Cyclosporin A and its nonimmunosuppressive analogue N-Me-Val-4-cyclosporin A mitigate glucose/oxygen deprivation-induced damage to rat cultured hippocampal neurons.

When mouse hippocampal neuronal cultures, 2-3 weeks in vitro, were transiently exposed to combined glucose and oxygen deprivation (100% argon, 5% CO2, in glucose-free medium) for 90 min, extensive neuronal degeneration had occurred after 24 h of reoxygenation. When these cultures were preincubated with cyclosporin A, a calcineurin inhibitor and a blocker of the mitochondrial permeability transition, neuronal death diminished by 30-50%. Similarly, the cyclosporin A analogue, N-Me-Val-4-cyclosporin A, a potent blocker of the mitochondrial permeability transition with no significant calcineurin blocking activity, decreased cell death by 70-80%. Both cyclosporin A and N-Me-Val-4-cyclosporin A markedly attenuated calcium-induced swelling of isolated mouse brain mitochondria by blocking the mitochondrial permeability transition. The potassium thiocyanate-stabilized binding of cyclophilin D to mouse brain mitochondrial membranes was completely prevented by cyclosporin A and N-Me-Val-4-cyclosporin A. Our results strongly suggest that the mitochondrial permeability transition is involved in oxygen/glucose deprivation-induced cell death in vitro. Cyclophilin D and other components of the mitochondrial permeability transition may be important targets for neuroprotective and anti-ischaemic drugs.

[Pharmacokinetic studies of 5-nitrox in kidney failure patients].

The authors carried out initial pharmacokinetic studies of 5-nitrox on patients with compensated, first and second degree of decompensated renal insufficiency in view of determining dosage and intervals for application of the preparation. They found regular changes in the profile of the curves, manifesting the concentrations of 5-nitrox in plasma and urine for each interval of time-with advancement of renal insufficiency the peak levels diminished and were seen at later intervals as the course of the curves were of plateau character in patients with second degree of renal insufficiency. The analysis of the results showed that treatment of patients with decompensated renal insufficiency was not justified due to the retention of relatively high plasma level up to the 12th hour and rather low, unefficient concentrations of urine. 5-nitrox could be used in a single dose of 200 mg at 8-hour intervals in patients with compensated first degree of decompensated renal insufficiency.

[Pathomorphological studies of the testes in various forms of hypogonadism in young men].

The authors studied 32 biopsies of testes of patients, aged 18 to 26 with various forms of male hypogonadism: Klinefelter's syndrome--16, primary hypotrophy pogonadism with other known and unknown etiology--6, cryptorchism--1, secondary hypogonadism--4, later puberty--3. The materials were studied by the standard histological techniques. The authors report about definite structural changes with the various forms of hypogonadism, manifested in the elements of seminal tubules, Sertoli's cells, basal membrane of the ducts and Leydig's cells, situated in the interstitial spaces.
