Whisky: a new provacative test for calcitonin secretion.

Oral whisky is a potent stimulus of calcitonin secretion. Peak increments of immunoreactive calcitonin are observed within 15 min after the ingestion of 50 ml of whisky; the magnitude of the response is similar to that observed during a four-hour calcium infusion. This procedure has several advantages over standard methods of stimulating calcitonin release in patients at risk of developing medullary carcinoma of the thyroid, and this is shown by a study in a large family with familial chromaffinomatosis.

Calcitonin in breast and lung cancer.

Immunoreactive calcitonin was found in extracts of seven out of eight consecutive breast carcinomas and four selected lung carcinomas, but not in extracts of benign breast lesions or normal tissues. This suggests that the high plasma calcitonin levels observed in patients with a wide variety of cancers reflect ectopic production of calcitonin by cancer tissue, and that the radioimmunoassay for calcitonin could prove to be of value in the detection and management of malignant disease, particularly breast cancer.

Separation of phenylthiohydantoin-amino acids by high-pressure liquid chromatography.

A method for the separation of all phenylthiohydantoin (PTH)-amino acids except PTH-arginine and PTH-histidine by high-pressure liquid chromatography on a silica column is described. Elution is performed with a concave solvent gradient from hexane-methanol-propanol (3980:9:11) to methanol-propanol (9:11). A complete run is achieved in 40 min with a pressure drop of 1000 p.s.i. over the 250 mm X 2.1 mm column. Eluted peaks of 2-5 nmole are easily detected by their ultraviolet absorption at 254 nm. This method is superior to existing gas-liquid and thin-layer chromatographic techniques since all PTH-amino acids except PTH-arginine and PTH-histidine may be both separated and quantitated in a single run of 40 min. The use of the technique in conjunction with an automated peptide sequence analyser is illustrated.

Feedback regulation of 25-hydroxycholecalciferol metabolism by vitamin D3.

1. In vitamin D-deficient chicks both vitamin D3 and 1alpha-hydroxycholecalciferol markedly decrease renal 1-hydroxylase activity and induce 24-hydroxylase activity. 2. Actinomycin D abolishes both effects. 3. These results are consistent with feedback regulation of vitamin D3 metabolism by a direct nuclear action of the vitamin or its metabolites on the kidney cells.

Calcitonin, parathyroidectomy and modelling of bones in the growing rat.

After calcitonin injection, parathyroidectomy, or both, in young growing rats, broadening of the proximal femoral metaphysis with lack of normal concavity of the medial and lateral contours was observed radiographically. Histologically, this abnormal modelling was associated with retarded osteocytic osteolysis. Further manifestations of decelerated resorption included retention and extension of the chondroid core in the secondary spongiosa, retention of cartilage in the cortex and a large increase in the number of cementing lines.
