A loss of GABAergic hippocampus innervation in rats with cobalt-induced epilepsy demonstrated by Wolman's fluorescence method.

The topohistochemistry of gamma-aminobutyric acid (GABA) was studied in the hippocampus of rats with epileptiform activity provoked by cortical implantation of cobalt-agar pellets and in control rats with implanted agar-pellets. Using the GABA specific fluorochroming procedure of Wolman (1971) a distinct decrease of GABA fluorescence was found especially in the pyramidal and granular layer, but only in the animals with a typical epileptiform activity of the EEG. The results support the hypothesis that there is a lack of GABAergic inhibition in epileptic individuals, but the anticonvulsive effect of GABA depends on structural and functional relations in the specific type of seizure.

[Role of the mesencephalon reticular formation in the effect of ethosuximide on a cortical epileptogenic focus].

Ethosuximide provoked in encéphale isolé cats a dose-dependent suppression or facilitation of strychnine focus spiking activity, and in combination with high frequency stimulation of formatio reticularis mesencephali a significant activation of the cortical focus. The results seem to show that the influence of ethosuximide on the formatio reticularis mesencephali is one--if not the main--component of the drug effect.

[Photically released trains of potentials following local application of ethosuximide into the corpus geniculatum laterale].

The influence of ethosuximide injected into the lateral geniculate body (LGB) on photically evoked afterdischarges (PhAD) and recruiting responses (PhR) after 0,3/s or 7/s light flash series was compared with the effect of intraperitoneally administered ethosuximide in freely moving albino rats with chronic electrodes. Whereas i. p. application of ethosuximide reduces or abolishes photically evoked afterdischarges and recruiting, administration into the LGB does not change these potentials or the photic evoked potential. This points to an action not immediately in the LGB.

[Hypersynchronous EEG activity and conditioned avoidance reflexes in rats].

The influence of dimethylsulfolane on active avoidance conditioning with simultaneous registration of EEG was investigated. Dimethylsulfolane, which shows some pharmacological properties of pentylenetetrazole causes a decrease of the amount of conditioned reactions. There was no strong correlation between the impairment of learning ability and the number of EEG spindle discharges after dimethylsulfolane.

Effects of ethosuximide upon psychomotor responses and electromyographic parameters of healthy individuals.

The purpose of this paper was to investigate the effects of anticonvulsant ethosuximide upon physiological parameters, psychomotor reactions and electromyographic criteria. Ethosuximide increased the reaction times to flashes and the choice reaction time in most subjects, often provoked a greater amount of errors during choice reactions and decreased the average heart frequency. The functional activation during the tasks was diminished. The EEG showed no marked qualitative or quantitative changes. Ethosuximide reduced the maximum conduction velocity of the motor nerve and changed the action potential duration but not the action potential shape. There is no exact parallelism between the increase in reaction time and decrease of nerve conduction velocity. The drug effect upon psychomotor reactions seems to be caused by reduction of vigilance and an inhibitory effect upon the individual's motor responsiveness. Like other anticonvulsants, ethosuximide may alter the electrical properties of all excitable membranes and, by this ubiquitous site of action, exerts the described effects.
