Expression of argyrophilic proteins in the nucleolar organizer regions of human thymocytes and thymic epitheliocytes under conditions of coculturing with vilon and epithalon peptides.

Vilon stimulated and Epithalon suppressed the expression of argyrophilic proteins in nucleolar organizer regions of thymocytes and epithelial cells, stimulating or reducing, respectively, the formation, assembly, and transport of ribosomes into the cytoplasm and thus determining the intensity of protein synthesis in these cells. A direct mitogenic effect of Vilon was also revealed: this peptide promoted thymocyte transformation into proliferating blast cells.

[Ultrastructural criteria of clinical prognosis in hormonally non-active adrenocortical tumors].

The investigation was concerned with histological and ultrastructural features of adrenocortical tumors of the adrenals, which have a differential-diagnostic and clinico-prognostic relevance. Histological and electron-microscopical examination of 60 tumors (adrenocortical adenoma--12; adrenocortical cancer--48) was carried out and the findings were compared with clinical data. No significant correlation between histological pattern of adrenocortical tumors and survival was established. Ultrastructural evidence showed electron-microscopical examination to be a reliable procedure for making prognosis of adrenocortical cancers. There was a direct correlation between the level of differentiated cells and degree of their differentiation, on the one hand, and the quality of prognosis and survival, on the other. The significant predominance of differentiated cells was characteristic of adrenocortical adenomas.

Ultrastructural criteria of malignancy of adrenal medullary tumor.

Histological analysis and electron microscopy of 12 benign pheochromocytomas and 9 malignant pheochromoblastomas showed that there are no reliable histological differences between cells of benign and malignant tumor of the adrenal medulla. The ratio of ultrastructurally differentiated and undifferentiated cells in the tumor can reflect their maturity; the more ultrastructurally undifferentiated and less differentiated cells in the tumor, the higher is the malignant potential of this tumor.

[Sarcoma arising from Langerhans cellsĭ].

A rare case of tumor arising from Langerhans cells in the tongue and neck area in a 37 year-old man is presented. It was a polymorphocellular sarcoma with bean-like twisted nuclei. Electron microscopy identified granules of Langerhans (Bierbeck), multiple tubulo-vesicular structures, ring-like plates, Golgi apparatus, lysosomes and dendritic processes. Total leukocytic antigen was assayed in tumor cells but no expression of S-100 protein found.

[Malignant non-Hodgkin's lymphomas forming from the germinal-center cells of the lymphoid follicles in baboons from a high-risk stock].

Histological, cytochemical and ultrastructural investigation of immunologically typed B-cell non-Hodgkin's malignant lymphomas (NHL) of primates (model system on baboons) revealed 15 cases of malignant lymphomas originating from germinal centre cells of lymph nodes follicles. By the tumour cell type centroblastic (CB), centroblastic/centrocytic (CB/CC) and centrocytic (CB), malignant lymphomas were distinguished (according to Kiel classification). In case of CB NHL, tumours, as a rule, are of nodular type. Tumours, in which centrocytic infiltration predominates, are characterized by diffuse type of growth in lymphoid and nonlymphoid organs. Generalized process affects mainly lymph nodes and to considerably lower degree involves spleen and nonlymphoid parenchymatous organs.

[B-cell and T-cell immunoblastic lymphomas of baboons].

The results of morphological study of B- and T-cellular immunoblast baboon lymphomas are presented. Differences are found both in the structure of tumour cells and in the histostructure of tumours as a whole. The tumor cells of B-immunoblastic lymphomas expressed mainly IgM. The tumour cells of certain T-immunoblastic lymphomas being investigated by means of monoclonal OKT4 antibodies had a surface phenotype peculiar to T-helper cells.

[Leukemogenesis and bone marrow hematopoietic disorders in baboons with immunoblastic malignant lymphomas of B- and T-cell types].

Leukemization in baboon B- and T-cell immunoblastomas was investigated. Immunologic identification of tumor was carried out using standard immunological and cytochemical tests, e. g. identification of several types of E-rosette forming lymphocytes, surface and cytoplasmic immunoglobulin-carrying lymphocytes and assay of such acid hydrolases as acid phosphatase, acid alpha-naphthyl acetate esterase and beta-glucuronidase in lymphoid cells. Leukemization of immunoblastomas with bone marrow infiltration by tumor immunoblasts and the appearance of the latter in peripheral blood were observed rather frequently--in 7 of 10 cases of B-cell and in all 4 cases of T-cell immunoblastoma. Immunoblastoma progression was accompanied by a rise in lymphoid element level in peripheral blood and a lesser increase in bone marrow. At terminal stage, both patterns of immunoblastoma sometimes involved the development of leukemoid reactions of a myeloid type and disturbances in bone marrow hematopoiesis which manifested themselves by mild anemia and a slightly pronounced failure to produce platelets.

[Ultrastructural characteristics of malignant non-Hodgkin's lymphoma in baboons].

The ultrastructure of 29 malignant non-Hodgkin's lymphomas of baboons including prolymphocytic lymphoplasmacytic, lymphoblastic, and immunoblastic malignant lymphomas is described. The ultrastructure of the cells of B-cell malignant lymphomas is characterized by a more developed strands system of rough endoplasmic reticulum in the tumor cell cytoplasm and the presence of structural abnormalities in the protein synthesis in some B-cell malignant lymphoma cells in the form of Russell's and Dutcher bodies. Analogous baboon and human tumors were shown to have a high degree of cell ultrastructure similarity. Rare forms of baboon malignant non-Hodgkin's lymphomas including a large-cell variant of T-cell lymphoblastic lymphoma with large multilobated nuclei and a variety of B-cell lymphoma with an intermediate degree of cell differentiation have been found. Malignant lymphoma of large multilobated T-cell type and lymphoma of the intermediate lymphocytic type occur rarely in human pathology too.

The establishment of the suspension Epstein-Barr virus producing cell lines from patients with tumoral diseases.

Isolation of the Epstein-Barr virus (EBV) in suspension lymphoblastoid cell lines from human patients with tumor diseases, mainly malignant lymphoma, has been described. It has been shown that the EBV was isolated from human patients with myeloid type of leukemia in 75% of cases. A similar virus was also isolated from patients with Hodgkin's disease and leukemoid reaction of the myeloid type for lung cancer. Morphological, cytochemical, immunological, and cytogenetic characteristics of the cell lines in which the EBV is replicated have been investigated.

[Intrauterine transmission of induced hemoblastosis in monkeys, associated with type-C oncornavirus].

The possibility of intrauterine transmission of hemoblastosis was investigated in a group of 118 M. arctoides females with induced viral hemoblastosis. In the course of observations pregnancy occurred in 9 cases. Four of seven babies, delivered at term have developed hemoblastosis. The main signs of the disease were revealed in babies from the first days of their life. Hemoblastosis was accompanied with the appearance of a specific surface antigen and the presence of the "C" type oncornavirus in blood and organs of the babies, and also in cells of one embryo. Viral hemoblastosis was successfully passaged from one of the babies to three juvenile monkeys of the same species.

[Dipeptidyl aminopeptidase-IV as a histochemical marker of differential diagnosis of large cell anaplastic CD30+-lymphoma and Hodgkin's disease].

Using histochemical methods, we studied distribution of dipeptidylaminopeptidase-IV (DPP-IV) in tumor cells of 16 patients with non-Hodgkin's malignant lymphomas (NHL) including B-cell NHL (10 cases), pleomorphic T-cell lymphoma (1 case), CD30+ anaplastic large cell lymphoma (ALCL) of T-cell (1 case) and ALCL of null-cell type (4 cases) and of 13 patients with Hodgkin's disease (HD). The results indicate that tumour cells of pleomorphic T-cell NHL and ALCL of T- and null-cell type showed DPP-IV activity. In contrast, no DPP-IV activity was seen in the tumor cells of B-cell NHL (lymphocytic, centroblastic/centrocytic, centroblastic, immunoblastic), in Berezovsky-Reed-Sternberg and Hodgkin's cells of different HD variants. These results demonstrate that difference in DPP-IV activity between tumor cells of ALCL and HD may be diagnostically important for separation of ALCL from HD and moreover may be used in verification of the borderline between HD-like ALCL and ALCL-like HD. It is possible that DPP-IV activity contributes to pathogenesis of ALCL and may determine clinical behaviour of this NHL being involved in autocrine and paracrine regulation of tumor cell growth of ALCL.

[Significance of argyrophilic proteins of the nucleolar organizer region in differentiation of benign and malignant growth of thyroid epithelial tumors].

Argyrophilic proteins of nucleolar organizers (NO) were measured in 47 benign (24 follicular and 8 atypical adenomas) and malignant (8 papillary and 7 follicular carcinomas) tumors. The content of argyrophilic NO proteins was significantly higher in malignant cells. The proportion of cells with at least 5 silver grains per cell was 16.8 and 22.8% in follicular and papillary carcinoma, respectively, and 3.2 and 12% in follicular and atypical adenoma, respectively. These data can be used as accessory signs in differential diagnosis.

[Expression of argyrophilic proteins from the nucleolar organizer regions as an index of maturity degree of benign and malignant adrenal tumors].

Expression of APNOR was studied in 26 benign and malignant tumors of adrenal cortex and medulla. The histochemical method with silver nitrate was used. Expression of APNOR argyrophilic proteins in adrenocortical carcinomas was 4.59 times higher than in adrenocortical adenomas and 2.63 times higher in pheochromoblastomas than in pheochromocytomas. This index may be recommended as an additional method for differential diagnosis of benign and malignant adrenal tumors, particularly in difficult border line cases as well as determination of prognosis and metastatic potential of these tumors. Cytospecificity of APNOR expression in different types of adrenal cells was established.
