Heterozygous insertions alter crossover distribution but allow crossover interference in Caenorhabditis elegans.

The normal distribution of crossover events on meiotic bivalents depends on homolog recognition, alignment, and interference. We developed a method for precisely locating all crossovers on Caenorhabditis elegans chromosomes and demonstrated that wild-type animals have essentially complete interference, with each bivalent receiving one and only one crossover. A physical break in one homolog has previously been shown to disrupt interference, suggesting that some aspect of bivalent structure is required for interference. We measured the distribution of crossovers in animals heterozygous for a large insertion to determine whether a break in sequence homology would have the same effect as a physical break. Insertions disrupt crossing over locally. However, every bivalent still experiences essentially one and only one crossover, suggesting that interference can act across a large gap in homology. Although insertions did not affect crossover number, they did have an effect on crossover distribution. Crossing over was consistently higher on the side of the chromosome bearing the homolog recognition region and lower on the other side of the chromosome. We suggest that nonhomologous sequences cause heterosynapsis, which disrupts crossovers along the distal chromosome, even when those regions contain sequences that could otherwise align. However, because crossovers are not completely eliminated distal to insertions, we propose that alignment can be reestablished after a megabase-scale gap in sequence homology.

Cauda equina syndrome secondary to constipation: an uncommon occurrence.

Suspected spinal cord or cauda equina pathology is an emergency that must be dealt with swiftly, with appropriate referral and investigations. Constipation is a clinical diagnosis of exclusion, but should not be forgotten as a cause of major morbidity when severe in nature. It can mimic an acute abdomen, sepsis and even spinal cord pathology. We describe a case of cauda equina syndrome in a young man. The pathology responsible was found to be constipation with faecal impaction, once nerve and bony pathology were excluded. When treated, there was almost immediate resolution of the neurologic signs and urinary retention.

When you assume...the reality of implementing a legally mandated substance abuse treatment program.

In 2000, the voters of California approved the Substance Abuse and Crime Prevention Act (SACPA), mandating that substance-abusing offenders who met the eligibility criteria receive treatment and not jail time. In preparation for implementation, the Santa Clara County Department of Alcohol and Drug Services Research Institute worked to identify the number and type of clients who would be eligible to receive treatment. The purpose of this article is to analyze the differences between a profile of SACPA-eligible clients that was based on pre-SACPA implementation clients and assumptions that were made concerning eligibility for the program and the reality of post-SACPA implementation clients. This article examines causes and consequences of the discrepancies between policy and implementation in the area of substance abuse treatment.

Development of an algorithm for an EEG-based driver fatigue countermeasure.

PROBLEM: Fatigue affects a driver's ability to proceed safely. Driver-related fatigue and/or sleepiness are a significant cause of traffic accidents, which makes this an area of great socioeconomic concern. Monitoring physiological signals while driving provides the possibility of detecting and warning of fatigue. The aim of this paper is to describe an EEG-based fatigue countermeasure algorithm and to report its reliability. METHOD: Changes in all major EEG bands during fatigue were used to develop the algorithm for detecting different levels of fatigue. RESULTS: The software was shown to be capable of detecting fatigue accurately in 10 subjects tested. The percentage of time the subjects were detected to be in a fatigue state was significantly different than the alert phase (P<.01). DISCUSSION: This is the first countermeasure software described that has shown to detect fatigue based on EEG changes in all frequency bands. Field research is required to evaluate the fatigue software in order to produce a robust and reliable fatigue countermeasure system. IMPACT ON INDUSTRY: The development of the fatigue countermeasure algorithm forms the basis of a future fatigue countermeasure device. Implementation of electronic devices for fatigue detection is crucial for reducing fatigue-related road accidents and their associated costs.
