Morpho-functional changes in human tendon tissue.

Insertion tissue biopsies of right arm common extensor tendons from 11 patients with chronic lateral epicondylitis were processed for light and electron microscopy. The subjects were aged between 38 and 54 years (only one was 25). The specimens showed a variety of structural changes such as biochemical and spatial alteration of collagen, hyaline degeneration, loss of tenocytes, fibrocartilage metaplasia, calcifying processes, neovascularization and vessel wall modifications. Tissue alterations were evident in limited zones of the tendon fibrocartilage in which the surgical resection was generally visible. The areas where the degenerative processes were localized, were restricted and in spatial contiguity with morphologically normal ones. The observed cases presented histological and electron microscopic findings that characterize lateral epicondylitis as a degenerative phenomenon involving all tendon components.

Giant cell tumor of tendon sheath: a light and electron microscopic study.

A neoformation has been surgically withdrawn from third finger extensor tendon of the right hand of a 52 year male subject. Light (LM) and electron microscope (EM) observations from a number of tissue fragments allowed the identification of tumor nature, which appeared a giant cell tendon sheath. Moreover, some structural patterns have been described and compared to the previously reported cases. In areas of major cell density, parenchyma does not show lobular or gland-like organization; on the other hand, wide zones characterized by an amorphous matrix, progressively replacing collagen and containing elongated cells, are present. Giant multinucleated cells, mostly localized close to collagen bundles, can also be revealed. Unexpectedly, no foam cells appear and no phagocytized cell debris can be identified in giant multinucleated cells. Engulfed crystals are, differently, evidentiated by electron microscopy, both in mono- and multinucleated cell cytoplasm. Their electron density and their localization within cytoplasmic vacuoles suggest the presence of calcium. A correlation between giant cells and osteoclasts is then proposed. Multiple variously oriented centrioles support the possible mitotic genesis of multinucleated giant cells, which never show, on the other hand, fusion features. Siderosomes and residual bodies are also present. An unusual, diffuse thickening of nuclear lamina, only interrupted at nuclear pore level, is described and discussed.

Calcific chronic lateral epicondylitis: a histological and ultrastructural study.

Fragments of insertion tissue from right arm common extensor muscle have been collected from a 25-year patient with chronic lateral epicondylitis. Specimens, processed for light (LM) and electron (EM) microscopy, evidentiated a variety of degenerative alterations, such as focal hyalinosis, lipoidosis, collagen fiber redistribution, calcifications and vascular changes. Evidence of collagen normal function maintenance and turnover have been also observed in tenocytes.

Early events of liver regeneration in rats: a multiparametric analysis.

5-Bromodeoxyuridine (BrdU), a synthetic analogue of thymidine, has been utilized in vivo to detect the proliferation which occurs in the liver after two-thirds surgical hepatectomy. Immunocytochemical detection of BrdU incorporation has been carried out at both the morphological and flow cytometrical level, while structural changes of regenerating liver have been investigated, using Mallory-Azan-stained paraffin sections, by means of an image analyser. The results obtained show that in vivo DNA synthesis progression throughout S phase follows a pattern similar to that previously described in vitro in both 3T3 fibroblasts and Friend erythroleukemia cells and also demonstrate a precise correlation between morphological patterns of BrdU incorporating cells and their lobular distribution. Moreover, the activation of at least two proliferation waves can be detected from 18 to 34 h after hepatectomy: the former, starting from adjacent regions of contiguous lobules, apparently induces an irregular increase of lobular dimension; the latter, involving both inner and peripheral lobular domains, seems to be correlated with the appearance of nodule-like structures at the lobule periphery. In view of these results the role of the hepatic acinus and the hypothesis of a streaming of parenchymal cells during liver regeneration have been discussed.

Pathology from hydroxyapatite deposits in periarticular tissues. First conclusions on etiopathogenesis.

Biopsies of periarticular tissues from patients with episodes of chronic periarthritis were examinated by histological, ultrastructural and chemico-crystallographic methods. In all samples the mineralogical observations at the optical microscope, showed aggregates of microcrystalline incrustations, whose ultrastructural morphology has been characterized by SEM. The histological observations showed necrosis of collagen fibres and microcrystallization process in cavities and in metaplastic fibrocartilage. The HA deposits in the periarticular tissues, in the next stage of the process, are surrounded and demolished by macrophages and multinucleated giant cells. At the same time granulation tissue carries out the repair process, that begins with the formation of thin parallel collagen fibers interposed with many fibroblasts and numerous small vessels.

Morphological aspects of steroid hormone action on rat hepatocyte.

A morphological study was performed on the effect of HC treatment on normal and adrenalectomized rat liver. The observation of liver fragments shows that the hormone treatment can balance only partially the hormonal deficiency caused by ADX, resulting in particular changes of both hepatic architecture and fine ultrastructural organization.

Scanning electron microscopy, chemical and histologic analysis in human periarthritic shoulder biopsy.

In the present study biopsies were analyzed, taken from the left shoulder of a patient who, according to the radiological diagnosis, was suffering from calcific periarthritis. In both optical and electronic microscopy the mineralogical observations showed crystalline aggregations, while the chemical analysis, carried out with an Edax EDS spectrophotometer, confirmed the presence of Ca and P in them, in the ratio typical of biological apatites. The histological observations clearly indicate a change in the metabolism of the tissues present. In the samples observed the damage to the muscular tissues is easily seen while the connective tissue, though apparently less compromised, shows the presence of numerous calcifications whose damage cannot be revealed solely through histologic observations. The authors, linking the structural histologic alterations observed to the presence of hydroxyapatite granules as well as to the patient's painful symptoms, believe all these observations are the result of a chronic process.

Further observations on pineal brain sand formation and evolution in man.

Acervuli obtained from fragments of the human pineal glands of subjects of both sexes and age ranging from 23 to 87 years were analyzed by light microscopy after histochemical stains and by EDS-microanalysis. It was found that the sub-units and acervuli are positive to P.A.S., Alcian Blue pH 2.5, Gomori-Bargmann procedures for the presence of proteins, glycoproteins, proteoglycans and of the neurosecretory material in different layers of the sub-units and acervuli. We suppose that the increase of Mg++ in the sub-unit from the core to the periphery is responsible for the inhibition of following hydroxyapatite deposition and for growth. We suggest that the presence of glycoproteins and proteoglycans can represent the aggregation factor for bindings between disulphide bonds and calcium.

Addiction time and value of Z indicators in Rorschachs of heroin users.

Rorschach test scores for short-term (1 to 3 yr.) and long-term (5 or more yr.) heroin users (ns = 25 each) were compared, to ascertain whether the use of heroin influences the organizational activity, namely, the form level of each response (as measured by Zf and Zd indices). Zd values were significantly lower in long-term than in short-term users, while Zf values approached significance. These findings support the hypothesis that cognitive functioning is impaired along with addiction time.

A preliminary study of human pineal gland concretions: structural and chemical analysis.

Acervuli and fragments of pineal gland obtained from 33 subjects of both sexes and age ranging from 1 to 87 years, (30 autopsy and 3 biopsy specimens) were analyzed by light microscopy, transmission and scanning electron microscopy, X-ray diffraction and X-ray energy dispersive microanalysis. It was found that primary mineralization occurs in an organic matrix formed by pinealocytes and that hydroxyapatite also takes place in mineral deposition. From our analysis, the formation of acervuli appears to be age and sex independent and can be possibly related to the secretory activity of the gland.

Bone mineral alterations and Mg content in aging.

The authors propose to determine whether the quantity of bone mass reduction linked to aging is accompanied by qualitative modifications of the mineral structure. To this end, 18 samples of cancellous bone from the femoral heads of two groups of patients (Groups A & B), were examined. Group A was made up of 8 old osteopenic patients suffering from fracture of the femur neck (age 62-84). Group B consisted of 10 young non-osteopenic subjects (age 34-53). Through chemical analysis a statistically significant percentage increase in Mg++ was noted in Group A and, by X-Ray diffraction, significant presence of beta-TCP. Mineral structure alterations occur in the process of aging of the skeletal tissue.

Histochemical and ultrastructural study of the human pineal gland in the course of aging.

A histochemical and ultrastructural study was carried out on the pineal gland of 2 day- to 86-year-old subjects. Specimens were obtained by stereotaxis in the course of neurosurgery; other specimens were autoptical pineal fragments. Histochemical and ultrastructural analyses showed lipofuscin pigments, frequently localized in the proximity of blood capillaries and of cytoplasmic microacervuli. The brain sand amount does not appear to be age-related because microacervuli are barely present or frequently absent in the oldest subjects. Its involvement in the secretory activity rather than in gland atrophy is also suggested. The close relationship between cytoplasmic microacervuli and intermediate filaments suggests a possible role of the cytoskeleton in the formation of the brain sand.

Effect of water irrigations on human colonic mucosa structure after sigmoidostomy.

The colonic mucosa of patients with sigmoidostomies, who were operated on for rectal cancer, and treated thereafter for different periods of time with daily water irrigations to obtain complete evacuation, was investigated by histologic and histochemical methods. Microscopic examination of the test specimens showed that the epithelial continuity, the characteristic brush border, and the positive mucous reaction to PAS and Alcian blue-Alcian yellow methods were not significantly changed with respect to controls. In some test specimens a remarkable number of mitoses were evident in the crypts. Since this could result from water irrigation stimuli and/or might represent an early manifestation of a restored carcinogenetic process, specific investigations were performed on the 0-acylated sialic acids of the colonic mucins, which have been reported to represent markers of early malignant changes in colorectal epithelial cells. The results did not reveal alterations of the sialomucins in the treated specimens with respect to controls.
