2/8 translocation in a Japanese Burkitt's lymphoma.

A new translocation between chromosomes 2 and 8, t(2p-; 8q+), was found in fresh lymphoma cells from a Japanese patient with Epstein-Barr virus-carrying Burkitt's lymphoma, and in a lymphoma cell line derived from this patient. There was no 14q+ translocation, as has been previously described in African and North American Burkitt's lymphomas.

Analysis of pure pancreatic juice in patients with chronic alcoholism.

To elucidate early biochemical changes of pancreatic juice and their reversibility in chronic alcoholics, pure pancreatic juice was collected from 23 chronic alcoholics by endoscopic retrograde catheterization of the papilla. Samples were collected at 1 minute intervals for 20 minutes after intravenous injection of secretin (Eisai, 1 U/kg) and for 10 minutes after CCK-PZ injection (Boots, 1 U/kg). Volume, bicarbonate concentration, protein concentration and three hydrolases were determined. Following results were obtained. (1) Five patients showed hypersecretory state. Four of the five patients showed hyperconcentration of protein. (2) Seventeen patients showed hyposecretory state. Lipase secretion was most frequently affected (94%). Maximal bicarbonate concentration was the next to be affected (82%). Amylase and chymotrypsinogen secretion were less frequently affected (65%). Flow rate was least frequently affected (24%). (3) It was suggested that exocrine dysfunction in chronic alcoholics is reversible in an early stage and that sequence of events with advancement of the stage is hypersecretion, hyperconcentration of protein, normalization of water secretion with a decrease in lipase secretion and maximal bicarbonate concentration, a decrease in amylase and chymotrypsinogen output, and finally a decrease in flow rate.

Heavy meromyosin from skipjack tuna, Euthynus pelamis. Preparation and enzymic properties.

A method was developed to obtain heavy meromyosin (HMM) from the tryptic digest of skipjack tuna dorsal myosin. The tuna HMM thus obtained was shown to be homogeneous on gel filtration-gel electrophoresis, and on ultracentrifugation. The sedimentation constant (S20,w) was estimated to be 6.1S for tuna HMM. The ATPase activity of tuna dorsal HMM was found to be very similar to that of rabbit skeletal HMM in many respects: KCl concentration dependence, pH dependence, effect of pCMB, kinetic parameters (Vmax and Ka) in actin activation, and Arrhenius activation energy. The only difference found between tuna HMM and rabbit HMM was in heat denaturation behavior: the ATPase activities of tuna HMM were approximately four times as sensitive to heat inactivation as those of rabbit HMM. Thus, tuna HMM should represent a good experimental material for investigations of the molecular basis of susceptibility to denaturation, and of the characteristics of fish myosins in general. A new type of heat denaturation of myosin was observed. It occurred in a very early stage of heat treatment of either tuna dorsal myosin or rabbit skeletal myosin; however, it did not occur upon heat treatment of HMM of either tuna or rabbit, and it was detectable in terms of the Mg-ATPase activity only when the activity was measured in the presence of untreated actin.

Actins

Detection of human B cells and chronic myelocytic leukemic cells by rosette formation with sheep erythrocytes and fresh human sera.

A new method of detecting the C3b receptor is reported. A particular merit of this method is that anti-RBC rabbit antiserum is not required. Rosettes were formed with human B lymphocytes, B lymphoblasts and granulocytes, using sheep erythrocytes (SRBC) sensitized with fresh human serum (FHS). T lymphocytes and T lymphoblasts did not form rosettes. The percentage of cells forming rosettes with this method approximated the percentage of rosettes formed with EACm. However, FHS coated SRBC did not react with most cells of B cell type chronic lymphocytic leukemia (CLL), whereas EACm rosette formations showed a definite reaction. On the other hand, 34--58% of cells of chronic myelocytic leukemia (CML) bound with the indicator red cells. SRBC sensitized with fresh rabbit or guinea pig serum formed rosettes with PBL, tonsil cells, B lymphoblasts and granulocytes. Complement and IgM antibody were required for this reaction, as in EAC rosette formation.

In vitro release of eosinophil chemotactic factor of anaphylaxis (ECF-A) from guinea pig skin.

An in vitro release of the eosinophil chemotactic factor of anaphylaxis (ECF-A) in anaphylactic reactions was studied using skin slices from actively sensitized guinea pigs. ECF-A and histamine were released in vitro from actively sensitized guinea pig skin by antigen challenge. The magnitude of ECF-A release did not always parallel that of anaphylactic histamine release in guinea pig skin, suggesting that there may be no correlation between the amount of ECF-A and that of histamine released from the skin in anaphylactic reactions in guinea pigs.

Anaphylaxis

Decreasing effects of glycerol-fractions extracted from ox diaphragm muscles on acetylcholine-induced contractions of smooth muscles.

Extracts of 5% glycerol obtained from ox diaphragm muscles were fractionated into four (A, B, C and D) with (NH4)2SO4. The activity as acetylcholine (ACh) receptor-like substance containing fraction was evidenced as follows; the ACh-induced contraction of tracheal muscles decreased with addition of the fraction, and such could not be attributed to the reaction with ACh receptors of tracheal smooth muscles. Fraction D had the most potent activity in the presence of neostigmine. This reaction induced by fraction D was reversed by addition of d-tubocurarine (d-TC). Fraction D was fractionated into three (I, II and III) by gel filtration on Sephadex G-75 with 50 mM phosphate buffer (pH 7.5). The purified fraction III was identified by electrophoresis, UV and visible absorption spectrum, and ion-exchange chromatography to be myoglobin. Pure myoglobin also proved to have a decreasing effect on ACh-induced contraction.

Heterotransplantation of human leukemic B-cell, T-cell and null-cell lines in hamsters.

Human leukemic B-cell (BALL-1), T-cell (TALL-1) and null-cell (NALL-1) lines have been established from three patients with acute lymphoblastic leukemia (ALL). To study the heterotransplantability and in vivo growth characteristics, attempts were made to transplant these ALL cell lines into newborn Syrian hamsters treated with rabbit anti-hamster thymocyte serum. Intraperitoneal implantation of 1.8-3.5 x 10(7) cells gave rise to invasive tumors in all recipients after 15 to 41 days. In addition to a common in vivo feature of mesenteric and retroperitoneal tumors, BALL-1 line was characterized by infiltration of the skin, massive ascites and bone marrow invasion. TALL-1 cells infiltrated various organs including the lymph nodes, liver, gallbladder, spleen, bone marrow, central nervous system and eyes. NALL-1 line grew slowly, producing the least tumors, although there were distant metastases in the lungs. Tumor cells were detected in the blood of 2 of 3 BALL-1-bearing hamsters and in the blood of 4 of 5 TALL-1-bearing hamsters. Thus, these three ALL cell lines were found to exhibit a characteristic biological behavior in hamsters, which might be related to the different cell lineage.

Exocrine pancreatic function test by a synthetic peptide.

A new synthetic substance, N-Benzoyl-L-tyrosyl-p-aminobenzoic acid, is specially cleaved by pancreatic chymotrypsin after oral administration and the released p-aminobenzoic acid (P.A.B.A.) is absorbed and excreted in the urine. The P.A.B.A. recovery in the urine was examined to evaluate its diagnostic value as an exocrine pancreatic function test. The data permit the following conclusions: 1. There is a significant correlation between this test and maximal bicarbonate concentration, amylase output and volume of P.Z./C.C.K. secretin test. 2. More than one-half to two-thirds proximal or one-third distal of the pancreas must be removed before one can expect an abnormal result in this test. 3. This is a simple and useful test to detect exocrine pancreatic insufficiency of more than moderate degree but normal results may be obtained in minimal to mild exocrine pancreatic insufficiency. Only six of 11 cases (54.5%) with one abnormal factor of P-S test showed decreased P.A.B.A. recpvery, whereas 22 of 23 cases (95.7%) with two or three abnormal factors of P-S test showed decreased or borderline P.A.B.A. recovery.

Growth characteristics of human leukemic B-cell, T-cell, and null-cell lines serially transplanted in hamsters.

Human leukemic B-cell (BALL-1), T-cell (TALL-1), and null-cell (NALL-1) lines were established from three patients with acute lymphoblastic leukemia (ALL). Intraperitoneal transplantation of these ALL cell lines into immunosuppressed newborn hamsters resulted in the development of invasive tumors in all recipients, except 4 of 10 implanted with NALL-1 line. BALL-1 and TALL-1 lines were serially transplanted for 10 and 9 passages, respectively. In addition to a common in vivo feature of mesenteric and retroperitoneal tumors, BALL-1 line was characterized by infiltration of the skin, massive ascites, and bone marrow invasion. TALL-1 cells infiltrated various organs including the lymph nodes, liver, bone marrow, central nervous system, eyes, etc. NALL-1 line grew slowly, producing the least number of tumors. Leukemic cells were detected in the blood of 24 of 25 hamsters bearing BALL-1 and in the blood of 18 of 24 hamsters bearing TALL-1. Thus, these three ALL cell lines were found to exhibit a characteristic biological behavior in hamsters.
