[Changes in several characteristics of the erythrocyte membrane and erythrocyte structure in severe burns in animal experiments].

It is determined in experiments on rats that the IIIB degree thermal injuries of 20% of the total body surface significantly reduce the erythrocytic membrane's resistance and cause the production of peroxides in 15 minutes after trauma. Simultaneously the electrical properties begin to change. The structure of erythrocytes is also altered. In 30 minutes post injury these alterations are in progress, but in 60 minutes they are less expressed. The structure of the erythrocytes is significantly altered. The authors consider that the influence of membranotoxic factors--the products of free radicals' reactions and phospholipases--is accounted for all noticed alterations.

[Effect of administration of ampiox and guanosine triphosphate on the concentration of cyclic nucleotides in the muscle tissue of a zone of inflammation].

Experiments on rabbits with infiltrates induced by intracutaneous injection of staphylococcal culture were made to examine the content of cyclic nucleotides in the adjacent muscle tissue under the effect of administering ampiox alone or combined with guanosine triphosphate (GTP). Combined injection of ampiox and GTP to the infected animals produced a considerable elevation in the content of cAMP (more than 2 1/2-fold) and in the cAMP/cGMP ratio along with a beneficial therapeutic effect. The possible mechanisms of action of the agents administered are discussed.

[Mechanism of action of collagen on processes of regeneration].

The effect of collagen, its hydrolysate and glycin on metabolism of cyclic nucleotides in wound tissue was studied in experiments on animals. Collagen was found to reduce the cAMP level in muscles of the wound fundus, while the concentration of cGMP remained unchanged. Collagen hydrolysate induced unidirectional changes in cyclic nucleotides, whereas glycin opposite ones. The basal activity of cAMP phosphodiesterase was not changed. The mechanism of the stimulatory effect of collagen on wound healing is discussed.

[Repair processes in wound tissues of experimental animals following administration of glycine].

A study was made of the effect of glycine given in doses approximating the physiological ones on the repair of processes in rat wound tissues. It was disclosed that in the early periods of wound healing, glycine administration leads to the increased content of cAMP and cAMP/cGMP ratio in the wound muscle and then in the granulation tissue, which appears to promote the intensification of the repair processes manifesting in the changes in tissue metabolism (DNA, collagen), in anti-inflammatory action, as well as in a more rapid maturation of the granulation tissue and wound reduction. It was also found that the doses of glycine tested do not affect the content of insulin and hydrocortisone in the blood of experimental animals.

[Effect of cyclic guanosine monophosphate on certain indices of muscle tissue carbohydrate metabolism during the wound process].

The effect of intraperitoneal administration of cGMP (0.5 mg per animal) on carbohydrate metabolism of wound area muscle tissue was studied in experiments on rats with linear skin wounds. The content of glycogen, gluconeogenesis, activity of glycogen phosphorylase, lactate dehydrogenase and malate dehydrogenase were studied. Cyclic GMP induced a substantial activation of glycogen metabolism (elevation of gluconeogenesis, increase in the activity of glycogen phosphorylase) even the third day after the operation. The animals not given cGMP demonstrated such an activation only the fifth day following the operation. Under the effect of cGMP the activity of lactate dehydrogenase rose the third day after the operation. Thus cGMP administration to the animals with wounds leads to an earlier mobilization of energy resources thereby promoting the acceleration of wound healing.

[Dynamics of the change in cyclic nucleotide content in wound tissues].

Changes in the content of cyclic nucleotides (cAMP and cGMP) in the wound tissues (muscles and granulations) were studied in experiments on rats. A wound with skin defect in the dorsal area with a crushed underlying muscle served as an experimental model. It was shown that the cAMP content in the muscular tissue rises twice: during the 1st day and to a greater measure on the 7th day. The cGMP content slightly increases on the 1-4th day, drops on the 7th day and increases cAMP concentration changes similarly to that inthe muscular tissue: it increases on the 7th day and drops on the 14th day. On the contrary, the cGMP content curve in granulations is more monotonous, only a slight increase being observed on the 7th day.

[Inhibitors of trypsin-like proteolytic enzymes as means of preventing the development of secondary necrosis in burn wounds].

The effect of proteolytic inhibitor contrical on the experimental burn wound healing was studied in rats using biochemical, histological and histochemical methods. In control untreated animals with flame burn of 20% of body surface wound healing was associated with development of secondary necrosis, marked inflammatory reaction, augmented activity of proteases and peptidases. The use of contrical prevented secondary necrosis; this effect was apparently related to reduction of tissue proteolytic enzymes activity.

[Characteristics of the lactate dehydrogenase isoenzyme spectrum of the skin in the presence of thermal lesions].

Experiments were conducted on rats; a study was made of the activity of lactic dehydrogenase (LDH) and its isoenzymes in the zone of affection and in the adjacent areas of the skin at various periods after the burn infliction; on the 1st--8th day there occurred a reduction of the sum total LDH activity in the scab zone and the underlying tissue by 70--80%, and in the margin and the intact skin--by 50%. These changes were accompanied by shifts in the isoenzymatic LDH spectrum in the affected tissue; the activity of fraction 5 displayed a sharp rise on the 1st day and that of fractions 2 and 3--a reduction, on the contrary, by the 8th day there was seen some diminution of fraction 5 activity and an elevation of fractions 2 and 3 activity. The following picture is observed on the 14th--22nd day after the burn; the sum total LDH activity remains low, the isoenzymatic LDH spectrum in the margin and the scab is largely normalized, whereas in the underlying tissue there persist changes in the LDH enzyme ratio (a reduction of fractions 1--3 activity, and a rise of fraction 5 activity).

[Effect of the protease inhibitor--contrical--on granulation tissue metabolism].

Studies of the influence of contrycal on metabolism of rat granulation tissue showed that this protease inhibitor stimulated C14-glycine incorporation into collagen proteins and caused reorganization of the isoenzyme LDH spectrum; at the same time there was a delay in the maturation of the granulation tissue with its marked growth.

[Effect of contrical on the activity of dehydrogenases and their isoenzymes in the muscles and organs of animals with developing granulation tissue].

Studies of the influence of contrycal on condition of the enzyme systems of the muscles, liver, kidneys and heart of rats with developing granulation tissue showed contrycal, a protease inhibitor, to have a stimulating effect on the LDH and MDH activity and on their isozyme spectrum. The effect of the inhibitor was expressed in the changes occurring in the state of the enzyme system, both at the site of damage (granulations and the underlying tissue) and in a number of internal organs -- the liver and kidneys.
