Effects of secretory products of activated neutrophils on morphological composition and functional activity of peritoneal exudation cells during inflammation of staphylococcal origin.

Substance A5 isolated from supernatants of activated neutrophils from donors significantly increases the percentage of neutrophils and macrophages in the peritoneal exudation of mice on days 3 and 7 of staphylococcal inflammation and stimulates functional activity (lysosomal, phagocytic, and NBT-reducing) of these cells, reduced as a result of inflammation, on days 3, 7, and 14 of the inflammatory process.

[The role of neutrophils in the regulation of antimicrobial resistance].

The functional activity of neutrophils and macrophages and an immune response to sheep red blood cells decrease in mice with local staphylococcal infection in the early period. In the first 24 hours after contamination, the animals' neutrophils lose their capacity to produce immuno-stimulating agents, they begin secreting immunosuppressive factors. Administration of secretory products of activated neutrophils, the low-weight molecular peptide isolated from these cells to the induced animals restores the immunological responsiveness of the staphylococcus-infected animals, reduces the death of mice infected with S. aureus and S. typhimurium.

[Role of neutrophils in regulation of immune responsiveness and reparative reactions of damaged tissue].

After both thermal and mechanical injuries, mice are shown to have depressed functions of phagocytes (primarily neutrophils), suppressed ability of an immune response to antigens and decreased skin inflammatory and reparative activities. It is suggested that a neutrophilic secretory defect and a decrease in the production of stimulating factors might be a cause of the above effects. After thermal and mechanical injuries, injection of neutrophilokin extracted from latex-activated neutrophilic supernatant into the mice promoted recovered phagocytic functional activity, higher humoral and cellular immune response and increased the rate of standard burn wound healing. Neutrophilokin also accelerated clavus formation in mice with mechanical injury. Thus, it can be concluded that neutrophils and their low-molecular secretory products play an important role in regulating immune and inflammatory reparative homeostasis in thermal and mechanical injuries.

[The effect of the peptide fractions of neutrophilokines on the immune response of mice infected with Staphylococcus aureus].

The influence of immunostimulating and immunosuppressing neutrophilokine fractions on the immune response of mice with staphylococcal infection has been studied. When injected into mice with staphylococcal infection, the immunostimulating fraction of the supernatants of activated neutrophils has been found to restore the activity of the immune system of the infected animals. The secretory products of intact neutrophils do not change or decrease the capacity of the animals for immune response under the conditions of the infectious process.

[Study of latex-induced luminol-dependent chemiluminescence of neutrophils].

The effects of various polystyrene monodispersed latexes on neutrophil chemiluminescence (CL) induction have been studied. The studies have revealed that latexes may be employed as neutrophil CL activators. Latexes with a diameter of 1.7 micron could induce CL of the highest intensity. An increase or decrease in the diameter of latex particles reduced the intensity of neutrophil CL induction. Gamma-globulin or immunoglobulin coating of latex lowered, whereas zinc methacrylate coating enhanced the intensity of the reaction. Since latexes induce a maximum respiratory outburst in the first two minutes after their introduction in cellular suspension, they may be referred to as rapid inductors of neutrophil CL.

[Changes in the immunotropic activity of neutrophilokins in the course of experimental staphylococcal infection].

The influence of the products secreted by activated neutrophils (neutrophilokins) of mice, both intact and infected with staphylococci, on the activity of mouse spleen cells in the graft-versus-host reaction, immune response to sheep red blood cells and the antigen-presenting function of peritoneal macrophages was studied. Neutrophilokins of intact mice stimulated the activity of immunocompetent cells. Neutrophilokins obtained from infected mice on day 3 after infection produced an immunosuppressing effect. On day 7 after infection the immunostimulating activity of neutrophils was restored and showed practically no difference from the normal level.

[Specific reaction of neutrophils to bacterial chemoattractants].

The influence of bacteria and supernatants, obtained from the cultures of neutrophils preincubated with these bacteria, on the chemotaxis of neutrophils has been studied. Neutrophils preincubated with bacteria acquire the capacity for specific chemotactic deactivation in response to the contact with the culture of the same bacteria. During the phagocytosis of bacteria neutrophils secrete factors selectively attracting only those neutrophils which have been activated by bacteria of the same species.

[Effect of neutrophilokines on the immune response in mice].

The influence of different peptide fractions obtained from the intact and latex-stimulated neutrophils on the immune response was studied. It was demonstrated that neutrophils after stimulation synthesize the factors activating immune response, the intact neutrophils synthesize the suppressor factors of peptide nature.

[Functional activity of T- and B-lymphocytes in thermal burns].

The functional activity of B and T (helpers and suppressors) lymphocytes was studied in mice with thermal injury covering an area of 10 and 30% of the body surface. Following 10% burn the activity of B lymphocytes and T helpers increased while that of T suppressors dropped down. Insufficiency of T lymphocyte function developed after vast injury. At the same time B cells maintained their ability to form antibodies against sheep red blood cells.

[Proliferative reaction in the lymph node of recipients after the transplantation of the splenic cells of burned syngeneic donors].

Thermal burn was shown to induce in the splenocyte population in mice the appearance of cells capable of producing proliferative reaction in the regional lymph node. Spleen cells taken on days 1--2 after a burn were found to be most active. A direct relation between the number of transplanted cells and marked proliferation process in the lymph node was established. The proliferative process was mainly the consequence of the proliferation of donor cells. The process resulted, probably, either from the stimulation of cell division under the influence of proliferation factors, or from the activization of autoreactive lymphocyte clones.

[Immune response to ram erythrocytes and the cellular state of the monocytic phagocytosing system in the early period of a thermal injury].

The degree of immune response to corpuscular antigen and macrophage reaction within 24 hours after thermal injury were examined in a comparative study. The number of antibody-producing cells, antigen engulfment and elimination by the cells of peritoneal exudate, their adhesiveness to glass, and cathepsin activity in the cells of peritoneal exudate and the spleen were determined. Mice subjected to thermal burns showed suppressed immune response and decreased functional activity of the cells of the monocytic phagocyte system within the first 24 hours, which was manifested by a decrease in the adhesiveness of these cells to glass, and their ability to engulf and destroy xenogenic erythrocytes.
