Characterization of anticholinesterase-active 3-alkylpyridinium polymers from the marine sponge Reniera sarai in aqueous solutions.

From the marine sponge Reniera sarai 3-alkylpyridinium oligomers and polymers have been isolated. 3-Alkylpyridinium polymers are potent anticholinesterase agents; in addition, they show hemolytic and cytotoxic activities. Oligomers with a molecular weight lower than 3000 Da do not possess any significant activity. We report structural characterization of 3-alkylpyridinium polymers and their behavior in aqueous solutions. We found that biologically active polymers are composed of head-to-tail 3-alkylpyridinium units. According to MALDI-TOF spectrometry two species of polymers exist, the smaller with a molecular weight of 5520 Da and the larger with a molecular weight of 18,900 Da. Both polymers are soluble only in water, while low molecular oligomers are readily soluble in organic solvents. Polymers form large water-dissolved supramolecular structures with an average hydrodynamic radius of 23 +/- 2 nm and, therefore, cannot be separated with size-exclusion chromatography.

[The oxidative burst in the determination of immune response to synthetic materials].

Macrophages play an important role at the implant-tissue interface. Their activity is extremely complex but above all they are phagocytic cells. Phagocytosis is accompanied by a sequence of biochemical reactions known as "the oxidative burst" (BO). The fundamental physiological significance of this phenomenon is to provide the phagocytes with an efficient microbicidal mechanism. Our aim was to prepare an objective method for screening biomaterials that could influence macrophagic defence mechanisms against a possible implant contamination.

[Influence of suture materials on cell viability: a comparative experimental study].

Among the conditions that a material must satisfy to be considered biocompatible, most important is the non negative influence on the cellular viability of the surrounding tissue (cytocompatibility). The aim of our work was to evaluate in vitro, whether different types of materials (usually used in surgery), alter the viability parameters or the morphological characteristics of the cultured cells in contact with them.

Aneurysmal bone cysts of the pelvis.

Twenty-three cases of pelvic aneurysmal bone cysts treated at the Istituto Ortopedico Rizzoli were reviewed after a mean follow-up of 7 years. Eighteen cysts involved the anterior arch, four extended into the iliac wing and the anterior arch, and one invaded the entire hemipelvis. The acetabulum was involved in 56.5% of the cases. Fourteen patients were treated with surgery (curettage 11; resection 3), and five with radiation therapy; two patients had both modalities; two additional patients refused any treatment after biopsy. The overall recurrence rate was 13% (one case after curettage, one after radiation therapy, and one after combined treatment). Significant complications affected the final functional result in four of seven patients who received radiation therapy, while only one minor complication was seen in the surgical group.

Prognostic significance of serum lactic acid dehydrogenase in Ewing's tumor of bone.

Pretreatment serum lactic acid dehydrogenase (LDH) levels were examined in 82 patients with Ewing's tumor of bone (23 metastatic and 59 localized). The percentage of patients with increased serum LDH levels was significantly higher in the metastatic group than in the group of patients with localized disease (87 vs. 45%). After treatment with combined therapy, in this latter group the recurrence rate was significantly higher in patients with elevated serum LDH levels at admission than in those with normal serum LDH levels (50 vs. 9%). Of the 16 patients with localized disease who relapsed, 13 (81%) had elevated serum LDH levels at the time of recurrence. These results indicate that in Ewing's tumor serum LDH levels may serve as useful prognostic markers and could be valuable in monitoring the effectiveness of combined therapy.

[Lupic nephropathy in childhood: morphologic analysis of 18 cases].

Serum IgE concentration was determined in 17 newborn and 171 children with ages between 1 and 12 months. In the first group, blood was taken from the umbilical cord, and in the second, we used a periferic blood sample. The selection implied the exclusion of children with any suspicion of atopic disease, family history of atopy and recent viral infection. The serum IgE concentration was determined by enzyme linked immunoabsorbent assay (PRIST). The sex, race ( white and non white) and age were analyzed in the children studied. During a follow-up period (24 months) seven children (3,7%) developed atopic symptoms. The serum IgE concentration of the groups studied was higher than in other studies. The mean IgE in each group was as follows : newborns = 0.24 IU/ml; 1-3 months = 1.57 IU/ml; 4-6 months = 7.72 IU/ml; 7-9 months 12.07 IU/ml; 10-12 months = 12.14 IU/ml.

Ultrastructure of CD57+ cells isolated from human tonsils or blood.

The presence of CD4+, CD57+ T cells in the germinal centers has been reported by several authors. The CD57 antigen is also expressed by natural killer (NK) cells. We purified CD57+ cells from human tonsils and blood by microdissection, rosetting with sheep red blood cells and magnetic cell sorting (MACS) and examined the ultrastructural morphology of these cells. Clear differences were found in cell aspect: blood NK contained large granules which were not found in the tonsillar CD57+ cells. These latter appeared medium-sized and not fully activated. After immunolabeling, the tonsillar CD57+ cells were mainly found in the light zone of the germinal centers.
