[Effect of antilymphocyte globulin on growth and erythroid differentiation of K-562 cells].

The influence of antilymphocytic globulin (ALG) on the proliferative activity and erythroid differentiation of erythromyeloid K-562 cells was studied. It has been established that ALG produces a dose-dependent inhibiting effect (20-150 micrograms of ALG/ml) on the proliferative activity of K-562 cells and induces erythroid differentiation of these cells that does not depend on the concentration of ALG used in the test. Pretreatment of K-562 cells with ALG increases their sensitivity to the following growth-inhibiting action of cytosine-arabinoside.

[Changes in the functional properties of NK-cells and the cytostatic activity of granulocytes in acute leukemia patients].

Effector activity of natural killers (NK) was studied, and functional state of NK and granulocytes was evaluated in 23 normal subjects and 28 acute leukemia patients (12 of them being in clinico-hematological remission up to 2 years). In acute leukemia significant disorders are observed in the system of nonspecific antitumor defence that is evidenced by a drastic decrease of NK activity due to changes in the functional properties of NK--their capacity for producing conjugates and blast cell lysis, low sensitivity to exogenous interferon, and lowered spontaneous cytostatic of granulocytes.

[Functional changes in the natural killer cells of patients with acute leukemia].

The results of study of NKC activity (51Cr release assay, conjugate-forming and lytic activity) in healthy persons and patients with acute leukemia (AL) in active phase and in remission are presented. It has been shown that the depression of the NKC activity in acute phase of AL (6 times as low as in norm) and in remission (3 times as low as in norm) depended upon disturbances of the processes of conjugate formation, lytic and recycling activity. Correlation has been found between percentage of lytic conjugates and that of cytotoxicity. The increase of NKC activity in remission seemed to be due to tendency to normalization of functional state of NK-cells (conjugate-forming and recycling activity).

[Low-molecular weight nuclear RNA in the lymphocytes of healthy persons and chronic lympholeukemia patients].

Low-polymer nuclear RNA (1) of lymphocytes was studied comparatively in healthy individuals and patients with lympholeukemia. In 7.5% polyacrylamid gel both in normal lymphocytes and their leukemic analogs 9 fractions were identified which possess similar electrophoretic mobility. The correlation of separate fractions 1 in pathological lymphocytes was shown to differ from that in lymphocytes of healthy donors. The study of the metabolic activity 1 indicated that one of the fractions (D) is metabolically inert under conditions of a 1-hour label, while in normal lymphocytes one fraction was revealed, whereas in chronic lympholeukemia--two fractions being recorded only by their radioactivity.

[Nuclear and cytoplasmic poly A-containing RNA in normal and leukemic human leukocytes].

The method of affinity chromatography on a poly(A)-cellulose column was used to study the content and radioactivity of poly(A)-rich RNAs in the nuclei and cytoplasma of lymphocytes of healthy subjects and patients suffering chronic lymphatic leukemia. It was found that in the nuclei of normal and leukemic lyphocytes the amount of polyadenylated RNAs was higher than that in cytoplasma. The content of poly(A)-rich RNAs in the normal lymphocytes nuclei was 6.7%, whereas in leukemic ones--4.3% of the total nuclear RNA, in the latter, however, the specific radioactivity of polyadenylated RNAs was 1.5 times as that in normal cells. Poly(A)-containing RNAs normally make up as much as 3.1% of the total amount of cytoplasmic RNA, while in leukemic lymphocytes--1%. The specific radioactivity of poly(A)-rich RNAs in normal and leukemic lymphocytes cytoplasma was on average 3 times higher than in other classes of RNA.

[Characteristics of poly(A) rich RNA in human lymphocytes under normal conditions and in chronic lympholeukemia].

The presence of poly(A) containing RNA bound specifically to millipore filters, adsorbed on poly(U) cellulose and resistant to the pancreatic RNAs effect has been demonstrated in total RNA of human lymphocytes. These fractions show a DNA-like nucleotid content, a rapid turnover and they are characterized by sedimentation constants within the range of 9.5--15 S. On the average, 24.6 per cent of the applied initial radioactivity of total RNA is absorbed on membrane filters of normal lymphocytes, whereas from 13.3 to 23.3 per cent- in the poly(U) cellulose column. The study of lymphocytes total RNA in patients with chronic lympholeucosis has indicated a great heterogeneity in the content of poly(A) sequences rich RNA fractions.

[Phosphoribosyl pyrophosphate and its metabolic enzymes in the erythrocytes in certain forms of anemia].

Content of phosphoribosyl pyrophosphate and the activity of ribose phosphate pyrophosphokinase and AMP-pyrophosphorylase were studied in erythrocytes of healthy persons and of patients with various types of anemia. Deficiency of phosphoribosyl pyrophosphate and a decrease in the activity of enzymes studied were observed in erythrocytes under microspherocytic and hypoplastic anemia. The alterations correlated with impairments in energy metabolism. At the same time an activation of phosphoribosyl pyrophosphate enzymatic system and an increase of its content in erythrocytes did not depend on energy metabolism in Marchiafava--Micheli disease.

[Age composition of the erythrocyte population, ATP and 2,3-diphosphoglycerate composition in erythrocytes in different forms of hemolytic anemia].

The total erythrocyte population was subdivided into three age groups using as criteria the stability of red blood cells to osmotic shock and the values of glucose-6-phosphate dehydrogenase (G6PD) activity. The "old" erythrocytes constituted about 8%, "mature"--87% and "young"--5% under normal conditions. In hereditary microspherocytosis 70% of erythrocytes possessed a decreased osmotic resistance and higher G6PD activity as compared with normal state. Two categories of erythrocytes was found among "young" cells in Marchiafava-Micheli disease; one of them possessed normal osmotic resistance, the other--decreased osmotic resistance. The age composition of the erythrocyte population was normal in functional hyperbilirubinemia. Content of ATP was decreased in erythrocytes of patients with hereditary microspherocytosis, it was increased in Marchiafava-Micheli disease and approached the normal level in patients with functional hyperbilirubinemia. Concentration of 2,3-diphosphoglycerate was similar to the normal level in erythrocytes of patients with functional hyperbilirubinemia; an inverse correlation was observed between contents of hemoglobin and of 2,3-diphosphoglycerate in blood of patients with hereditary microspherocytosis and with Marchiafava-Michel disease.
