[The effect of sclareol lactone and sclareol glycol on artificially induced lung metastases of Lewis lung carcinoma (a preliminary report)].

The prophylactic effect of tetralabdanes, obtained by chemical decomposition of the natural diterpene sclareol (Il. Ognianov and T. Somleva) on the growth of artificially induced lung metastases was studied. After intravenous administration of 25 mg/kg of sclareol-lacton 30 minutes before the transplantation of tumorous cells of Lewis [correction of Luis] lung carcinoma the number of metastases was reduced with 37.5% but in a dose of 50 mg/kg from--33 to 63%. In a dose of 100 mg/kg of sclareol-lacton metastases were increased with 2--7%. Sclareolglycol administered in a dose of 25 mg/kg under the same experimental conditions, reduced lung metastases with 38%, but in a dose of 50 mg/kg--from 26% to 61%. Its administration in a dose of 100 mg/kg stimulated their formation with 62%.

[M-echocardiographic determination of left-ventricular diastolic noncompliance, diastolic wall stress and the rate of relaxation of the circumferential muscle fibers].

New noninvasive contemporary indices for assessment of left ventricular diastolic function are presented. They can be obtained by the simultaneous recording of the M-echocardiographic left ventricular dimensions and the apexcardiogram. These indices are: left ventricular diastolic compliance, left ventricular wall stress and the velocity of the circumferential fibre shortening. These indices are statistically significantly changed to a greater extent in hypertrophic cardiomyopathies and to a lesser extent in ischemic heart disease and arterial hypertension and after nifedipin treatment they improve considerably. The left ventricular diastolic wall stress was studied in both types of left ventricular load: by volumetric load a twofold increased stress of the wall was registered while by tension load the stress of the wall was almost normal.

[Experiment to potentiate the effect of cyclophosphamide combined with tempidon and caffeine in leukemia L1210 and Lewis hepatic carcinoma].

The authors carried out experiments on mice DBA/2 and BDF1 with leucosis L1210 and lung carcinoma of Lewis and found that psycotropic preparations tempidone and caffeine did not manifest antitumorous effect and toxicity after the used doses and scheme of treatment. Tempidone enhanced antitumorous action of small and mean doses of cyclophosphamide in mice with leucosis L1210. In the triple combination (tempidone, caffeine and cycl phosamide) the caffeine enhanced this action of tempidone without raising toxicity in mice with leucosis L1210, but not in mice with lung carcinoma of Lewis.

[Effect of talliblastine on several hematologic indices].

The authors carried out a study on the blood formation of healthy experimental animals after treatment with the potential antitumours agent taliblastine in view of clarifying its influence. The experiments were undertaken on inbred mice DVA/2 and rats on the Wistar strain. Taliblastine was administered intraperitonealy: in ice-aingly and in a 5-day course, but in rats only in a 5-day course. The substance was administered after single treatment with doses of LD50 maximal threshold limit dose (MTD), but in the dourses with MTD and 1/2 of MTD. They examined the changes in hemoglobin, number of leucocytes, erythrocytes, thrombocytes and reticulocytes, but in rats-the number of bone-marrow cells as well. The values of hematological indices were determined 24 hours, 10 and 30 days after conclusion of the administration of taliblastine. It was established that taliblastine did not affect substantialy the values of hemoglobine and caused transitory changes in the absolute number of leucocytes and lymphocytes.

[Influence of taliblastin and vinblastine on serum agglutinins determined by 2 methods].

The authors examined the influence of the potential antitumorous preparation taliblastin and the well known cytostatic vinblastin on the formation of serum agglutinins in mice. Two methods were used--the classical reaction of agglutination and the method for inhibition of passive migration of erythrocytes (IPM) described by Schweicer and Gilisen (1974). It was established that vinblastin markedly inhibited the formation of specific agglutinating antibodies, while taliblastin was considerably weaker immunosupresor; The method of IPM was more sensitive than the classical reaction of agglutination and gave a possibility for better determination of immunosuppresive action of cytostatics.

[Toxicological characteristics of thaliblastine[].

The toxic action of the antiphlogistic drug thaliblastin, containing 93-95 per cent of the alcaloid thalicarpine was investigated. With its single intraperitoneal injection to mice the DL10 of the drug amounts to 273-306 mg/kg, DL50-325-350 mg/kg and DL90-370-402 mg/kg. With its 5-day long administration the respective figures for mice are 102-165, 125-201 and 154-243 mg/kg and for rats--170, 203 and 272 mg/kg. The respective figures for a 20-day long introduction to mice are 155, 185 and 220 mg/kg. Introduction of thaliblastin in doses of DL10 single and multiply results in a marked loss of the mass in mice.
