Inhibition, reactivation, and determination of metal ions in membrane metalloproteases of bacterial origin using high-performance liquid chromatography coupled on-line with inductively coupled plasma mass spectrometry.

High-performance liquid chromatography coupled on-line with inductively coupled plasma mass spectrometry (HPLC-ICP-MS) was used for the characterization of metal ions in several metalloproteases of bacterial origin. The different components of the bacterial extracts were separated on a size-exclusion column. The eluent of the HPLC system was continuously transported to the ICP-MS system for rapid, reproducible, and sensitive analyses of trace elements in the metalloproteases. Two different membrane proteases from Bacillus cereus and Pseudomonas aeruginosa were characterized to be zinc metalloproteases using enzymological methods and HPLC-ICP-MS. The zinc content was determined to be three molecules of zinc per protein molecule for the B. cereus protease and one molecule of zinc per protein molecule for the P. aeruginosa protease. For another purified protease, a periplasmic alanyl aminopeptidase of P. aeruginosa, the lack of protein-bound metal ions could be clearly determined-a confirmation that this main aminopeptidase of P. aeruginosa belongs to the cysteine protease family. The presence of nonionic detergents can influence the distribution of trace elements during the HPLC separation. Therefore, the use of these substances should be avoided during enzyme purification for metal analyses or they should be exchanged later for zwitterionic and ionic detergents with more strongly dissociating properties.

Isolation of a cDNA coding for an ubiquitin-conjugating enzyme UBC1 of tomato--the first stress-induced UBC of higher plants.

A clone of an ubiquitin-conjugating enzyme (UBC) was isolated from a lambda-ZAP-cDNA library, generated from mRNA of tomato (Lycopersicon esculentum) cells grown in suspension for 3 days. The open reading frame called LeUBC1, encodes for a polypeptide with a predicted molecular mass of 21.37 kDa, which was confirmed by bacterial overexpression and SDS-PAGE. Database searches with LeUBC1 showed highest sequence similarities to UBC1 of bovine and yeast. By Southern blot analysis LeUBC1 was identified as a member of a small E2 subfamily of tomato, presumably consisting of at least two members. As revealed by Northern blot analysis LeUBC1 is constitutively expressed in an exponentially growing tomato cell culture. In response to heat shock an increase in LeUBC1-mRNA was detectable. A strong accumulation of the LeUBC1-transcript was observed by exposure to heavy metal stress which was performed by treatment with cadmium chloride (CdCl2). The cellular uptake of cadmium was controlled via ICP-MS measurements. The data suggest that like in yeast, in plants a certain subfamily of UBC is specifically involved in the proteolytic degradation of abnormal proteins as result of stress.

Pseudomonas cepacia endophthalmitis.

A 72-year-old white man who had undergone surgical trabeculectomy and extracapsular cataract extraction with a posterior-chamber lens implantation in the left eye suffered from chronic iridocyclitis for eight months. He subsequently presented with acute hypopyon and vitritis. Anterior-chamber and vitreous cultures were positive for Pseudomonas cepacia. The infection was successfully treated with subconjunctival piperacillin, intravitreal cefotaxime, and intravenous piperacillin and gentamicin. To our knowledge, this is the first documented case of Pseudomonas cepacia endophthalmitis.

Ocular absorption and metabolism of topically applied epinephrine and a dipivalyl ester of epinephrine.

An analogue of epinephrine (EPI), dipivalyl epinephrine (DPE), has been found to reduce intraocular pressure (IOP) significantly at lower concentrations than EPI itself. In order to understand the reason for this increased activity, the ocular penetration, distribution, and metabolism of these two compounds were compared. About 10 times as much DPE as EPI was absorbed by rabbit eyes, with the cornea as the major repository for the increased amount of drug absorbed. Comparison of the partition coefficients of the two compounds showed DPE to be from 100 to 600 times as lipophilic as EPI. The radioactive materials found in the aqueous humor after treatment with either compound had the same mobility in a thin-layer chromatographic system. The data indicate that following the increased penetration of the lipophilic prodrug, DPE is hydrolyzed to EPI in the eye.
