Fluorescent microscopic study of epithalon binding in maternal and fetal rabbit tissues in health and under conditions of placental insufficiency.

Epithalon (regulatory tetrapeptide) labeled with dansil (fluorescent stain) easily penetrates into all tissues and organs of pregnant rabbit females and through the placenta into fetal organs. Incorporation of labeled epithalon in placental tissues is more often observed in fetuses developing under conditions of placental insufficiency than in normal fetuses.

Expression of argyrophilic proteins in the nucleolar organizer regions of human thymocytes and thymic epitheliocytes under conditions of coculturing with vilon and epithalon peptides.

Vilon stimulated and Epithalon suppressed the expression of argyrophilic proteins in nucleolar organizer regions of thymocytes and epithelial cells, stimulating or reducing, respectively, the formation, assembly, and transport of ribosomes into the cytoplasm and thus determining the intensity of protein synthesis in these cells. A direct mitogenic effect of Vilon was also revealed: this peptide promoted thymocyte transformation into proliferating blast cells.

Ultrastructure of the pineal gland after gamma-irradiation under conditions of inhibition of adrenocortical function.

Electron microscopy showed that whole-body gamma-irradiation in sublethal doses led to the appearance of injuries in pinealocytes, glial cells, and vessels of the pineal gland in rats. Limitation of the nonspecific effect of gamma-irradiation via inhibition of adrenocortical function with metopirone in physiological doses reduced the radiation-induced ultrastructural damage to the pineal gland.

Ultrastructure of the rat duodenal endocrine cells after prolonged irradiation.

We propose classification of duodenal endocrine cells of intact rats based on ultrastructural signs of secretory granules and subdivided these cells into 10 basic types. The effect of long-term irradiation in a total dose of 2.5 Gy on ultrastructural organization of duodenal apudocytes was studied. Irradiation induced nonspecific changes of cell organelles in apudocytes. Differences in the ultrastructural disorganization were detected between different types of apudocyte populations and between different types of endocrine cells. Under conditions of adaptation to radiation apudocytes released the secretory product not only through molecular extrusion and exocytosis, but also via degranulation.

Immunohistochemical and morphometric analysis of effects of vilon and epithalon on functional morphology of radiosensitive organs.

Studies of the effects of vilon and epithalon on functional morphology of the thymus, spleen, and duodenum in intact rats and rats exposed to single whole-body gamma-irradiation in a dose of 6 Gy showed that vilon stimulated proliferative activity of thymocytes and enhanced proliferative potential of stem cells in the intestine, thus stimulating the postradiation recovery of critical organs. Epithalon decelerated metabolic processes in the duodenal mucosa and suppressed hemopoiesis and lymphopoiesis in the spleen.

Age-specific ultrastructural postradiation changes in intestinal endocrine cells.

Age-specific ultrastructural postradiation changes in the main types of duodenal endocrine cells (apudocytes) were studied in rats 6 and 12 months after single whole-body irradiation in doses of 7 and 7.5 Gy. Ultrastructural disorganization of different severity was detected, which depended on apudocyte type and term postradiation. Degranulation was the basic mechanism of hormone secretion in delayed periods after the exposure.

Modulating effects of epithalamin and epithalon on the functional morphology of the spleen in old pinealectomized rats.

Immunohistochemical and morphometric analysis showed that epithalamin and epithalon produced similar effects on the functional morphology of the spleen in pinealectomized rats. Both peptides prevented hyperplasia of lymphoid cells in follicular germinative centers induced by pinealectomy and potentiated the decrease in extramedullary hemopoiesis. These findings confirm the data on functional relationships between the pineal gland and immune system. The effects of epithalamin and epithalon on cell and tissue homeostasis in the spleen of old pinealectomized rats can be regarded as a manifestation of the general regulatory effect of these peptides.

Hormonal function of nonendocrine cells: role of new biological phenomenon in the regulation of homeostasis.

Recent studies revealed a new biological phenomenon: hormone synthesis in nonendocrine cells. Here we review hormone production by 4 types of nonendocrine cells of different origins, localizations, and functions and the role of this biological phenomenon in the maintenance of homeostasis. Our results and published data suggest that hormonal function is a general biological property not specific for only neuroendocrine cells, but rather typical of all living cells independently of their origin and role in the body.

[Effect of cycloferon on dissemination of Lewis lung carcinoma in mice and rhabdomyosarcoma ra-23 in rats].

Tumor growth and proliferative activity of tumor cells were suppressed and the number of pulmonary metastases in C57B16 mice decreased 3.3-fold following seven injections of cycloferon (100 mg/kg body) to induce interferon production. Injections were carried out 1-16 days after subcutaneous transplantation of Lewis lung carcinoma. After mice were immunized with ovine red blood cells, cycloferon administration raised thymus-dependent humoral immune response. After eight injections of cycloferon (50 mg/kg body) into rats, from day of intravenous transplantation of rhabdomyosarcoma RA-23 until day 20, no significant effect on metastasizing into the lung was recorded. However, single injection of cyclophosphamide 50 mg/kg inhibited metastasis formation. The highest suppressor effect was registered with combination cycloferon-cyclophophamide treatment: mean weight of metastasis decreased by half, as compared with treatment with cyclophosphamide alone. Both drugs caused karyotypical abnormalities to occur in metastatic cells. Tumor growth and spreading suppression after cycloferon should be attributed to cytotoxic antitumor action, cell proliferation inhibition and immunomodulating effect.

The effect of the peroxisome proliferator ciprofibrate on the gastric mucosa and particularly the gastrin cell.

The peroxisome proliferator ciprofibrate induces hypergastrinemia without inhibiting acid secretion. The present study was carried out to assess the effect of ciprofibrate on serum gastrin and gastrin (G) cells in different strains of rats and to compare the effect of ciprofibrate with other lipid-reducing agents (lovastatin and simvastatin) which have a different mechanism of action. Serum gastrin was determined by a radioimmunoassay method, G cell density by histomorphometry after immunostaining for G cells, and gastrin, somatostatin and histidine decarboxylase (HDC) mRNA abundance by Northern blot analysis. Ciprofibrate (100 mg/kg/day for three weeks) induced a marked hypergastrinemia (P < 0.01) in male and female Fischer rats as well as in female Wistar rats. Simvastatin and lovastatin did not affect serum gastrin. Antral G cell density increased significantly in female Wistar rats (P < 0.05) and non-significantly in the other rats after ciprofibrate. Both gastrin and somatostatin mRNA abundance in antral mucosa increased markedly and significantly (P < 0.01) after ciprofibrate treatment. The present study shows that the peroxisome proliferator ciprofibrate induces hypergastrinemia secondary to an increased storage and synthesis of antral gastrin. Since somatostatin mRNA abundance also increased, the present study suggests that ciprofibrate and possibly other peroxisome proliferators in sufficient concentrations have a stimulatory effect on endocrine cells.
