Structurally designed novel furogamma lactams as inhibitors for bacterial propagations.

Some novel furogamma lactams have been synthesised by one step condensation of arylaminomalonates with substituted furyl acryloyl chlorides. The annulation of substituted monocyclic gammalactams followed by cyclization produced novel tricyclic furogamma lactams. Some of these furogammalactams are found to exhibit Gram-positive and Gram-negative antibacterial activity at very high concentrations.

To the mechanism of spermine-FBS cytotoxicity toward K562 human myelogenous leukemia cells.

The effects of several compounds acting through adenylate cyclase system and/or influencing prostaglandin biosynthesis on spermine-FBS cytotoxicity to human myelogenous leukemia K562 cells were studied. Salbutamol, a beta 2-adrenoceptor agonist inhibited to a certain extent spermine-FBS cytotoxic action to K562 cells, and propranolol, a beta 2-adrenoceptor antagonist, did not affect this inhibition. Aminophylline, an inhibitor of cyclic nucleotide phosphodiesterase, acted suppressing spermine-FBS cytotoxicity to K562 cells. Pretreatment of the cells with dexamethasone did not significantly alter salbutamol-related inhibition of spermine-FBS cytotoxicity. Indomethacin, an inhibitor of cyclooxygenases directly involved in prostaglandin biosynthesis, did not interfere with protective terbutaline effects against spermine-FBS cytotoxicity to K562 cells during the 24-hour period.

[The effect of hormones on tumor growth].

It is well documented that estrogen influences the growth of mammary carcinoma. Estrogen-induced response of target tissue includes protein-biosynthesis as well as stimulation of cell proliferation. Estrogen stimulates tumor cell proliferation in vitro in a dose-related manner by shortening the cell cycle time primarily at the G1 duration and by increasing the number of dividing cells per cycle. The mechanisms of estrogen action on target cell proliferation are not understood well and because of controversial experimental results the role of functional estrogen receptors in estrogen-induced proliferation is not yet clear. In this work, we studied the effect of estrogen on the growth of cells derived from primary tumors (N = 4) and malignant pleural effusions (N = 6) from patients with histopathologically verified breast cancer. We correlated steroid receptor content of malignant tissue used with results obtained for estrogen-induced cell proliferation. It was shown that there was a positive correlation between steroid receptor content and effect of estrogen on cell proliferation in most of the cases studied (7/10). Our study also confirms the possibility of positive and negative influences of estrogen on modulation of tumor cell growth.

[In vitro culture of cells from patients with leukemia].

The development of cytological, cytochemical and immunological investigations of leukemic cells as well as new cytostatic drug combinations have led to great progress in the diagnosis and treatment of acute leukemia (AL). However, a heterogeneity in response to the same kind of therapy has been noted in patients with the same cytological type of the disease. This points out the importance of investigations of leukemic cells in culture which should provide more data on proliferation and reactivity of these cells. The population of tumor cells in heterogeneous in morphological and biochemical properties, in proliferative capacity and other characteristics. In cell culture it is possible to investigate the growth of cells and the effects of various agents on normal and malignant cells. In this study we investigated the growth of cells from bone marrow of patients with acute leukemias and acute phase of chronic leukemias. Cells from 17 patients were studied. The patients were from Internal Clinic A in Belgrade and the investigations were performed before treatment. Bone marrow cells were cultured in vitro on plastics and growth of the adherent cells was followed in the course of 2-3 weeks. The results show that bone marrow cells adhered and proliferated in 12/17 patients, there was low proliferation in 4/17 and none in 1/17. It is shown that in these conditions a successful growth of fibroblast-like shaped cells can be obtained with the bone marrow cells, but it is still not possible to correlate the course of the disease and the proliferation in vitro.(ABSTRACT TRUNCATED AT 250 WORDS)

Lymphocyte reactivity in Imuran-treated guinea-pigs and in vitro effect of colchicine.

Imuran treatment significantly diminished macrophage migration inhibition in immunized guinea-pigs and the percentage of E-rosette forming cells in all treated animals. 10(-6) M colchicine in vitro significantly increased the percentage of E-rosette forming cells in Imuran-treated animals. The results suggest possible alteration of the lymphocyte receptors after treatment with Imuran.
