[The effect of ultrasound, the polyene antibiotic amphotericin B and their combination on the biomembranes of yeast-like fungi in the genus Candida].

The effect of ultrasound (US) and the antibiotic amphotericin (A-B) on the structure of Candida albicans membranes. The most expression of destructive processes and appearance of free radicals in mycetic cells was observed in combination of US and A-B by chemiluminescence and ultrastructural analyses. The mechanisms of action of US and A-B on mycetic cells were discussed in the article. The expedience of the combination of US and A-B in candidiasis as a factor, increasing fungicidal effect on the candidiasis agent of Candida albicans.

Amphotericin B↗

[The cellular acid phosphatase activity in yeast-like fungi of the genus Candida exposed to ultrasound, polyene antibiotics and dyes].

The activity of one of the lysosomal membrane marker enzymes--acid phosphatase from the Candida yeast fungi on their exposure to ultrasound (US), polyenic antibiotics (amphotericin B and nystatin) dye antiseptics (ethacridine lactate, methylene blue), and their combinations was assayed. The impact of US and the drugs, in particular their combination, was found to be followed by activation of the fungal lysosomal apparatus function and increases in their catabolic processes. The highest rise in lysosomal catabolic activity was found when the polyenic antibiotics were used in combination with US, which reflects the higher damaging effect of this combination against Candida lysosomal membranes than the dyes and of these antibiotics and US alone. The studies provide strong evidence for the preference of the combined use of US and the polyenic antibiotics in candidiasis as a factor enhancing their fungicidal effect against Candida yeast fungi.

[The characteristics of the ultrastructural changes in yeast-like fungi of the genus Candida exposed to therapeutic-range ultrasound and nystatin].

The electron microscopic study of the ultrastructure of yeast-like fungi of the genus Candida under the action of nystatin and sonication, as well as the action of these two factors used in combination, was carried out. In this study the degree of preservation of the structure of fungal cells was evaluated and the percentage of cells with different ultrastructural changes was determined. Observations made 3 and 24 hours after sonication and treatment with nystatin, used in combination, showed that ultrastructural changes in fungal cells were more pronounced than in the cells subjected to the action of nystatin alone. 24 hours after the combined physico-medicinal treatment of Candida the percentage of nonviable blastospores among them was 5-fold higher than that of such cells among Candida subjected to treatment with nystatin alone and more than 20-fold higher than among Candida subjected only to sonication. These data on the increased fungicidal effect of nystatin used in combination with sonication substantiate the expediency of the above-mentioned physico-medicinal treatment in cases of candidal lesions of the skin and its appendages, which is one of the ways for increasing the effectiveness of the treatment of this mycosis.

[Comparative levels of amphotericin B in the skin and subcutaneous fatty tissue after cutaneous application of amphotericin ointment by phonophoresis and with preliminary treatment by dimethyl sulfoxide].

A comparative study of the amphotericin B contents in the skin and subcutaneous fatty tissue was performed on guinea pigs after local application of amphotericin ointment by phonophoresis and with preliminary treatment of the skin by dimethyl sulfoxide (DMSO). When the amphotericin ointment was used in combination with ultrasound the content of amphotericin B in the skin and subcutaneous fatty tissue 1, 3, 24, 48 and 72 hours after the application was much higher than that after the ointment local application without the ultrasonic treatment. When the amphotericin ointment was applied locally after the preliminary treatment with DMSO the maximum content of the antibiotic in the skin and subcutaneous fatty tissue was observed 3 hours after the application which was significantly higher than the content observed after the ointment application by phonophoresis and especially locally without the ultrasonic treatment. In 24, 48 and 72 hours the amphotericin B concentration in the skin and subcutaneous fatty tissue under any conditions lowered and in 24 hours had a tendency to level in the areas treated with ultrasound and DMSO. In 48 and 72 hours the highest concentrations of the antibiotic were in the skin and subcutaneous fatty tissue after the ointment application by phonophoresis.

[A comparative evaluation of amphotericin pharmacokinetics in the skin and subcutaneous fatty tissue when used dermally and by phonophoresis].

During the combined use of the ointment with amphotericin B and ultrasound after 1, 3, 24, 48 and 72 hours there was detected a higher content of the drug in the skin and the subcutaneous fatty tissue as compared with the epicutaneous application of the ointment without ultrasound. The drug contents in the insonified and non-insonified samples increased by the 3rd hour after the application except the insonified skin, where the maximal values were noted after 1 hour; subsequently (after 24, 48 and 72 hours) amphotericin B concentration gradually decreased. It remained significantly higher in the insonified samples than in the non-insonified ones. The data ground the expediency of using the ointment with amphotericin B in combination with ultrasound in candidosis that is one of the ways of increasing the effectiveness of treatment of this disease.
