New sample treatment for determination of linear alkylbenzene sulfonate (LAS) in agricultural soils by liquid chromatography with fluorescence detection.

A new sample-treatment procedure has been developed for determination of total linear alkylbenzene sulfonate (LAS), i.e. homologues and isomers, in agricultural soil. The procedure involves two steps, ultrasound-assisted extraction of LAS from the samples with methanol then clean-up of the methanolic extracts and preconcentration of the LAS by solid-phase extraction on two adsorbent cartridges (SAX and C18). The ultrasound-assisted procedure reduces extraction time (10 min in contrast with 6-12 h for conventional Soxhlet extraction) and requires only small volumes of organic solvent. The effect of different variables interacting in the ultrasound-assisted extraction process was studied. Finally, separation and quantification of the homologues and isomers of LAS was performed by liquid chromatography with fluorescence detection (LC-FD). 2-Octylbenzenesulfonic acid sodium salt (Na-2ØC8-LAS) was used as internal standard. The proposed method was satisfactorily used for determination of LAS in agricultural soil samples from the fertile plain of Granada (Spain).

Monitoring in a Lotka-Volterra model.

The problem of monitoring arises when in an ecosystem, in particular in a system of several populations, observing some components, we want to recover the state of the whole system as a function of time. Due to the difficulty to construct exactly this state process, we look for an auxiliary system called an observer. This system reproduces this process with a certain approximation. This means that the solution of the observer tends to that of the original system. An important concept for this work is observability. This means that from the observation it is possible to recover uniquely the state process, however, without determining a constructive method to obtain it. If observability holds for the original system, it guarantees the existence of an auxiliary matrix that makes it possible to construct an observer of the system. The considered system of populations is described by the classical Lotka-Volterra model with one predator and two preys and the construction of its observer is illustrated with a numerical example. Finally, it is shown how the observer can be used for the estimation of the level of an abiotic effect on the population system.

Dexmedetomidine or medetomidine premedication before propofol-desflurane anaesthesia in dogs.

The objective of this study was to evaluate dexmedetomidine as a premedicant in dogs prior to propofol-desflurane anaesthesia, and to compare it with medetomidine. Six healthy dogs were anaesthetized. Each dog received intravenously (i.v.) five preanaesthetic protocols: D1 (dexmedetomidine, 1 microg/kg, i.v.), D2 (dexmedetomidine, 2 microg/kg, i.v.), M1 (medetomidine, 1 microg/kg, i.v.), M2 (medetomidine, 2 microg/kg, i.v.), or M4 (medetomidine, 4 microg/kg, i.v.). Anaesthesia was induced with propofol (2.3-3.3 mg/kg) and maintained with desflurane. The following variables were studied: heart rate (HR), mean arterial pressure, systolic arterial pressure, diastolic arterial pressure, respiratory rate (RR), arterial oxygen saturation, end-tidal CO2, end-tidal concentration of desflurane (EtDES) required for maintenance of anaesthesia and tidal volume. Arterial blood pH (pHa) and arterial blood gas tensions (PaO2, PaCO2) were measured during anaesthesia. Time to extubation, time to sternal recumbency and time to standing were also recorded. HR and RR decreased significantly during sedation in all protocols. Cardiorespiratory variables during anaesthesia were statistically similar for all protocols. EtDES was significantly different between D1 (8.1%) and D2 (7.5%), and between all doses of medetomidine. Desflurane requirements were similar for D1 and M2, and for D2 and M4 protocols. No statistical differences were observed in recovery times. The combination of dexmedetomidine, propofol and desflurane appears to be effective for induction and maintenance of general anaesthesia in healthy dogs.

Co-digestion of ruminal content and blood from slaughterhouse industries: influence of solid concentration and ammonium generation.

At the present time, organic solid wastes from industries and agricultural activities are considered to be promising substrates for biogas production via anaerobic digestion. Moreover solids stabilisation is required before reutilization or disposal. Slaughterhouses are among the most important industries in Uruguay and produce 150,000 tons of ruminal content (RC) and 30,000 tons of blood per year. In order to determine the influence of the solids and blood contents, the ammonia inhibition and the inoculum adaptation co-digestion batch tests were performed. A set of experiences with TS concentration of 2.5%, 5% and 7.5% and different ratios of RC/blood were carried out using an inoculum from an UASB reactor. In all experiences fast blood hydrolisation was observed. A higher methane production was detected in the experiences with higher TS content. However, the fraction of solids degradation was lower in these experiences. A plateau in the biogas production was found. The free ammonia level, which was above the reported inhibitory levels, could explain this behaviour. After the inhibition period the biogas production restarted probably due to the biomass acclimatisation to the ammonia. In order to determine the inoculum adaptation a new experiment was performed. The inoculum used was the sludge coming from the first set of experiences. Based upon batch tests a 3.5 m3 pilot reactor was designed and started up. Ammonia inhibition was avoided by the start-up strategy and in two weeks the biogas production was 3.5 m3/d. The VS stabilisation with a solid retention time of 20 days was of 43%. The pilot reactor working at steady state had a TS concentration of 3-4% with a ratio of RC/blood of 10:1 at the entrance.

Start-up and steady-state results of a full-scale UASB reactor treating malting wastewater.

An Imhoff tank was reconstructed into a 250 m3 UASB reactor in order to treat a malting plant wastewater. The UASB was inoculated with sludge from an anaerobic lagoon used for slaughterhouse wastewater treatment. After two months of operation the reactor achieved full load with an HRT of 17 h, a COD removal higher than 80% and a biogas production of 300 m3/day (77% average methane content), with an organic loading rate of 3.6 kgCOD/m3.d (0.24 kgCOD/kgVSS.d). A yield coefficient of 0.09 gVSS/gCODrem was found from a mass balance. The fat present in the inoculated sludge (48 mg/gSSV) did not affect the start up performance. Sludge from the inoculum with high content of fat (270 mg/gSSV), was separated by flotation in the first week of operation. The COD removal efficiency was scarcely influenced by the reactor operation temperature (17-25 degrees C).

A maize defective-kernel mutant (longcell) characterized by tubular cells, severe morphological alterations and induction of cell death.

Seeds of the longcell mutant in maize (Zea mays L) have a defective-kernel phenotype: the embryo aborts at the early coleoptilar stage and the endosperm is reduced in size. Mutant embryos have severe alterations in morphogenesis. They have a suspensor-, an embryo axis- and a scutellum-like structure, but the shoot apical meristem (SAM) is not formed. Scanning electron microscopy showed that most of the cells in longcell embryos are tubular and abnormally enlarged. The level of expression of several genes involved in basic metabolism is not severely affected during early and mid embryogenesis, but storage molecule accumulation is reduced. Genes which in normal conditions are only expressed after germination, are expressed during kernel development in the longcell seeds. Mutant embryos undergo cell death in late embryogenesis. Nuclei in dying embryos are TUNEL positive, and different genes coding for hydrolytic enzymes are up-regulated. The expression of genes related to oxidative stress is also altered in longcell embryos. These results lead us to suggest that the longcell mutant may be cytokinesis-defective.

Diurnal variations in the plasma concentration of parathyroid hormone in dogs.

The plasma concentrations of parathyroid hormone (PTH), ionised calcium (Ca(2+)), total calcium, albumin and inorganic phosphorus, and the pH were measured in blood samples obtained from nine dogs during a period of 26 hours. The plasma pth levels fluctuated slightly during the day, by about 20 pg/ml, but there was a distinct peak (42.8 [8.8] pg/ml) at 07.00. Plasma Ca(2+) showed a diurnal pattern in which two peaks (increases of 0.03 mmol/l) were observed at 05.00 and 17.00, and the plasma concentration of inorganic phosphorus showed a similar pattern. There were no diurnal changes in total calcium or albumin.

Equilibrium, observability and controllability in selection-mutation models.

In this paper we shortly discuss the problem of the equilibrium in the well-known Fisher type selection-mutation model, also providing a formula for particular three-allele models. The considered continuous-time dynamics is a known extension of the classical model of natural selection given by Fisher. We also extend the existing investigation of the observability of Fisher's model to the case when another evolutionary factor, mutation is also present. Moreover, we prove a result of technical character, which makes it possible to apply the methodology of nonlinear systems with invariant manifold, to models of artificial selection. For an illustration, a class of three-allele systems is presented in which the controllability into equilibrium is guaranteed without any condition on the biological parameters.

Observability in dynamic evolutionary models.

In the paper observability problems are considered in basic dynamic evolutionary models for sexual and asexual populations. Observability means that from the (partial) knowledge of certain phenotypic characteristics the whole evolutionary process can be uniquely recovered. Sufficient conditions are given to guarantee observability for both sexual and asexual populations near an evolutionarily stable state.

Deletion of BCR region 3' in chronic myelogenous leukemia.

The t(9;22)(q34;q11) produces the BCR/ABL fusion gene which codifies a 210 kb protein with a strong tyrosine kinase activity and is involved in cellular development and growth. Because this translocation is a reciprocal event, it could give rise to a second fusion gene, ABL-BCR, on the derivative 9q+. We analyzed the influence of the 3' M-BCR deletion on the clinical picture at diagnosis and disease outcome in 57 patients with a clinical diagnosis of CML. Molecular studies were done on DNA from peripheral blood leukocytes or bone marrow with the restrictions enzymes BglII, EcoRI, HindIII, and BamHI, and the BCR 3' probe (transprobe 1) (Oncogene Science Inc.), which encompasses almost all of the 5.8 Kb of the M-BCR gene area. In 18 patients Southern blot analysis showed deletion of the 3' end of BCR gene (32.7%). There were no significant differences between patients with or without deletion, either in the clinical and laboratory data at the disease diagnosis or at the disease outcome. The absence of differences between the patients with and without 3' BCR deletion supports the hypothesis that the hybrid gene ABL-BCR does not have an important role in leukemogenesis in CML cases.

Plasma ionized calcium and parathyroid hormone concentrations in horses after endurance rides.

OBJECTIVE: To evaluate changes in plasma ionized calcium (Ca2+) and parathyroid hormone (PTH) concentrations in horses competing in endurance rides. DESIGN: Longitudinal clinical study. ANIMALS: 28 horses. PROCEDURE: Venous blood samples were obtained from horses before and after racing 80 km. Plasma pH and concentrations of Ca2+, PTH, inorganic phosphorus, albumin, lactate, and magnesium were measured. RESULTS: Overall, a significant decrease in mean (+/- SD) plasma Ca2+ concentration (from 6.44 +/- 0.42 to 5.64 +/- 0.42 mg/dl) and a significant increase in plasma PTH concentration (from 49.9 +/- 30.1 to 148.1 +/- 183.0 pg/ml) were found after exercise. Exercise also resulted in significant increases in plasma inorganic phosphorus, albumin, and lactate concentrations. No changes in plasma magnesium concentration or pH were detected after exercise. Plasma PTH concentration was not increased after exercise in 8 horses; in these horses, plasma PTH concentration decreased from 58.2 +/- 26.3 to 27.4 +/- 22.4 pg/ml, although plasma Ca2+ concentration was also decreased. CONCLUSIONS AND CLINICAL RELEVANCE: Plasma Ca2+ concentration was decreased after racing for 80 km, compared with values obtained before racing. In most horses, an increase in plasma PTH concentration that was commensurate with the decrease in plasma Ca2+ was detected; however, some horses had decreased plasma PTH concentrations.

Three- and four-membered rings from cycloadditions of 1,3-thiazolium-4-olates and aldehydes.

2-Aminothioisomünchnones, a well-known family of masked dipoles, react with aromatic aldehydes in a domino cascade reaction that produces episulfides (thiiranes) or beta-lactams (2-azetidinones). This sequence is initiated by a [3+2] dipolar cycloaddition followed by ring opening of cycloadducts and intramolecular rearrangement to afford these unusual ring contractions. The nature of the reaction products depends on the structural characteristics of the starting dipole and the experimental conditions. Episulfides are obtained selectively as cis isomers with respect to both aryl groups, whereas beta-lactams are produced as cis/trans mixtures. These structural features were determined unequivocally by X-ray crystallographic analysis. The beta-lactams still possessed a flexible acyclic chain containing sulfur, a salient lead modification of the bioactive cyclic penems and cephems. The preferential production of exo transition structures was rationalized with the aid of computational calculations at the B3LYP/6-31G* level.

Blanket development in a malting wastewater anerobic treatment.

A 3 m3 pilot UASB reactor was operated with malting wastewater. In order to find the aggregation characteristics, the sludge concentration profile was determined throughout time. Reactor behaviour concerning the physical properties of the sludge can be evaluated using this procedure. Moreover, organic load per VSS can be calculated from this profile. The organic load based per VSS is more accurate as a design parameter than the volumetric load. Under the study conditions imposed the COD removal efficiencies achieved were about 85%. Organic loads of 0.6-0.7 kg COD/(kgVSS.d) were reached. After operating at 30 degrees C, temperatures of 28 degrees C and 15 degrees C were tested, showing no decrease in COD removal efficiency. At week 15 the presence of granules was verified, and after that, sludge concentrations of 30-35 kg VSS/m3 were measured at the bottom of the reactor. The SVI of the granular sludge decreased to values below 20 mL/gVSS. The settlement velocity was 10 m/h, which is much greater than the recommended values for the settlement zone; therefore, no problems are to be expected in the settler regarding the sludge sedimentation. The biomass yield coefficient obtained was 0.10 gVSS/gCOD in agreement with literature values for brewering effluent.

Further studies on the partial double Donnan. Is isosmotic KCl solution isotonic with cells of respiratory trees of the holothurian Isostichopus badionotus Selenka?

As potassium, chloride and water traverse cell membranes, the cells of stenohaline marine invertebrates should swell if exposed to sea water mixed with an isosmotic KCl solution as they do when exposed to sea water diluted with water. To test this hypothesis respiratory tree fragments of the holothurian Isostichopus badionotus were exposed to five isosmotic media prepared by mixing artificial sodium sea water with isosmotic (611 mmol/l) KCl solution to obtain 100, 83, 71, 60 and 50% sea water, with and without 2 mmol/l ouabain. For comparison, respiratory tree fragments were incubated in sea water diluted to the same concentrations with distilled water, with and without ouabain. Cell water contents and potassium and sodium concentrations were unaffected by KCl-dilution or ouabain in isosmotic KCl-sea water mixtures. In tissues exposed to H(2)O-diluted sea water, cell water increased osmometrically and potassium, sodium and chloride concentrations decreased with dilution; ouabain caused a decrease in potasium and an increase in sodium but no effect on chloride concentrations. The isotonicity of the isosmotic KCl solution cannot be adscribed to impermeability of the cell membrane to KCl as both ions easily traverse the cell membrane. Rather, operationally immobilized extracellular sodium ions, which electrostatically hold back anions and consequently water, together with the lack of a cellward electrochemical gradient for potassium, resulting from membrane depolarization caused by high external potassium concentration, would explain the isotonicity of isosmotic KCl solution. The high external potassium concentration also antagonizes the inhibitory effect of ouabain on the Na(+)/K(+) ATPase responsible for sodium and potassium active transport.

Prevalence and viral persistence of TT virus in patients on hemodialysis.

The aim of this study was toevaluate the prevalence of TT virus (TTV) in 107 hemodialysis patients and 100 healthy individuals. Sixteen percent of hemodialysis patients and 2% of the control population were TTV positive (P<0.001). Sex, time on dialysis, transfusions, and alanine aminotransferase levels were not related to the presence of TTV DNA. TTV was more frequent in patients 50 to 70 (12/51) years of age and persisted in the serum of six of nine patients 8 months later. TTV was found in peripheral blood mononuclear cells of all patients with chronic infection, and sequence variation was noted in the peripheral blood mononuclear cells of two patients. No relationship between TTV infection and hepatitis was observed. Viral persistence and nonparenteral transmission may be possible in patients on hemodialysis.

Quantitative analysis of acid-base balance in show jumpers before and after exercise.

The acid-base status of venous blood was studied in 17 show jumpers before and after exercise using both a traditional and a quantitative approach. Partial pressure of carbon dioxide (PCO(2)), pH, haemoglobin, and plasma concentrations of sodium (Na(+)), chloride (Cl(-)), potasium (K(+)), ionized calcium (Ca(2+)), total proteins, albumin, lactate and phosphorus were measured in jugular venous blood samples obtained before and immediately after finishing a show jumping competition. Bicarbonate, anion gap and globulin concentration were calculated from the measured parameters. 'Quantitative analysis' of acid-base balance was performed utilising values for three independent variables: PCO(2), strong ion difference [SID = (Na(+)+ K(+)+ Ca(2+)) - (Cl(-)+ Lact)] and total concentration of weak acids [A(T)= Alb (1 paragraph sign23 pH - 6 paragraph sign31) + Pi (0 paragraph sign309 pH - 0 paragraph sign469) 10/30 paragraph sign97]; plasma concentrations of hydrogen ion ([H(+)]) were also calculated from these variables using Stewart's equation. No significant changes in blood pH were detected after the show jumping competition. Exercise resulted in a significant increase in lactate, Na(+), K(+), haemoglobin, total proteins, albumin, globulin and anion gap, and a decrease in bicarbonate, Cl(-)and Ca(2+). PCO(2)decreased after exercise while SID and A(T)increased. A significant correlation between measured and calculated [H(+)] was found both before and after exercise. However, individual [H(+)] values were not accurately predicted from Stewart's equation. In conclusion, even though pH did not change, significant modifications in the acid-base balance of horses have been found after a show jumping competition. In addition, quantitative analysis has been shown to provide an adequate interpretation of acid-base status in show jumpers before and after exercise.

Phylogenetic comparison of the DEN-2 Mexican isolate with other flaviviruses.

Recent attention has focused on the geographic variation of dengue viruses, since major epidemies may follow introduction of a new virus strain into susceptible populations. We cloned and sequenced a very interesting Mexican isolate (200787/1983) which is antigenically unique by signature analysis with respect to all other dengue-2 topotype viruses. This strain is also unique in biological behavior (neurotropism) and is of epidemiological significance in Mexico. The dengue-2 Mexican isolate sequence information was compared with that of other flaviviruses, analyzing the branching structure of the phylogenetic tree reconstructed from the E gene amino acid sequences. The E glycoprotein, is target for neutralizing antibodies and T-cell responses, and defines the tropism and virulence of flaviviruses. In the phylogram, our strain was located in the position of greatest dissimilarity within serotype-2. Also, frequency analysis of amino acids revealed a very different signature pattern from that found in viral serotype-2.
