Orosomucoid 1 and orosomucoid 2 types in the Taiwanese and Japanese: evidence for five new orosomucoid variants.

The genetic polymorphisms of orosomucoid, ORM1 and ORM2, were analyzed by isoelectric focusing on polyacrylamide gels and subsequent immunoprinting. Sera from 600 unrelated individuals in Taiwan and Japan were examined. Five new alleles, designated ORM1*7, ORM1*8, ORM1*4.2, ORM2*8 and ORM2*10, were observed together with common and rare alleles that have been found in the Japanese and the Filipinos. As compared with the Japanese, the Taiwanese have a higher frequency of ORM1*1, and a significantly lower frequency of ORM1*2.1.

Genetic polymorphism of human alpha 2HS-glycoprotein: characterization and application to forensic hemogenetics.

Human alpha 2HS-glycoprotein (AHSG) phenotypes were studied by isoelectric focusing of native and desialyzed samples followed by passive immunoblotting. At present a total of 15 different alleles could be distinguished. The usefulness of AHSG polymorphism in the field of forensic hemogenetics was demonstrated by markedly high variability at AHSG locus, reliability of phenotyping results and stability in blood stains.

Factor I (C3b inactivator) polymorphism among five populations in Eurasia.

Factor I (C3b inactivator) polymorphism in the Japanese (in Western and Southern Japan), Taiwanese, Nepalese and French was studied using isoelectric focusing on polyacrylamide gels. The exposure of passively blotted nitrocellulose membranes to glutaraldehyde vapor facilitated the subsequent immunodetection of a low concentration of factor I and permitted the reliable identification of the three phenotypes determined by two codominant alleles FI*A and FI*B. The data indicated a west-to-east genocline, ranging from France to Western Japan, in which FI*A changed from 0.006 to 0.120.

Genetic polymorphism of alpha 2HS-glycoprotein in a French population. Description of two new rare variants.

A French population was investigated for genetic polymorphism of alpha 2HS-glycoprotein (A2HS; nomenclature according to Human Gene Mapping 7, Los Angeles, 1983) using isoelectric focusing and immunoblotting. Three variants were observed together with two common alleles A2HS 1 and A2HS 2, whose frequencies were significantly different from the data in Canadians and Egyptians. An anodal variant to A2HS 1 was identical to a variant with two different nomenclatures reported by three different groups, indicating that there is a confusion in the A2HS nomenclature. The others were new variants with cathodal isoelectric points to A2HS 2 in the native state.

Genetic polymorphisms of orosomucoid and alpha-2-HS-glycoprotein in a Philippine population.

A Philippine population was investigated for genetic polymorphism of orosomucoid 1 (ORM1), orosomucoid 2 (ORM2), and alpha-2-HS-glycoprotein (A2HS) by isoelectric focusing. The allele frequencies were: ORM1*1 = 0.7904, ORM1*2 = 0.1687, ORM1*2.1 = 0.0409, ORM2*1 = 1.0000; A2HS*1 = 0.6870, A2HS*2 = 0.3130. Compared with the Japanese population, the Philippine population has higher frequencies of ORM1*1 and A2HS*2, and lower frequencies of ORM1*2.1 and A2HS*1.

Orosomucoid (ORM) typing by isoelectric focusing: evidence for an additional duplicated ORM1 locus haplotype and close linkage of two ORM loci.

Human serum orosomucoid (ORM) exhibits a high variability. Several alleles including a duplicated ORM1 gene (ORM1*2.1) have been identified at two functional ORM loci, ORM1 and ORM2. In this study a modified isoelectric focusing, in which glycerin was omitted from gels, was used to differentiate a new variant ORM1 5 from ORM2 3. The ORM1 5 band was always observed together with the ORM1 2 band. The simultaneous expression could be explained in terms of an additional duplicated ORM1 locus haplotype, ORM1*5.2, whose average frequency was .016 in two Japanese populations. The ORM2*6 found at a polymorphic frequency (.023) was demonstrated to be in association with ORM1*2, indicating the close proximity between ORM1 and ORM2 loci.

Orosomucoid (ORM) typing by isoelectric focusing: evidence for gene duplication of ORM1 and genetic polymorphism of ORM2.

It has been demonstrated that the genetic polymorphism of human serum orosomucoid (ORM) is controlled by polymorphic ORM1 and monomorphic ORM2 loci. In this study a Japanese family was encountered in which several members had puzzling electrophoretic patterns consisting of four bands. The ORM patterns were due to the products of a duplicated ORM1 locus haplotype (ORM1 2.1) or the products of new variant alleles at the ORM2 locus. The ORM1 2.1 haplotype is very common in the Japanese population, occurring at an allele frequency of 0.16. The increased occurrence of ORM12-1 and the heterogeneity in band intensity among ORM12-1 phenotypes could be explained in terms of a duplicated gene ORM1 2.1. The ORM2 locus proved to be polymorphic, with six alleles in the Japanese population.

Transferrin variants in Japan and New Zealand. Report of an unusually sialyzed TF variant.

The genetic polymorphism of transferrin (TF) was studied in 2,167 Japanese individuals and in 448 New Zealanders. The three TF C subtypes were identified, but TF C3 was absent from Japanese populations. In addition, three TF B and six TF D variants were observed, each of which occurred either in Japanese or in New Zealanders. Stepwise removal of N-acetyl-neuraminic acid (NANA) with neuraminidase revealed that the most cathodal variant TF DShinnanyo found in a Japanese family was characterized by having only two NANA residues.

The polymorphism of desialyzed alpha 2HS-glycoprotein (AHS): isoelectric focusing in 2.5 M urea as a method for identification of genetic variants.

Desialyzed plasma specimens were phenotyped for alpha 2HS-glycoprotein (AHS) using polyacrylamide isoelectric focusing (IEF) in the range pH 5-6 in 2.5 M urea, followed by immunoblotting. The technique used in this study is easy to perform and can differentiate the gene products of all the currently known variants of alpha 2HS-glycoprotein except for AHS 4.

Orosomucoid (ORM) typing by isoelectric focusing: evidence of two structural loci ORM1 and ORM2.

Orosomucoid (ORM) phenotyping was performed by isoelectric focusing and immunoprinting. The band patterns of desialyzed ORM indicated that the ORM system is controlled by two structural loci ORM1 and ORM2. In a total of 253 samples from two Caucasoid populations, five phenotypes determined by three polymorphic alleles, ORM1*1, ORM1*2, and ORM1*3 were identified. The ORM1*3 was characteristic of the Caucasoids. The ORM2 locus was monomorphic.

Phosphoglucomutase-1 subtypes: polymorphic occurrence of PGM1*7+ and geographical variation in Japan.

The distribution of gene frequencies in the phosphoglucomutase-1 (PGM1) system was investigated in two Japanese populations from Yamaguchi (Western Japan) and Okinawa (Southern Japan) using an improved isoelectric focusing method permitting the successful detection of the most anodal variant PGM1 3+. PGM1*7+ occurred with a polymorphic frequency of 0.012-0.021. A difference in the gene frequency was observed between the two populations. In comparison with neighboring populations, the Yamaguchi population was similar to Mongolians and Koreans in North China, and Okinawa to Zhuang in South China.

A new alpha 1-antitrypsin allele PI Poki: isoelectric focusing with immobilized pH gradients as a tool for identification for PI variants.

A new rare genetic variant PI Poki in the protease inhibitor system found in two Japanese subjects, was compared with some PI P variants using several electrophoresis systems. Although this variant was slightly anodal to P by separator isoelectric focusing, it could be readily distinguished by isoelectric focusing with immobilized pH gradients: the mobility of Poki became cathodal to P. This method proved to be a powerful tool for identification of PI variants.

Determination of alpha 2HS-glycoprotein phenotypes by isoelectric focusing and immunoblotting: polymorphic occurrence of HSGA*5 in Okinawa.

A print immunofixation is a very useful procedure for the demonstration of alpha 2HS-glycoprotein (HSGA) following polyacrylamide gel isoelectric focusing. However, this technique has the one disadvantage of requiring a large volume of expensive antiserum. In this paper an alternative detection system is presented which involves the non-electric transfer of HSGA from a focused gel to a nitrocellulose filter and the immunologic detection of HSGA immobilized on nitrocellulose. Using this method the distribution of HSGA polymorphism in the Nepalese and Japanese populations was investigated. All the populations tested were found to lack the common HSGA*3 of the Caucasians. A rare HSGA*5 in Honshu, a main island of Japan, was observed at polymorphic frequency in Okinawa, Southern Japan.

PI(alpha 1-antitrypsin) polymorphism in the Japanese: confirmation of PI*M4 and description of new PI variants.

PI phenotyping by separator isoelectric focusing (SIEF) was performed on a total of 1000 unrelated Japanese individuals from two different areas of Western Japan. The PI M1M4 subtype was observed together with the six common PI M subtypes. PI*M4 was confirmed to be present but rare in the Japanese. Several new PI variants were identified by comparison runs of each variant with previously reported genetic variants. The significance of treatment of serum with dithiothreitol (DTT) followed by iodacetic acid (IAC) in determination of PI variants is also described.
