[Studies on tannins from Tripterygium hypoglaucum (lévl.) hutch].

OBJECTIVE: To study the tannins from Tripterygium hypoglaucum. METHOD: Compounds were isolated by column chromatography and elucidated on the basis of spectroscopic analysis and comparison of references. RESULT: The compounds obtained were determined as (+)catechin, (-)epigallocatechin, procyanidin B-3, procyanidin B-4 and gallocatechin-(4 alpha-->8) epicatechin. CONCLUSION: All the compounds were obtained from this plant for the first time.

Relationship between hydrophobicity and structure of hydrolyzable tannins, and association of tannins with crude drug constituents in aqueous solution.

The hydrophobicity values of hydrolyzable tannins were evaluated by measuring the distribution of the compounds between n-octanol and water. Of 8 gallotannins and 13 ellagitannins examined, pentagalloylglucose (7), the major polyphenol of Paeoniae Radix, showed the largest partition coefficient value. In aqueous solution, pentagalloylglucose associated with various crude drug constituents, such as paeoniflorin, glycyrrhizin potassium salt, aconitine trifluoroacetate, liquiritin apioside and amygdalin. The 1H-NMR spectroscopic examination suggested that the association occurred preferentially at the most hydrophobic sites of the molecules. The association with these compounds inhibited the distribution of pentagalloylglucose into the n-octanol phase and adsorption on hide powder. In addition, the water solubility of the biologically active polymeric proanthocyanidins of rhubarb was increased by association with rhein 8-O-glucoside potassium salt, the major anthraquinone glycoside of rhubarb.

Screening of polyacetylenic alcohols in crude drugs using the ELISA for panaxytriol.

Polyacetylenic alcohols such as panaxytriol, panaxynol and panaxydol isolated from the roots of Panax ginseng C. A. MEYER have antiproliferative activity against various cultured tumor cells. Anti-panaxytriol antibody was obtained by immunizing rabbits with panaxytriol-bovine serum albumin conjugates. Although the antibody reactivity was directed mainly toward panaxytriol, there was a slight cross-reactivity with other polyacetylenic compounds. The antibody was, therefore, used for screening a large number of crude drugs for polyacetylenic compounds such as panaxytriol. Methanol-extracts from 31 crude drugs were examined. Significant reactivity was observed in 15 methanol-extracts from Aralieaceae, Compositae and Umbelliferae as reported by other investigators. Three out of the 15 crude drugs were selected for determination of the potent cross-reactive compounds. Four kinds of cross-reactive compounds were isolated by silica gel column chromatography, monitoring each fraction using the enzyme-linked immunosorbent assay (ELISA). Among them, panaxynol and heptadeca-1,8-diene-4,6-diyne-3,10-diol were identified from Saposhnikoviae Radix. Falcarindiol was newly identified from Peucedani Radix. A new polyacetylenic alcohol, 9,10-epoxy-16-hydroxy-octadeca-17-ene-12,14-diyne-1-al, was also isolated from Foeniculi Fructus. All these polyacetylenic alcohols inhibited the growth of a human gastric adenocarcinoma cell line, MK-1 cells, in a dose-dependent manner. These results indicate that the antibody against panaxytriol is an effective tool for "screening" antiproliferative polyacetylenic compounds.

Two new iridolactones and their glycosides from the roots of Patrinia scabra.

Investigation of the roots of Patrinia scabra afforded two new iridolactones, patriscabrol and isopatriscabrol. Their structures have been established by extensive analysis of their NMR spectra and an X-ray crystallographic analysis of patriscabrol. Four monoglucosides of each iridolactone were also isolated together with an apioside of glucosidic patriscabrol.

[Studies on the toxic constituents of Illicium simonsii Maxim].

A new sesquiterpenelactone compound was isolated along with the known anisatin and shikimic acid from pericarps of the fruit of Illicium simonsii Maxim., family Magnoliaceae, collected in Sichuan province. This new sesquiterpene lactone compound exhibited high toxicity in mice. The structure of which was elucidated as 2-oxo-6-deoxyneoanisatin (I) based on spectral evidences. The detailed physical and chemical data were presented.
