P < 0.05). At the end of 72 h, the hTERT mRNA expression in TP 0.100 g/L, 0.050 g/L and control group were 0.32 +/- 0.05, 0.41 +/- 0.04, 0.72 +/- 0.05, respectively (P < 0.05). The Western-blotting assay showed that hTERT protein was also decreased by tea polyphenols compared to control group. CONCLUSIONS: Tea polyphenols could inhibit the proliferation of Tca8113 cells and expression of the hTERT mRNA and protein in Tca8113 cell lines. This effect might be one of the mechanisms for anticancer function of tea polyphenols.

Theoretical solution of a spherically isotropic hollow sphere for dynamic thermoelastic problems.

The separation of variables method was successfully used to resolve the spherically symmetric dynamic thermoelastic problem for a spherically isotropic elastic hollow sphere. Use of the integral transform can be avoided by means of this method, which is also appropriate for an arbitrary thickness hollow sphere subjected to arbitrary thermal and mechanical loads. Numerical results are presented to show the dynamic stress responses in the uniformly heated hollow spheres.

Effects of breakfast with different calorigenic amounts on blood glucose, insulin and glucagon levels.

This study was aimed to investigate the relationship between breakfast and serum glucose, insulin and glucagon concentrations in order to establish a model breakfast appropriate for Chinese. Twenty-four volunteers were randomly assigned to four study groups: high carbohydrate breakfast, high fat and protein breakfast, the typical breakfast and fasting. Each subject had serum and urine samples collected while fasting and at 1,2 and 3.5 hours following the meal. The concentration of serum glucose, insulin and glucagon was measured. The levels of serum glucose in group A, B and C differed significantly at 1 and 2 hour after meal compared to those at fasting (P<0.05). The serum glucose in group A increased insignificantly after meal. The serum insulin levels were in group A, B and C significant different compared with control group(P<0.05). Those peaked at 1 hour after meal, with group C rising the furthest. Compared with the fasting group, the serum glucagons rose and maintained the increase after breakfast in group A, B and C (P<0.05). The data suggested that various diets with different calorigenic amounts increased hormone concentration to various extents. We found that a breakfast rich in carbohydrates could maintain proper blood glucose level.
