Effects of intravenous injection of two different strains of Corynebacterium parvum in the mouse.

A strain of C. parvum, CN6134, known to have antitumour activity, caused thrombosis in the sites where the organism is phagocytosed. It bound to macrophages in vitro and activated the alternate pathway of complement. A strain of C. parvum, CN5888, which fails to show antitumour activity, did not show thrombosis. It did not bind to macrophages or activate guinea-pig complement. It did, however, cause marrow infarction which seems to result from a diminished clearance of the organism from the circulation.

Malignant histiocytosis: a clinico-pathological study of 12 cases.

The clinico-pathological features of 12 cases of malignant histiocytosis (histiocytic medullary reticulosis), aged 12-60 years, were studied. Special emphasis was put on the analysis of the haematological, cytological and histopathological findings of the disease. Bone marrow aspirates and biopsies from involved tissues (liver, lymph node, spleen and skin) were found best suited for establishing the diagnosis. Cytochemical and ultrastructural data support the concept that the neoplastic cells belong to the mononuclear-phagocyte system. Three untreated patients died in less than 6 months. Median survival of the nine treated patients was 1 year. Seven patients were treated with the combination chemotherapy CHOP and four achieved a complete remission. Three of them are alive and continue in unmaintained remission for more than 1 year, surviving so far for 18+ to 31+ months. Two cases showed evidence of CNS involvement, one during life and the other as a post-mortem finding. The differential diagnosis from other related or unrelated disorders and the possible aetiological factors responsible for the disease are discussed in detail.

A comparison using quantitative enzyme cytochemistry between leucocyte alkaline phosphatase levels in the bone marrow and blood of the rat.

Leucocyte alkaline phosphatase (LAP) levels have been compared in neutrophil polymorphs in bone marrow and blood in the rat, using a quantitative enzyme cytochemical technique. It has been shown that LAP levels are higher in bone marrow than blood, both in normal animals and those with experimentally-induced local inflammatory lesions. In the latter group both marrow and blood LAP levels were higher than in controls; blood levels rose because of the outpouring of cells with moderately high LAP levels into the blood and marrow levels rose because only cells with very high LAP levels remained. The inverse relationship between neutrophil age and alkaline phosphatase content, previously demonstrated in the peripheral blood of the rat, has also been shown to exist in the bone marrow.

Staining of sulphatides in metachromatic leukodystrophy with Alcian blue at high salt concentrations.

Alcian blue combines with purified sulphatide in 1.OM magnesium chloride. In tissue sections from patients with metachromatic leukodystrophy, sulphatide is stained by Alcian blue in 0.8 M magnesium chloride, and the staining can be abolished by prior treatment with chloroform and methanol. The simplicity of the technique, its specificity and ease of interpretation recommend Alcian blue staining at high salt concentrations as a routine method in the diagnosis of metachromatic leukodystrophy.

Histological changes of intravascular coagulation in mice resulting from intravenous injection of C. parvum.

A histological study was made of liver and other tissues of mice at intervals after injection of C. parvum. As early as 20 min after injection occasional thrombi surrounded by polymorphs were seen in the alveolar capillaries. A general thrombosis rapidly developed in the hepatic vessels with consequent hepatic necrosis. Thrombi also developed in splenic vessels. This intravascular coagulation was prolonged, lasting at least 7 days. There was evidence that fresh thrombi were being formed throughout this time. The thrombosis was mirrored by a fall in platelet counts. The kidney was uninvolved in the reaction. The mechanism of this reaction and its possible relevance to the antitumour action of C. parvum is discussed.
