[Effect of mononuclear phagocytes on the differentiation of syngeneic, allogeneic and xenogeneic hematopoietic stem cells].

The colony formation in spleen of lethally irradiated syngeneic or hybrid recipients was studied after transplantation of bone marrow cells, with or without macrophages from lymph nodules or from peritoneal cavity of mice, cells of macrophage-like cell line J-774, and monocytes from peripheral blood of healthy donors. The direction of stem cell differentiations in the presence of all the types of mononuclear phagocytes was seen to change from mainly erythroid to mainly myeloid one. The ratio of erythroid to myeloid colonies became equal to 0.5-0.9 instead of 2.0, when bone marrow cells were injected with equivalent quantity of mononuclear phagocytes. This new regulatory function of mononuclear phagocytes is discussed.

[Changes in the leukocyte phagocytic activity of donor blood after its UV irradiation. II. Simulation of the effect of the autotransfusion of UV-irradiated blood].

The UV-irradiated blood of healthy adults was supplemented with non-irradiated blood in the ratio 1:10. The phagocytic activity (PhA) of monocytes and granulocytes was seen to increase markedly in the whole mixture of blood. In this case the rise of PhA was pronounced 1.4-1.7 times as much as in the case of the non-supplemented, directly UV-irradiated blood. The enhancement of PhA depends on its initial level and may occur simultaneously with structural changes of the cell surface components. It seems reasonable to propose that PhA stimulation may be one of the earliest mechanisms in immunocorrection by UV-irradiated blood therapy.

[Changes in the leukocyte phagocytic activity of donor blood after its UV irradiation. I. Its relation to the irradiation dose and initial level of phagocytic activity].

Phagocytic activity of human mono- and granulocytes increased markedly after UV blood irradiation in the apparatus "Izolda" used in hospitals of the USSR for medical treatment. With the rise of irradiation dose the ratio of cells ingesting latex particles increased, although the average number of particles ingested per cell decreased. The integrative phagocytic index poorly depended on the irradiation dose. In patients with a low initial level of phagocytic index, after UV blood irradiation it became more pronounced than in those with the initial elevated level. The enhancement of phagocytic activity is the result of a direct UV-stimulation of cells. This stimulation not mediated by irradiated blood plasma is known to inhibit the phagocytic activity of leucocytes. A possible mechanism of phagocytic activity stimulation is discussed.

[Dynamics of change in the antibacterial activity of mouse peritoneal cells during the development of experimental influenzal infection].

The method of the radiometric determination of the antibacterial activity of peritoneal cells in the process of the development of viral infection has been approved. The genetically determined specific level of the antibacterial activity of these cells in different strains of mice has been established. The development of the lethal form of experimental influenza A in mice has been shown to be accompanied by the mobilization of nonspecific protective factors, including the system of interferons and the antibacterial activity of peritoneal cells. The latter factor is, seemingly, insufficient for ensuring the necessary level of protection in the animals, whose death occurs at a shorter time than that necessary for the development of specific immune response.

[Dynamics of the dermograms in patients with focal pneumonia].

Dermograms were examined over time in patients with focal pneumonia running an acute, protracted and unfavourable course. Prognostically significant differences in the dermograms were revealed in patients of the three groups. Attempts were made to correct the deficiency of the migration of immunocompetent cells to the cutaneous window in patients with pneumonia taking an unfavourable course.

[Use of lipostabil to correct lipid metabolism disorders in patients with ischemic heart disease].

A comparative study of biochemical and immunologic patterns showed lipostabil treatment to be effective in less than half of the patients with unstable angina. A method is developed for in vitro testing of individual blood mononuclear sensitivity to lipostabil effects. The proposed set of biochemical and immune assays allows the selection of patients subject to lipostabil treatment and provides additional criteria for the assessment of its efficiency.

[Dependence of the migration activity of blood granulocytes from healthy donors and from bronchial asthma patients on the presence of cellular mediators].

The migration activity of granulocytes in human blood has been shown to depend both on the locomotor properties of these cells and on the presence of cell mediators secreted by blood mononuclears and by granulocytes themselves. Granulocytes in the blood of patients with the atopic form of bronchial asthma differ from granulocytes in healthy donors by sharply decreased spontaneous migration activity. Granulocytes and mononuclears in the blood of patients with the atopic form of bronchial asthma differ from the corresponding cells of donor blood in the activity of cell mediators secreted by them and influencing the migration of granulocytes in the donors.

[Activation of glucose-6-phosphate dehydrogenase in mouse peritoneal macrophages by sera from patients with ischemic heart disease].

The purpose of this study was to compare the in vitro metabolic changes in mouse peritoneal macrophages exposed to sera of CHD patients and healthy donors. Oxidative (metabolic) burst was estimated by the activity of one of the limiting enzymes of hexose monophosphate shunt--glucoso-6-phosphate dehydrogenase. Pronounced enhancement of glucoso-6-phosphate dehydrogenase activity accompanied the exposure of macrophages to sera of patients with acute myocardial ischemia during the first two days after the onset of anginal pains. The correlation was established between the activity of creatine kinase in the sera of patients with acute myocardial infarction and the ability of these sera to enhance glucoso-6-phosphate dehydrogenase activity in mouse peritoneal macrophages.

[Mononuclear cell reactions in the combined treatment of patients with a cutaneous melanoblastoma of the lower extremities].

Changes in lymphocyte and monocyte counts in peripheral blood of patients who had undergone combined treatment for malignant cutaneous melanoma of low extremities were evaluated. The posttreatment counts were compared with those for mononuclear cell infiltration of tumor stroma. Mononuclear cell levels in blood and in tumor stroma versus patients' survival time were analysed. A moderate correlation between lymphocyte and monocyte (macrophage) levels in tumor stroma, on the one hand, and survival time, on the other, was established.

[Effect of the secretion products of mononuclear phagocytes on the proliferative activity of connective tissue cells].

3H-thymidine uptake was used to study the effect of the media conditioned by murine peritoneal macrophages and human peripheral blood monocytes on proliferative activity of 3T3 fibroblasts, smooth muscles of the umbilical vein and bone marrow fibroblasts. Secretory products of murine macrophages and human blood monocytes inhibited the proliferation of all 3 types of target cells. Latex phagocytosis by macrophages led to the increase in the inhibitory activity of the media conditioned by macrophages, whereas upon latex phagocytosis by monocytes the inhibitory action of the conditioned media was unchanged. Addition of rodexman to monocyte cultures did not produce any effect on the inhibitory action of the conditioned media. Activation of monocytes by lipopolysaccharide resulted in the diminution of the inhibitory action of the monocyte-conditioned media only with respect to smooth muscle cells. Lipopolysaccharide did not affect the action of the media conditioned with macrophages.

[Participation of products of macrophage origin in regulating monocytopoiesis].

Products secreted by mouse peritoneal macrophages phagocytizing latex particles in monolayer culture in vitro were found to induce monocytosis in vivo. They inhibited mouse bone marrow colony formation and simultaneously stimulated the accumulation of immature macrophages in semisolid agar culture. The addition of these products to the liquid bone marrow culture resulted in reduction of proliferative activity of non-adherent cells and in increase of proliferative activity of adherent cells as revealed from 3H-thymidine uptake. The possible role of the products secreted by macrophage in feedback regulation is discussed.

[Macrophage activation by polysaccharides from yeast-like fungi].

The effect of polysaccharides isolated from yeast-like fungi on diverse activities of murine peritoneal macrophages was studied in experiments in vitro. Yeast polysaccharides had a nonspecific activating effect on macrophages in tissue culture. They enhance the adhesion of macrophages to glass, stimulate phagocytosis and intracellular digestion, and change the chemotactic activity. The activity of polysaccharides directly correlates with their molecular weight. There is a relationship between the biological activity and chemical structure of polysaccharides. The beta-structural polysaccharides are capable of activating the functions of macrophages, while beta-structural dextrans do not possess such properties.

[Improvement in the functional and morphological mononuclear blood cell indices under the influence of miscleron].

Taking into account the conception of metabolic immunosuppression, arising in the case when fatty acids but no glucose serves as the basic energetic substrate, the authors have studied the effect of miscleron (clofibrate, athromid-S) on a number of hormonal-metabolic and immune indices. It is shown that miscleron in addition to decreasing the blood level of cholesterol, beta-lipoproteids, triglycerids, free fatty acids, cortisol and reducing 17-ketosteroids excretion, produces higher indices of the blasttransformation reaction, induced by FHA, an increased phagocytic index of monocytes, a rise in the level of peroxidase and lysosomal activity of monocytes. Miscleron and other substances lowering the lipid level in blood should be examined in patients with nonlymphoid types of tumors both with the aim to eliminate metabolic immunosuppression and to retard the rate of tumor cells growth due to a reduced level of low density lipoproteins.
