Verrucous carcinoma of urinary bladder.

The authors have encountered a recent case of verrucous carcinoma of the urinary bladder in a seventy-three-year-old man. The difference in its clinical presentation and pathologic growth pattern from the usual squamous cell carcinoma is striking. The case report, a detailed pathologic description of the lesion, and review of the scanty literature are presented.

Presence of two forms of fumarase (fumarate hydratase E.C. 4.2.1.2) in mammalian cells: immunological characterization and genetic analysis in somatic cell hybrids. Confirmation of the assignment of a gene necessary for the enzyme expression to human chromosome 1.

Two major forms of fumarate hydratase have been resolved in extracts prepared from a wide variety of mammalian cells by electrophoresis. Fractionation experiments with human and mouse cells suggest that one form (the slower migrating) is localized in the mitochondria, whereas the other form is predominant in the cytoplasm. Analysis of the segregation of the enzyme forms in human-mouse somatic cell hybrids indicates that a gene(s) necessary for the expression of both forms can be assigned to human chromosome 1(confirmation of a previous assignment by van Someren et al., 1974). Electrophoretic analysis suggests that the two forms may be interrelated. Furthermore, they both exhibit identical reactivity toward anti-fumarate hydratase antiserum. It is suggested that a modification of one form may occur in vivo and that the modification may be important in determining the intracellular localization of the enzyme.

Differences in the products of mitochondrial protein synthesis in vivo in Human and mouse cells and their potential use as markers for the mitochondrial genome in human--mouse somatic cell hybrids.

The proteins synthesized in the mitochondria of mouse and human cells grown in tissue culture were examined by electrophoresis in polyacrylamide gels. The proteins were labelled by incubating the cells in the presence of [(35)S]methionine and an inhibitor of cytoplasmic protein synthesis (emetine or cycloheximide). A detailed comparison between the labelled products of mouse and human mitochondrial protein synthesis was made possible by developing radioautograms after exposure to slab-electrophoresis gels. Patterns obtained for different cell types of the same species were extremely similar, whereas reproducible differences were observed on comparison of the profiles obtained for mouse and human cells. Four human-mouse somatic cell hybrids were examined, and in each one only components corresponding to mouse mitochondrially synthesized proteins were detected.

Plasmacytoma of the thyroid.

Three cases of plasmacytoma of the thyroid are described and their clinical and histological features recorded. Two were considered to be true extramedullary plasmacytomas, the third the initial manifestation of a disseminated myelomatous process. Plasmacytomas of the thyroid are shown to have many features in common with malignant lymphomas in this site, including an association with Hashimoto thyroiditis in more than half the reported cases. No certain method of assessing the prognosis in any individual case has yet been described.
