[The effect of the in-vitro interaction of antibacterial preparations on Brucella melitensis].

The character of drug interaction in 18 various combinations of sisomicin, amikacin, gentamicin, cefotaxime, doxycycline, trimethoprim and nitroxaline was studied by the chess board method with respect to 25 strains of Brucella melitensis. The synergistic effect with respect to the majority of the cultures was observed with the use of the combinations of gentamicin, amikacin or sisomicin with doxycycline. The combinations of the aminoglycosides with cefotaxime and sisomicin or gentamicin with rifampicin had a summation effect on the majority of the cultures. With the use of the combinations of cefotaxime with nitroxaline or trimethoprim and rifampicin with nitroxaline the effect on the majority of the cultures was indifferent. The effect was mainly antagonistic with the use of the combinations of amikacin with rifampicin, gentamicin with trimethoprim and cefotaxime with doxycycline. The effect of the combinations of gentamicin with nitroxaline and cefotaxime with rifampicin was different with respect to different strains of B. melitensis. It was indifferent, antagonistic or summation depending on the strain.

[Sensitivity of Brucella melitensis to various antibiotics, drug preparations and their combinations].

In vitro activity of 13 antibiotics and drug preparations as well as some of their combinations was studied with respect to Brucella melitensis. It was shown that the strains of B. melitensis were sensitive to sisomicin, amikacin, gentamicin, cefotaxime, rifaprim +, nitroxoline and trimethoprim. Variations in the doxycycline sensitivity depending on the strain were observed. The strains of B. melitensis were resistant to cephalexin, phosphomycin, chinoxydine, dioxydine and oxolinic acid. Combinations of sisomicin and trimethoprim at a ratio of 1:10 and sisomicin and nitroxoline at a ratio of 1:2 had a summation effect.

[Bacteriocinogenicity of brucellae isolated in foci in the Caucasus and their evaluation from taxonomic viewpoints].

The aim of the study was to elucidate the possibility of using bacteriocinogenicity of Brucella as taxonomic feature, to determine their phylogenetic relation to other microorganisms by their bacteriocinogenic properties and to investigate the physicochemical properties of brucellacin and conditions for its stable detection. The Brucella cultures were isolated in the Caucasus. Investigation of their capacity for production of bacteriocin according to the procedure described by M.A. Konstantinova and A.D. Garmazova (1979) revealed that 62.1 per cent of the 216 cultures tested produced brucellacin. Isolation of bacteriocin with the methods developed was shown possible in all of the tested strains of B. melitensis, B. abortus, B. suis and in most of the strains of B. ovis. The methods also provided an increase in the synthesis and activity of brucellacin. The analysis of the characteristic features of bacteriocinogenicity and the properties of bacteriocin allowed recommending the use of additional taxonomic features for identification and differentiation of Brucella. Sensitivity of the indicator strains of Brucella to bacteriocins of other species (F. tularensis, Campylobacter fetus intestinalis B-8833, Y. enterocolitica, Vibrio cholerae and E. coli Fredericq) was noted which was additional evidence of the phylogenic relation between the above organisms. Investigation of the physicochemical properties of brucellacin confirmed the suggestion of the protein nature of the active principle of brucellacin and its similarity in different Brucella species.

[Taxonomic position and ecology of the causative agent of brucellosis isolated from murine rodents in regions of the northern foothills of the Greater Caucasus. II. The ecological and pathogenetic characteristics of Brucella strains isolated from murine rodents].

To establish the possibility of the circulation of brucellae among the population of myomorphous rodents, the mass survey of these rodents was carried out in the mountainous and foothill regions of the Caucasus. In the area of the northern foothills of the Caucasian Mountains 23 Brucella cultures were isolated from 2715 rodents under examination; these cultures proved to be similar to B. suis 1330 in respect to their pathogenicity and the character of pathomorphological changes induced in guinea pigs. Experiments on white mice demonstrated the possibility of brucellar contamination through sexual route and through feces.

[Comparative evaluation of the spectrum of lytic effects of bacteriophages TB, Wb, Fi, BK 2 and R on various Brucella species].

The lytic activity of Brucella phages TB, Wb, Fi, BK2 and R on various Brucella species has been studied. Phage TB has been found to lyse B. abortus, B. suis and B. neotoma, phage Fi has proved to be no different from phage TB, while phage Wb has shown higher activity in respect of B. suis; phage BK2 has been found to lyse B. abortus and, to a lesser extent, B. suis and B. melitensis; phage R has shown low activity, while splitting out phages capable of lysing B. abortus cultures in the S-form. All these phages multiply well in the corresponding indicator strains grown in Martin's broth. The Brucella cultures isolated from myomorphous rodents in the northern foothills of the Caucasus have been found to be similar to B. suis in respect to their spectrum of phage sensitivity, differing from B. suis only by forming smaller negative colonies in the presence of phage Wb.

[Brucella L-forms, their revertants and initial cultures].

The transformation of Brucella into a relatively stable bacterial L-form leads to a sharp decrease in virulence and the loss of their capacity for agglutination under the action of commercial brucellosis antiserum. When reverting from the L-form, Brucella cells partially restore their virulence and agglutinability. The pronounced restoration of these properties can be observed in revertants from the L-forms, obtained in vivo. All revertants are less virulent than the initial strains. The transformation into the L-forms is accompanied by a sharp decrease in their electrokinetic mobility, which is indicative of profound transformations of their peripheral structures leading to the exposure of the electrically inert layers of the cell wall.

[Comparative cytoenzymatic study of infectious and vaccinal processes in experimental brucellosis].

In experiments on 240 guinea-pigs metabolic changes were found to occur in peripheral blood cells in the process of the development of brucellosis infection and after immunization. The degree and character of the activity of lysosomal enzymes depended on the time of infection and immunity formation. In comparison with the vaccinal culture of Br. abortus 19, the virulent culture of Br. abortus 544 induced greater changes in the activity of esterase, acidic and alkaline phosphatase in neutrophils, in the activity of acidic phosphatase in lymphocytes, as well as in the number of lymphocytes containing this enzyme. The data on enzymatic activity are recommended for use as a differential test for evaluating the character of the infectious and vaccinal processes.

[Comparative evaluation of methods of inducing Brucella L forms and a study of their adaptibility in the body of animals].

The action of bicillin-3 on Brucella directly in the body of animals enhances their potential ability to convert into L-forms. One of the factors contributing to the generalization of the infection caused by the L-forms of Brucella and to their prolonged persistance in the body is the presence of an L-transforming agent (in this case, bicillin-3), which indicate that the appearing L-forms of Brucella are obviously antibiotic-dependent.

[Principles of epidemiologic and epizootologic research in foci of brucellosis].

Principles of epidemiological and epizootological investigations in a brucellosis focus are presented in this work. A founded determination of a focus and methodological approaches to its study and systematization of the data concerning all the foci of a definite administrative territory is given. Also approaches to the realization of out-patient observation over the population in the brucellosis foci are characterized. The author believes that only such scientifically founded and all-round approach to the study of epizootological and epidemiological regularities of the brucellosis foci existence would aid successful solution of the most important problems pertinent to reduction of brucellosis incidence among humans.

[Immunogenic properties of the cellular components of Brucella].

A study was made of immunogenic properties of the cell wall and other cellular components of brucellae obtained by mechanical disntegration of bacteria. The greatest immunogenic properties proved to be possessed by the cell walls. In using the vaccine consisting o the living vaccine of the Br. abortus strain 19 and the cell walls of brucellae the efficacy of the preparation was not reduced in experiments on guinea pigs and mice.

[Results of a study of the diagnostic qualities of brucellosis and tularemic antigenic erythrocytic diagnostica].

The authors elaborated methods of preparation of brucella and tularemia antigenic erythrocytic diagnostic agents which were characterized as highly specific, specific and stable preparations in mass examination of humans and animals at various stages of the vaccinal and infectous processes. The simplicity of obtaining specific antigens intended for the sensitization of formalinized erythrocytes and stability of the results of results of reproduction of the methods of preparation of the antigenic erythrocytic diagnostic agents offered a possibility of recommending the mentioned methods of industrial preparation of the diagnostic agents.
