Chronic electrical stimulation of the chinchilla cochlea.

The scanning electron microscope (SEM) was used to investigate morphological changes to the chinchilla cochlea following chronic extracochlear electrical stimulation. Silver ball electrodes were placed on round window membranes and various types and levels of electrical pulsatile stimulation was given. Positive or negative square wave pulse (with DC offset) stimulation caused severe damage to the organ of Corti. In the most severely damaged area, most outer hair cells were missing and degenerative changes to pillar cells were observed. Some damage to the inner hair cells was also evident. These changes were limited to the basal turn. Some damage to stereocilia of first row outer hair cells was seen when symmetrical, biphasic, charge balanced pulses were used.

Cochlear effects of cryosurgery.

A total of 21 cochleas in 13 chinchillas were treated with cryosurgery and the damage to the organ of Corti was evaluated with the scanning electron microscope. The apical half-turn was treated in 11 of the cochleas and the basal half-turn in 10. Apical exposure to the cryoprobe generally resulted in severe damage to sensory cells in the apical turn whereas basal turn exposure generally resulted in mild damage to sensory cells in the basal turn. On occasion severe damage from apical turn exposure resulted in severe damage that extended to the basal turn. First row outer hair cells were the most susceptible to damage and inner hair cells least susceptible.

Hearing and hair cells.

Animal experiments and human temporal bone studies have indicated that hearing losses may occur in ears that have normal populations of sensory cells. The possibility exists that the hair cells have been rendered non-functional because of ultrastructural damage. Preliminary investigation of cilia on hair cells in areas of suspected damage shows no difference from cilia in similar areas of control ears.

Use of the CO2 laser in otology.

There are conflicting reports of the effect on the inner ear of the use of a carbon dioxide (CO2) laser. In four cats, we made middle ear lesions with a CO2 laser. Subsequent evaluation by inverted-phase and scanning electron microscopy revealed no inner ear pathology attributable to the CO2 laser. Apparently, appropriate use of the CO2 laser does not harm the inner ear.

An electron microscopic study of the vestibular sensory epithelium.

The osmium-thiocarbohydrazide preparation technique previously used in conjunction with scanning electron microscopy for assessment of cochlear anatomy has been used to study vestibular structure. The technique allows a detailed study of vestibular anatomy which, when supplemented with transmission electron microscopy, gives a more complete picture of vestibular structure than previously presented.
