[Tocopherol-binding proteins in membranes of rat liver mitochondria].

The use of the adsorption chromatography on the hydroxyl apatite makes it possible to yield and partly purify the triton X-100-solubilized mitochondrial protein fraction of the rat liver able to bind specifically [3H]-alpha-tocopherol. The method permits removing simultaneously both free detergent and [3H]-alpha-tocopherol from the protein mixture without disturbance of the established equilibrium. When compared with methods used for the removal of free hydrophobic ligands in the in vitro binding experiments, the applied method is the most effective.

[Effect of vitamin E and ubiquinone-9 on activity of ubiquinone-dependent enzymic systems of rat liver mitochondria].

It is shown that the activity of succinate- and NADH-ubiquinone-reductase systems as well as the ubiquinone content in the liver mitochondria of E-hypovitaminotic rats decrease as compared to their level in the liver mitochondria of healthy animals. Under conditions of E-hypovitaminosis alpha-tocopheronolactone and ubiquinone-9 3 h after intravenous injection to rats produce a pronounced normalizing effect on the ubiquinone content and on the activity of ubiquinone-dependent enzymic systems. It is assumed that a positive effect of vitamin E and its metabolite on the activity of these enzymic systems is to a considerable extent connected with an increase in the ubiquinone content in the rat liver mitochondria.

[Possibility of preventing muscular dystrophy in E avitaminosis with alpha-tocopheronolactone and diludin].

The efficacy of pharmacopeial alpha-tocopheryl acetate, alpha-tocopheronolactone and the antioxidant diludin was studied as to the possibilities of preventing E-hypovitaminosis muscle dystrophy in rabbits. alpha-Tocopheronolactone action was similar to that of pharmacopeial alpha-tocopheryl acetate as regards E-vitamin activity that was manifested by an increase in the body mass, decrease in the urine creatine index and maintenance of normal ubiquinone content, as well as of succinate-ubiquinone-reductase, NADH-ubiquinone-reductase and NADH-cytochrome c-reductase activity of rabbit skeletal muscle and heart mitochondria. On the contrary , as a result of the antioxidant administration, no direct correlation was found between ubiquinone content and activity of ubiquinone-dependent enzymatic systems of mitochondria. The data obtained attest to high E-vitamin activity of alpha-tocopheronolactone. The action of the latter was disclosed to be different from that produced by the antioxidant.

[Determination of adenine nucleotide and inorganic phosphate contents of liver of rats with E-hypovitaminosis and administration of alpha-tocopherol and ubiquinone-9].

It is shown that in the rat liver with E-hypovitaminosis the ATP content, sum of adenine nucleotides, and the phosphate potential value ([ATP]/[ADP.]Pi) decrease, whereas the amount of ADP, AMP and Pi remains practically unchanged. 3h after intravenous administration of alpha-tocopherol of ubiquinone to rats with E-hyovitaminosis the mentioned indices normalize, which evidences for a considerable increase in the level of bioenergetic processes. The direction of the mentioned changes corresponds to changes in the ubiquinone content and activity of the respiratory chain redox enzymes. The participation of vitamin E in processes of oxidative phosphorylation may be connected with the metabolism control and ubiquinone functioning.

[Efficiency of ubiquinone and p-oxybenzoic acid in prevention of E-hypovitaminosis-induced development of muscular dystrophy].

It is shown that E-hypovitaminosis-induced muscular dystrophy in rabbits is accompanied by a sharp decrease in the body mass, an increase in the urine creatine-index, a decrease in the vitamin E and ubiquinone contents in the liver and skeletal muscle tissues. In the myocardium mitochondria a decrease in the vitamin E content and an increase in the ubiquinone content are observed. The activity of NADH-cytochrome c-, NADH-ubiquinone- and succinate-ubiquinone-reductase also varies in mitochondria of the studied tissues. In myocardium organellas a direct dependence is found between the content of ubiquinone, NADH- and succinate-ubiquinone-reductase activity and an inverse one-between its content and the activity of the NADH-cytochrome c-reductase system. It is established that p-oxybenzoic acid as well as vitamin E prevents development of muscular dystrophy and causes changes analogous in direction in the activity of the ubiquinone-dependent enzymic systems of mitochondria. Ubiquinone-9 is less efficient in preventing the development of muscular dystrophy.

[Energy metabolism in newborn calves].

It has been studied that peculiarities of redox processes of glycolysis and of citrate cycle in newborn calves under oxygen shortage has been caused by diarrhea under acute disturbances of digestion. It was shown that concentration of oxalo-acetate, L-ketoglutarate, glutamate increases and concentration of malate lactate, pyruvate decreases when partial pressure of oxygen rises in the venous blood of calves. Change of activity succinate dehydrogenase depends on the degree of pathological process.

[Effect of tocopherol, its analogs and the antioxidant ionol on lipid peroxidation in vitro].

Ionol, alpha-tocopherol and its analogues (tocoferenquinones with C6 and C1 in the side chain, tocopherol without the side chain and two tocopherols with shortened to C6 side chain both with saturated and with unsaturated bond at the end) were studied for their effect on the formation rate of malonaldehyde in the tissue and liver mitochondria of vitamin E-deficient rats in vitro. Preparations were used in two concentrations: 35 mg per 1 ml of the medium--in a dose suppressing processes or lipid peroxidation; and 5 mg per 1 ml of the medium--in a dose activating biosynthesis of ubiquinone in the animal tissues. Tocopherol effect on lipid peroxidation processes in the liver tissue and mitochondria is shown to be different and to depend on its concentration in the medium. Ionol is efficient only in high doses under the conditions in vitro. Tocopherol analogues (their side chain being shortened to six carbon atoms) display maximum activity when they inhibit lipid peroxidation both in the liver tissue and mitochondria in the both concentrations under study which supposes their high efficiency under the conditions in vivo.

[Incorporation of 1-14C-acetate into unsaponifiable lipids of slices of regenerating rat liver].

Incorporation of 14C into unsaponifiable lipids, sterols and their presursors, hydrocarbons, isolated from sections of the rat regenerating liver studied after their 6-hour incubation with 1-14C-acetate. Intensity of the label incorporation into unsaponifiable lipids and sterols increases significantly the first three days after hepatectomy and decreases on the 15th day, intensity of incorporation into hydrocarbons rises significantly the first six days and lowers on the 15th days after hepatectomy. The label is observed to be incorporated more intensively into the total unsaponifiable lipids and hydrocarbon fraction as compared to sterols, that may evidence for activation of these compounds biosynthesis at initial stages.

[Anti-cataract activity of a vitamin E analog].

Anti-cataract activity of vitamin E analog with shortened side chain--2'-4'-methyl-pentenyl-acetoxy-2,5,7-tetramethylchroman has been studied. It is shown on the model system that the analog of vitamin E inhibits the increase of the lens agent fluorescence under irradiation of its homogenates by polychrome light. Taking no negative effect on the lens capsule epithelium, the drug normalizes the content of vitamin E in the blood and increases activity of Na+, K+, ATPase in the cortex and capsule of rabbits lenses in dynamics under simulation of light cataract in vivo.
