[Obtaining the allergens from yeast-like fungi of the genus Candida, producers of fodder protein, for hygienic standardization of the strains].

Separated were surface antigens' complex preparations from the native biomass of Candida yeast-like fungi, by using the beta-mercaptoethanol-induced undestructive extraction technique. The preparations were analysed by the highly effective liquid chromatography technique, double immunodiffusion in agar, immunoelectrophoresis, and in experiments on intradermal titration on intact and sensibilized guinea-pigs. It was indicated that the complex preparations of surface antigens extracted from C. maltosa, C. valida and C. scotti were characterized by low reactogenicity and high precision in experimental intracutaneous allergotesting on guinea-pigs. The immunochemical and allergenous activity was higher in preparations with molecular masses of 600 kD. These complex preparations can be used as allergens in the hygienic forming of fodder protein producent strains.

[Isolation of antigen-active biopolymers from Candida maltosa culture fluid for obtaining allergens of diagnostic value for examination of industrial microbiology workers].

The membrane filtration technique and column gel chromatography+ (with sepharose 6B or sephacryl S-200) were used to receive fractions of KJ Candida maltosa, strain VSB 899, which were studied with highly effective liquid chromatography, immunochemical reactions and intracutaneous allergotesting on guinea-pigs sensibilized with VSB 899 live cultures. In the whole KJ strain VSB 899 concentrate, 8 components were identified, six of which had molecular masses from 350,000 to 18,000 and had no analogue in the surface antigen preparation (SAP) from the same strain of yeast-like fungi. It was shown that the SAP-related antigen determinants were present in predominantly polysaccharide-containing fractions. In intracutaneous allergotesting, chromatographic fractions with molecular masses of 50,000 and 18,000-35,000 were most manifesting. It was supposed that still further purification of the allergen will increase its specificity.

[An evaluation of the degree of serological affinity of the surface structures in Candida maltosa and Candida albicans].

The complex preparation of surface antigens was obtained by the treatment of C. maltosa whole cells with beta-mercaptoethanol and their separation into 8 fractions by means of ion exchange chromatography on DEAE cellulose. The sensitizing capacity of these fractions was studied in the allergic dermal test on guinea pigs and their immunochemical activity, in the immunodiffusion test with homologous antiserum and with the gamma-globulin fraction of antiserum to C. albicans. All fractions induced delayed hypersensitivity, more or less intensive, in guinea pigs. The agar immunodiffusion test revealed that the complex preparation contained two groups of fractions differing in their antigenic composition. Fractions of group 1 reacted equally well with homologous and heterologous antisera. Fractions of group 2, eluting at NaCl concentrations from 0.1M to 0.4M and having very high precipitation activity in reactions with homologous antiserum, showed considerably lower capacity for reaction with antiserum to C. albicans, which suggested that they contained antigenic structures differing from the antigenic determinants of C. albicans and thus ensuring specific reactions in cases of candidal sensitization induced by C. maltosa.

[Effect of blastolysin on the ultrastructure and functional activity of mast cells and eosinophils].

Blastolysin is shown to cause degranulation of mast cells, resulting in a reduction of their histamine content and in their release of mediators that activate eosinophilic leukocytes (EL). These accelerate the deamination of the histamine released by mast cells to the extracellular medium. It is possible that blastolysin acts directly on EL granules which are largely responsible for histamine inactivation.

[Effect of blastolysin on the ultrastructure and phagocytic activity of peritoneal macrophages].

Morphology and functional activity of peritoneal macrophages were studied at different intervals after the administration of blastolysin to Wistar rats. Alteration of the macrophage ultrastructure and their functional activity proving the stimulatory effect of the preparation on cells is shown. The signs of stimulation were revealed 30 minutes after the administration of the drug, reached a maximum on the 7th and 14th days and was manifested by structural changes of the cell surface, lysosomal apparatus, mitochondria and by activation of phagocytosis.

[Immunochemical study of surface glycoconjugates of yeast-like fungus of the Candida genus].

Two complex preparations (extract I and extract II) of surface antigens were obtained from the yeast-like fungus Candida maltosa by treatment of intact cells with beta-mercaptoethylamine and pronase. Immune diffusion in agar gel revealed antigenic heterogeneity of the preparations. Both of the extracts were found to have at least 3 antigenic components. The extracts induce hypersensitivity of intact guinea pigs and candida sensitization on the 12th day after the injection of Candida maltose intact cells into the animals. The common antigen components of extracts I and II were found by means of immunochemical and chromatographic assays. The extracts preserved the antigenic activity after enzyme hydrolysis with pronase.

[Isolation of surface antigens of Candida maltosa responsible for Candida-sensitization and their immunochemical characteristics].

Surface antigens from Candida maltosa were shown to be of heterogeneous nature while common component was present in all the preparations. These antigens were able to induce both the slow type hypersensitivity and the reaction related apparently to the later phase of immediate hypersensitivity. Hypersensitivity of slow and immediate types developed in response to invasion of living culture of C. maltosa A strain. The data obtained suggest that surface antigens from Candida might be used for production of diagnostic allergens.
