Murine metabolic HFpEF is associated with altered mitochondrial substrate handling and S-nitrosylation remodeling.

Heart failure with preserved ejection fraction (HFpEF) is a heterogeneous condition with incompletely defined myocardial mechanisms. Here, using a two-hit murine model of cardiometabolic HFpEF induced by high-fat diet and endothelial nitric oxide synthase inhibition, we define a mitochondrial metabolic phenotype characterized by altered substrate handling, redox stress, and S-nitrosylation remodeling. While global proteomic changes were modest, metabolomic profiling revealed selective remodeling of tricarboxylic acid cycle intermediates, increased dicarboxylic acids, and altered redox-associated metabolites, consistent with mitochondrial metabolic and redox imbalance in this experimental setting. S-nitrosylation proteomics demonstrated a highly organized and bidirectional remodeling pattern affecting proteins involved in fatty acid/lipid metabolism, carbohydrate metabolism, mitochondrial energy metabolism, amino acid and organic acid metabolism, nucleotide/co-factor metabolism, and redox defense. Stable isotope tracing showed reduced glucose-derived and increased palmitate-derived acetyl-CoA in HFpEF, whereas Na-βHB reduced palmitate contribution and increased βHB-derived acetyl-CoA without restoring glucose contribution, indicating substrate redistribution and preserved ketone oxidation. Na-βHB supplementation increased oligomycin-sensitive respiration in freshly prepared left ventricular tissue, partially normalized selected TCA-cycle intermediates, reduced mitochondrial ROS and the NADH/NAD+ ratio, restored the GSH/GSSG ratio, and improved diastolic function without altering ejection fraction. Together, these findings define a redox-sensitive mitochondrial metabolic state in the HFD/l-NAME model and identify ketone supplementation as a partial metabolic rescue strategy in this context. At the same time, these findings highlight an important limitation of the murine HFD/l-NAME model, which should be interpreted as an experimental system for studying high-fat-induced cardiometabolic stress rather than as a metabolic equivalent of human HFpEF.

A modified algorithm for the improvement of composite interval mapping.

Composite interval mapping (CIM) is the most commonly used method for mapping quantitative trait loci (QTL) with populations derived from biparental crosses. However, the algorithm implemented in the popular QTL Cartographer software may not completely ensure all its advantageous properties. In addition, different background marker selection methods may give very different mapping results, and the nature of the preferred method is not clear. A modified algorithm called inclusive composite interval mapping (ICIM) is proposed in this article. In ICIM, marker selection is conducted only once through stepwise regression by considering all marker information simultaneously, and the phenotypic values are then adjusted by all markers retained in the regression equation except the two markers flanking the current mapping interval. The adjusted phenotypic values are finally used in interval mapping (IM). The modified algorithm has a simpler form than that used in CIM, but a faster convergence speed. ICIM retains all advantages of CIM over IM and avoids the possible increase of sampling variance and the complicated background marker selection process in CIM. Extensive simulations using two genomes and various genetic models indicated that ICIM has increased detection power, a reduced false detection rate, and less biased estimates of QTL effects.

14-3-3gamma affects dynamics and integrity of glial filaments by binding to phosphorylated GFAP.

Recent findings indicated a protective role of GFAP in ischemic brain, injured spinal cord, and in neurodegenerative disease. We previously demonstrated that 14-3-3gamma, once thought to be neuronal specific, was up-regulated by ischemia in astrocytes and may play a specific protective role in astrocytes. Here we report that 14-3-3gamma associates with both soluble and filamentous GFAP in a phosphorylation- and cell-cycle-dependent manner in primary cultured astrocytes. The amount of association increases during G2/M phase due to more phosphorylated GFAP. Moreover, this interaction is independent of vimentin, another type III intermediate filament protein in astrocytes which forms glial filaments with GFAP. A series of domain deletion mutants and substitution mutations at phosphorylation sites (from serine to alanine) on GFAP demonstrated that serine 8 in the head domain is essential for the direct association of GFAP to 14-3-3gamma. Overexpression of 14-3-3gamma destroyed the integrity and affected the movement of GFAP intermediate filaments. This data demonstrates that 14-3-3gamma contributes to the regulation of dynamics of GFAP filaments, which may contribute to the stability of the cytoskeleton and the mechanisms of central nervous system neurodegenerative disease.

Evaluation of binding selectivity of a polyamide probe to single base-pair different DNA in A.T-rich region by electrospray ionization mass spectrometry.

In this study, electrospray ionization mass spectrometry (ESI-MS) was used for the evaluation of the binding selectivity of a polyamide probe to single-base pair different DNA in an A.T-rich region. In this procedure, DeltaIr(dsn) was introduced as a parameter to compare the binding affinities of the polyamides with the duplex DNA. The results show that ESI-MS is a very useful tool for analysis of binding selectivity of a polyamide probe to single-base pair different DNA.

Aluminum and copper in drinking water enhance inflammatory or oxidative events specifically in the brain.

Inflammatory and oxidative events are up-regulated in the brain of AD patients. It has been reported that in animal models of AD, exposure to aluminum (Al) or copper (Cu) enhanced oxidative events and accumulation of amyloid beta (Abeta) peptides. The present study was designed to evaluate the effect of a 3-month exposure of mice to copper sulfate (8 microM), aluminum lactate (10 or 100 microM), or a combination of the salts. Results suggest that although Al or Cu may independently initiate inflammatory or oxidative events, they may function cooperatively to increase APP levels.

Epitaxial crystallization of isotactic poly(methyl methacrylate) on highly oriented polyethylene.

The annealing behavior of amorphous i-PMMA thin films on highly oriented HDPE substrates was studied by transmission infrared spectroscopy and electron diffraction. The i-PMMA thin film on highly oriented HDPE exhibits a much faster crystallization rate than usual, providing not only a good method for the preparation of crystalline i-PMMA thin and ultrathin film, but also the convenience to observe the crystallization process by infrared spectroscopy in situ. The overall crystallization kinetics of the i-PMMA thin film on the highly oriented HDPE layer was also explored in this work, and an Avrami exponent of about 2 was obtained. The accelerated crystallization behavior indicates a special interaction between HDPE and i-PMMA, which favors the nucleation and crystallization of i-PMMA. This special interaction leads also to an oriented alignment of i-PMMA on the HDPE substrate with both polymer chains parallel, i.e., the occurrence of heteroepitaxy, which could be verified by the polarized infrared spectra and electron diffraction pattern. Electron diffraction analysis further demonstrated that the contact planes of this epitaxial system are (100) lattice planes of both polymers. This can be explained in terms of a two-dimensional lattice matching.

Investigation of non-covalent complexes of HIV-1 promoter DNA and polyamides containing N-methylpyrrole by electrospray ionization mass spectrometry.

Eight novel polyamides containing N-methylpyrrole were designed to target the sequence (5'-CTGCATATAAGCAG-3'/5'-CTGCTTATATGCAG-3') of the TATA box element of the HIV-1 promoter DNA. The non-covalent complexes of the promoter DNA and the polyamides were investigated by electrospray ionization (ESI) mass spectrometry, which provided strong evidence for the binding of the novel polyamides to the sequence of the TATA box element. It also revealed that polyamide 2 (PyPyPyPybetaDp), a potent binder of HIV-1 promoter DNA and a lead molecule for the design of new anti-HIV-1 drugs, had the highest binding affinity with the TATA box element DNA among these polyamides by examining the stoichiometry and the selectivity.

Investigation of noncovalent complexes between beta-cyclodextrin and polyamide acids containing N-methylpyrrole and N-methylimidazole by electrospray ionization mass spectrometry.

Electrospray ionization (ESI) mass spectrometry was utilized to investigate noncovalent complexes between beta-cyclodextrin (beta-CD) and five novel polyamide acids containing N-methylpyrrole and N-methylimidazole. The 1:1 binding mode was specified by examining the binding stoichiometry from ESI mass spectra. It found that polyamide acids with beta-CD have binding affinities in the order: ImImImbetaCOOH > ImPyImbetaCOOH > ImPyPybetaCOOH > PyPyPybetaCOOH > NO(2)PyPyPybetaCOOH. The method gives, simultaneously, the binding constants between beta-CD and polyamide acids based on a novel linear equation.

On the alpha --> beta transition of carbon-coated highly oriented PVDF ultrathin film induced by melt recrystallization.

Poly(vinylidene fluoride) (PVDF) is one of the polymers which exhibit pronounced polymorphic crystalline forms, depending on crystallization conditions. Four different crystalline modifications, i.e., alpha, beta, gamma, and delta, have been reported so far. Among them, even though the alpha-form is the most common one, the beta-phase is the one that has attracted the widest interest due to its extensive piezo- and pyroelectric applications. During the past few decades, a substantial amount of work has been done in attempts to characterize these crystal modifications and transformations among them. It was well documented that the alpha-form PVDF can be easily obtained through melt crystallization of the PVDF at atmospheric pressure. Its beta-counterpart can, however, only be directly obtained by growth from solution, molecular epitaxy on the surface of potassium bromide, melt crystallization at high pressures, or by applying with a strong electric field. Transformation from its alpha-phase to beta-phase has been achieved by mechanical deformation, while the retransformation could be conducted through melt recrystallization at atmospheric pressure. In the present work, the recrystallization behavior of carbon-coated melt-drawn oriented PVDF ultrathin films at atmospheric pressure was studied by means of transmission electron microscopy and electron diffraction. The results indicate that through vacuum evaporating a thin carbon layer on the surface of highly oriented alpha-PVDF ultrathin film, not only has its high orientation been preserved after a complete melting and recrystallization process, but an alpha --> beta transition of PVDF has also been achieved through melt recrystallization at atmospheric pressure. This technique can be successfully used for preparing highly oriented beta-PVDF ultrathin films, especially patterned microstructures of PVDF with its highly oriented polar beta-phase and nonoriented nonpolar alpha-phase.
